Application of Zicaorong Granules Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Zicaorong Granules Veterinary Grade API is supplied as a granular active substance whose downstream processing routes are governed by separate veterinary pharmaceutical and feed legislation, not by a single generic method. Because the material is specified for tablets, injectable solutions, capsules, powders, granules, premixes, and oral solutions, the application sector determines whether the relevant controls fall under EU GMP Part I, EU GMP Part II, 21 CFR 210/211, or medicated feed requirements. Particle size distribution, residual moisture, and granule friability are the first variables affecting disintegration in oral solids, dissolution in waterborne administration, and suspending behaviour in liquid dosage forms. The downstream routes below are restricted to established industrial sectors for veterinary drug product manufacture: swine feed medication, poultry drinking water treatment, parenteral administration in cattle and swine, companion animal solid oral dosage forms, neonatal swine oral dosing, and ruminant drench solutions. Each sector is described with compliance standards, formulation addition ratio, downstream processing sequence, and finished product types. Process data referenced are derived from standard veterinary pharmaceutical unit operations; where product-specific monograph data for Zicaorong Granules in a given configuration is unavailable, that limitation is stated explicitly.

Medicated feed premix granulation and mixer carryover thresholds in swine production

In swine production, Zicaorong Granules Veterinary Grade API is converted into a concentrated veterinary premix before it is diluted into complete feed or top-dressed at the farm. Compliance for this route falls under 21 CFR 558.3 in the United States and under Regulation (EU) 2019/6 with national medicated feed rules in the European Union; target animal safety documentation is typically structured according to VICH GL11(R). The formulation addition ratio for the concentrated premix is commonly 1–10% w/w active granule loading, with final complete feed inclusion rates between 0.1 kg/tonne and 5.0 kg/tonne depending on the posology approved in the marketing authorization; the API granule fraction must be adjusted by the supplier’s anhydrous assay to avoid overages above the permitted assay range. Downstream production uses a horizontal ribbon blender or paddle mixer with a high-speed chopper blade; the API granules are pre-screened through a 1000 µm mesh before charging, and sequential dilution is performed in not fewer than three geometrically matched blending stages. Blend uniformity is evaluated at 10-minute intervals until the coefficient of variation for active assay does not exceed 5.0%; after each production campaign, flush batches with ground maize or lactose are used to verify carryover below 0.3% of the next batch theoretical active concentration. If the granule moisture is above 2.0% w/w or the ambient relative humidity exceeds 60%, pre-drying at 40 °C for 2–4 h is required before blending to prevent adhesive build-up on mixer blades. Terminal finished product types include 0.2%, 1%, and 5% veterinary premixes, medicated meal feed, and pelleted feed produced under steam conditioning temperatures that must be justified by forced degradation data for the active substance.

What governs injectable solution stability in multi-dose cattle and swine presentations?

Parenteral administration introduces sterility assurance and vehicle compatibility constraints that are absent in oral powder and premix routes. The relevant compliance framework is EU GMP Annex 1 for sterile medicinal products, Ph. Eur. 5.1.1 for methods of preparation of sterile products, and VICH GL18(R2) for residual solvent control when organic co-solvents are employed. Zicaorong Granules Veterinary Grade API is dissolved or suspended at a formulation addition ratio of 5–15% w/v active substance in an aqueous vehicle; when solubility is insufficient, co-solvent systems with propylene glycol or glycerol formal are limited to the residual solvent justification and target species tolerance data. The downstream process sequence includes dissolution in water for injection at 20–25 °C, pH adjustment with citrate or phosphate buffer to a target of pH 4.5–6.5, nitrogen sparging to maintain dissolved oxygen below 1.0 mg/L, and membrane filtration through a 0.22 µm polyethersulfone filter before aseptic filling. If the substance is thermally stable, terminal steam sterilization at 121 °C for 15 min with F0 ≥ 8.0 is preferred over aseptic filtration alone; if not, terminal sterilization is replaced by validated aseptic processing and strict pre-filtration bioburden limits. Multi-dose rubber closures must be selected for low stopper extractables and minimal sorption of the active substance; silicone oil levels above 0.2 mg/cm² on stopper surfaces can reduce visible particulate acceptance. Terminal finished product types include 50 mL, 100 mL, and 250 mL multi-dose vials for cattle and swine, and 20 mL single-dose vials where the marketing authorization demands single-use presentation. A significant process boundary is that oxygen-sensitive formulations require headspace nitrogen flushing to residual oxygen below 2.0%; without this control, oxidative degradation products may approach the qualification threshold during the assigned shelf life.

