Direct-compression tableting of Spirit Veterinary Grade API into veterinary tablets for companion-animal anti-infective therapy requires particle-size conditioning on a conical screen mill fitted with a 0.8 mm round-hole screen. The API is pre-blended with microcrystalline cellulose and pregelatinized starch at 12 rpm in a 500 L double-cone blender for 18 min, after which magnesium stearate is introduced through a 0.5 mm screen during the final 3 min to limit over-lubrication. A 27-station rotary tablet press using 8 mm round biconvex tooling operates at turret speed 30–60 rpm, precompression force 6–10 kN, and main compression force 12–18 kN; ejection force above 1.5 kN triggers review of feed-frame desiccant performance and punch-face condition. Active load is set between 5.0% w/w and 30.0% w/w; a representative 100 mg strength contains 20.0% w/w Spirit Veterinary Grade API, 35.0% w/w microcrystalline cellulose, 39.5% w/w lactose monohydrate, 4.0% w/w crospovidone, 1.0% w/w colloidal silicon dioxide, and 0.5% w/w magnesium stearate. Blend uniformity acceptance is RSD ≤5.0% for ten stratified samples assayed by HPLC with UV detection. Tablet hardness is held at 80–120 N, friability ≤1.0%, and disintegration ≤15 min in 37°C water. Dissolution uses USP <711> apparatus II with 50 rpm paddle speed in 0.1 M HCl, requiring ≥80% release of label claim within 60 min. Pre-drying of API and excipients to loss-on-drying ≤2.0% is required when ambient RH exceeds 60%; otherwise punch-face adhesion, weight drift, and capping increase. The core tablets are film-coated with aqueous HPMC-based dispersion to a target weight gain of 3.0%, using inlet air temperature 60–70°C and pan speed 6–10 rpm. Release testing includes USP <905> uniformity of dosage units, Ph. Eur. 2.9.3 uniformity of mass of single-dose preparations, ICH Q3D elemental impurities, and VICH GL18 residual solvents. Terminal finished products are 50 mg, 100 mg, and 250 mg film-coated tablets packed in 10-tablet aluminum blisters.
In a terminal-sterilization line, Spirit Veterinary Grade API is dissolved in Water for Injection at 60°C under nitrogen sparging until a clear solution is obtained. The formulation contains Spirit Veterinary Grade API at 5.0%–20.0% w/v, sodium chloride 0.9% w/v for tonicity, and pH adjusted to 4.5–5.5 with 1 M HCl or 1 M NaOH. The bulk solution is passed through a 0.45 µm prefilter and two 0.22 µm PVDF sterilizing-grade filters in series, then filled under Grade A conditions into siliconized Type I glass vials. Where thermal stability data support, terminal sterilization is performed at 121°C for 15 min with F₀ ≥12; otherwise, aseptic processing is validated to a sterility assurance level ≤10⁻⁶. Bacterial endotoxin limits are calculated from K/M = 5 EU/kg divided by the highest intended dose of 10 mg/kg, yielding 0.5 EU/mg, tested per Ph. Eur. 2.6.14 Method A. Particulate matter is controlled per USP <788> Method 1 with limits of ≤6000 particles ≥10 µm and ≤600 particles ≥25 µm per container. Residual solvent limits follow VICH GL18 for methanol, ethanol, and acetone. The filling line uses validated peristaltic pumps with fill volume tolerance ±5.0%; headspace oxygen is held below 2.0% after nitrogen flushing. Process temperatures above 60°C during dissolution should not be extended beyond 4 h without degradation profiling. Terminal finished products are 50 mL, 100 mL, and 250 mL injection vials for intramuscular or subcutaneous administration in cattle and swine.

Water-dispersible powder lines are built around dry blending rather than granulation because Spirit Veterinary Grade API must disperse rapidly into drinking-water systems at use rates of 0.5–1.0 g/L. The addition ratio is 20.0%–50.0% w/w active substance, with the balance selected from glucose monohydrate, anhydrous citric acid, and polyvinylpyrrolidone K30 as wetting carriers. The API and carriers are sieved through 0.5 mm, blended in a ribbon mixer at 20 rpm for 20 min, and packed into laminated aluminum sachets at residual moisture ≤1.5%. Water dispersibility is confirmed by dissolving 5 g in 1 L water at 25°C; pass criterion is no more