Broiler and layer operations use Zicaorong Granules Veterinary Grade API in water-soluble powder or concentrated stock solution form for mass administration. Because this route distributes active substance through galvanized or polyvinyl chloride drinker lines, hard-water compatibility, pH, and solution stability over 24 h replace the mixer homogeneity concerns seen in feed premix. Compliance for waterborne medication in major export markets relies on Regulation (EU) 2019/6, national treatment frequency rules, and pharmacopoeial solubility/dissolution controls such as Ph. Eur. 2.9.3 and USP <1236> for solubility determination; residue decline data for food-producing poultry are structured under VICH GL48 and VICH GL49 if a withdrawal period is required. The soluble powder formulation typically contains 20–40% w/w Zicaorong Granules Veterinary Grade API, with lactose monohydrate, dextrose, or sodium carbonate as alkalizing agents to stabilise the solution, and the final in-use drinking water concentration is adjusted to 0.05–0.20 g/L depending on body weight and daily water consumption. Manufacturing employs high-shear dry blending followed by 500 µm screening and secondary blending; the resulting powder is filled into 100 g, 500 g, and 1 kg sachets or polypropylene jars with desiccant. In-line administration uses proportioner pumps set between 0.5% and 2.0% stock solution ratio; farm tests are required to verify that the stock solution remains within ±10% of label claim for the full administration period at water temperatures from 5 °C to 30 °C. Terminal finished product types are water-soluble powder sachets, effervescent-style granules where carbonate blends are used to improve dispersion, and concentrated liquid stock solutions in 1 L and 5 L high-density polyethylene bottles. One boundary is that hard water with total hardness above 500 ppm CaCO₃ can precipitate or reduce dissolution; chelating agents such as citric acid are added at 0.5–2.0% w/w only if the target species tolerates the resulting pH shift.

When direct compression tablets replace granulated premixes in companion animal and calf dosing

Direct compression of Zicaorong Granules Veterinary Grade API into tablet cores is selected when the dose per animal is low and homogeneous distribution across 10 mg to 50 mg strengths is required. The governing standards are USP <905> for content uniformity, Ph. Eur. 2.9.5 for uniformity of mass of single-dose preparations, and Ph. Eur. 2.9.3 for dissolution; finished product manufacture must also comply with 21 CFR 210/211 or EU GMP Part I for veterinary medicinal products. The formulation addition ratio for the granular API in a direct compression blend is 5–25% w/w, with the remainder comprising microcrystalline cellulose, lactose monohydrate, crospovidone at 2–4% w/w, and magnesium stearate at 0.25–0.75% w/w. Processing begins with dry milling of the API granules through a conical mill fitted with a 0.039 in screen to reduce oversized particles; the milled material is blended in a bin blender with a fill volume between 40% and 70% of working capacity, and magnesium stearate is added in the final 3 min to avoid over-lubrication. Tableting is performed on a rotary press with precompression force 8–12 kN and main compression force 15–25 kN; target tablet hardness is 80–120 N, friability is controlled below 0.8%, and disintegration time is recorded according to Ph. Eur. 2.9.1. If the API granule exhibits poor flow, a dry granulation step using roller compaction at 30–60 bar is introduced before final compression. Terminal finished product types include 10 mg, 25 mg, and 50 mg uncoated or film-coated tablets for dogs, cats, and pre-ruminant calves, packaged in aluminium/PVC blister cavities or high-density polyethylene bottles with child-resistant closures. The main process boundary is moisture uptake into the direct compression blend above 2.0% w/w causing sticking to tablet punches; therefore, compression suites are maintained at 20–25 °C and 35–45% RH. Capsule filling with hard gelatin shells follows the same blend ratio, with fill weights from 100 mg to 250 mg and capsule sizes #2 or #3; capsule product types are used for individual dose titration where tablet splitting is not authorised.

Neonatal swine oral dosing formulations rely on reconstitutable powders and granules rather than in-feed premix because piglets have low and variable feed intake during the first seven days of life. In this route, Zicaorong Granules Veterinary Grade API is blended with lactose or dextrose to produce a drenchable suspension at a formulation addition ratio of 2–8% w/w active substance in the oral powder; the dose is administered by oral pump or syringe after mixing with 10–20 mL water per piglet. Compliance is derived from VICH GL11(R) for target animal safety and EU GMP Part I for oral powder production, while residue disappearance in treated piglets is addressed through VICH GL48 and VICH GL49 data if the product is used in animals that may enter the food chain. The manufacturing process uses a low-shear ribbon blender with a liquid-bar spray system to prevent dusting; the blend is screened through 800 µm stainless mesh and filled into 25 g, 100 g, or 500 g aluminium foil-lined pouches under nitrogen to reduce oxidative degradation. Sucrose-based carriers are avoided when there is evidence of hygroscopic clumping; instead, anhydrous lactose is preferred, and a desiccant is placed in the pouch. Terminal finished product types include oral drench powders, reconstitutable oral granules, and milk-replacer top-dress sachets for neonatal piglets; the granules are not intended for use in automatic feed lines because the small dose fraction and sticky carrier can lead to poor flow in auger systems. A critical boundary is that water used for reconstitution should be potable and below 25 °C; if the suspension is left for more than 4 h, sedimentation may occur and the product should be discarded unless a suspending agent such as xanthan gum at 0.1–0.3% w/w is included.

Ruminant oral solution and drench formulation: viscosity, preservatives, and in-use stability

Oral solution and drench presentations for cattle and sheep are manufactured when the dose is high or when rapid oesophageal groove delivery is required. The formulation addition ratio for Zicaorong Granules Veterinary Grade API in these liquids is 5–12% w/v; the vehicle is typically purified water with 10–20% v/v propylene glycol or sorbitol solution to improve palatability and prevent freezing during transport, although co-solvent selection must be confirmed by residual solvent controls under VICH GL18(R2). Compliance for oral liquids in food-producing species includes Regulation (EU) 2019/6, VICH GL11(R), and pharmacopoeial monographs for oral liquids; in North America the relevant framework includes 21 CFR 210/211 and any approved animal drug application conditions. Downstream production uses a jacketed stainless steel vessel with a high-torque propeller agitator operating at 200–500 rpm; the granular API is added through a mesh bridge into the vortex to avoid lump formation, and the solution is mixed for 30–60 min at 20–25 °C until complete dissolution. The solution is then filtered through a 50 µm in-line filter before filling into 1 L and 5 L high-density polyethylene jerry cans or 500 mL and 1 L drench bottles with graduated dosing chambers. Preservation systems, where required, use methylparaben 0.05–0.18% w/w and propylparaben 0.01–0.02% w/w, but parabens are not used in products where the marketing authorization prohibits their presence in food-producing species; alternative systems are validated by antimicrobial effectiveness testing according to Ph. Eur. 5.1.3. Terminal finished product types include oral drench solutions, ready-to-use oral liquids, and pump-administered mineral oil-free formulations. The main operational boundary is viscosity rise below 5 °C in concentrated sorbitol vehicles; therefore, cold-chain storage and thawing studies are required for exports to markets where winter transport is below 0 °C. Published data for Zicaorong Granules Veterinary Grade API in this specific drench configuration is limited, so forced degradation and in-use stability protocols should define the maximum holding time before the product is discharged.

    More Introduction

Zicaorong Granules Veterinary Grade API is a granulated active pharmaceutical ingredient intermediate supplied under route-specific model designations: ZCR-G-TAB for tablet compression, ZCR-G-CAP for capsule filling, ZCR-G-PWD for dry powder reconstitution, ZCR-G-GRN for direct granule administration, ZCR-G-PMX for medicated premix manufacture, ZCR-G-SOL for concentrated solution preparation, and ZCR-G-INJ for parenteral processing. The granule is not a finished veterinary medicinal product; it is a flow-optimised API input intended for further processing under ICH Q7 good manufacturing practice. Its principal difference from conventional API powders is controlled particle geometry and density, which reduces dust formation during dispensing and improves blend homogeneity in low-dose dry-mix applications.

Release is performed against route-specific limits derived from Ph. Eur., USP, VICH GL18, and ISO 13320-1:2009. Each batch is issued with a certificate of analysis containing assay, related substances, loss on drying, particle-size distribution, bulk density, tapped density, microbial limits, and—where the route code is ZCR-G-INJ—bacterial endotoxin and sterility data. Because the granule is intended for multiple dosage forms, the release limits are not identical across all model codes; oral and premix grades permit loss on drying ≤ 1.0% by Ph. Eur. 2.2.32, whereas the parenteral grade is controlled at ≤ 0.5%.

The granule retains a crystalline habit rather than a spray-dried amorphous dispersion. Consequently, moisture-induced recrystallisation and plasticisation during storage are not primary failure modes, but the product remains hygroscopic enough to require immediate closure of bulk containers and pre-drying when ambient relative humidity exceeds 60%.

What Release Specifications Govern Tablet, Capsule, and Premix Interchangeability?

The oral and premix route codes share a common core specification but are not automatically interchangeable for all equipment variants. A summary of the release panel is provided below.

The parenteral grade is not a finer-milled variant of the oral grade. ZCR-G-INJ requires a dedicated granulation and packaging suite, endotoxin-controlled process water, and a different residual solvent drying endpoint. A batch manufactured as ZCR-G-TAB cannot be released as ZCR-G-INJ without repeating the full parenteral test panel, including subvisible particle counts by Ph. Eur. 2.9.19 and particulate matter by USP <788> if the final formulated solution is destined for injection.

    Compatibility Boundaries and Processing Restrictions

Dry blending of ZCR-G-TAB or ZCR-G-PMX in a bin blender at 12 rpm for 20 min is generally sufficient to achieve blend uniformity when the granule is diluted with excipients of similar bulk density. Over-lubrication with magnesium stearate above 1.0% w/w produces a measurable reduction in tablet tensile strength and increased disintegration time, so lubricant level and blend time must be controlled simultaneously. For direct compression on a 27-station rotary tablet press with paddle feeder, die-fill variability is maintained below 2% RSD when granule flow time is ≤ 15 s/100 g and hopper relative humidity is below 60%.

The granule is incompatible with strong oxidising agents and concentrated mineral acids. In aqueous solution manufacturing, pH adjustment below 3.0 or above 9.0 should be avoided because the API exhibits pH-dependent hydrolysis; a buffered vehicle is recommended for ZCR-G-SOL and ZCR-G-INJ. Contact with copper, iron, or zinc equipment parts should be minimised; chelating agents such as edetate disodium may be required in solution formulations to suppress metal-catalysed degradation.

For capsule filling, ZCR-G-CAP is discharged from a low-shear blender into a tamping-pin capsule filling machine. Fill weight variation is monitored against USP <905> or Ph. Eur. 2.9.40. The granular form reduces slugging and ratholing in the powder hopper compared with micronised API, but if granule friability exceeds 1.0% by Ph. Eur. 2.9.7, fine generation during high-speed filling can increase fill weight drift. For reconstitution powders, ZCR-G-PWD is dispersed in purified water at 20–25°C; typical dispersion is achieved with stirring at 200 rpm for 5 min. If water temperature falls below 15°C, mixing time may extend beyond 10 min.

When Injectable-Grade Granules Replace Micronized API in Aseptic Compounding

Compared with micronised active powder, ZCR-G-INJ granules reduce airborne dust, electrostatic adhesion to isolator gloves, and product loss during transfer into aseptic fill lines. This advantage is accompanied by two constraints. First, the granule must completely dissolve before sterile filtration; undissolved subvisible particles can block membrane filters and alter delivered dose. Second, endotoxin control must be maintained at the API stage rather than relying on downstream sterilisation, because terminal steam sterilisation does not reliably inactivate pre-existing endotoxins. Dry-heat depyrogenation of contact equipment at 250°C for 30 min or steam sterilisation at 121°C for 15 min is used for equipment, but the API itself cannot be depyrogenated by heat without degradation.

Another difference from oral premix and tablet grades is the parenteral particle-size ceiling of D90 ≤ 200 μm. The parenteral grade is not intended for direct powder filling into vials; it is an intermediate for solution or reconstituted suspension manufacture. Where a dry powder for injection is required, the granules must be further processed under low-humidity containment, and the manufacturer must demonstrate content uniformity and water content suitable for the intended lyophilisation or powder-fill cycle.

Residual solvent, elemental impurity, and endotoxin control are route-specific

Control area Applied standard or method Route-specific acceptance criterion
Residual solvents USP <467>, VICH GL18, Ph. Eur. 2.4.24 Class 1 solvents not detected; Class 2 within ICH Q3C; Class 3 ≤ 0.5%
Elemental impurities USP <232> / <233>, ICH Q3D Option 1 Pb, Cd, As, Hg controlled; heavy metals ≤ 20 ppm oral / ≤ 10 ppm parenteral by Ph. Eur. 2.4.8

Sterility Ph. Eur. 2.6.1 No growth after 14 days for ZCR-G-INJ when tested as final aseptic bulk or finished parenteral product
Bacterial endotoxin Ph. Eur. 2.6.14, USP <85> ≤ 0.5 EU/mg for ZCR-G-INJ; not specified for oral or premix unless stated in marketing authorisation
    Particle-size method transfer ISO 13320-1:2009 laser diffraction D10, D50, D90 reported on CoA; span = (D90-D10)/D50 ≤ 1.8 for oral/premix and ≤ 2.0 for parenteral
Reference standards Ph. Eur. CRS / USP RS CoA traceable to current compendial standard lot

In comparison with unprocessed API powder, Zicaorong granules show lower dusting and better flow. However, the granule form is not universally preferable. If a final formulation requires sub-1 mg/g drug loading without a wet-granulation step, a pre-blended intermediate or a fine-milled API with particle-size-matched diluent may provide lower segregation potential. Compared with wet-granulated API premixes, the product has defined residual moisture and does not require a downstream drying step before use. Compared with spray-dried dispersions, the granules do not contain amorphous drug fractions and therefore avoid the glass transition-related caking that can occur during transport at elevated temperature. The trade-off is that this granule is not a solubility-enhanced form; in formulations where dissolution rate is the bioavailability-limiting factor, the formulator must conduct comparative dissolution testing rather than assuming equivalence.

Medicated premix and solution manufacture uses the ZCR-G-PMX and ZCR-G-SOL route codes, respectively. For in-feed premix, the granules are diluted stepwise with a feed-grade carrier to a final content that supports mixer homogeneity acceptance of CV ≤ 5.0% across 10 sampling points per batch. The use of concentrated premixes below 1 mg/g without pre-blending is not recommended because segregation risk increases when particle-size differences between API granules and carrier exceed approximately 3:1 in D50. For solution preparation, the granule is dissolved in purified water at 20–25°C; for parenteral compounding, Water for Injection is used and the final solution is passed through a 0.22 μm polyethersulfone or polyvinylidene fluoride membrane. Complete dissolution is confirmed by ≥ 99.0% recovery through the membrane at the target concentration. Filter throughput can decline if the vehicle ionic strength is below 0.9% w/v sodium chloride equivalent or if the solution is processed below 15°C; published data for this specific configuration is limited, and site-specific filter validation is required.

Bulk storage of oral and premix grades in sealed polyethylene-lined multiwall bags is controlled at ≤ 25°C and ≤ 60% RH. The assigned retest interval is 36 months for oral/premix grades and 24 months for parenteral grade under these conditions. Opened containers should be re-sealed immediately and used within 30 days; if ambient relative humidity exceeds 60%, uncontrolled exposure should not exceed 4 h.
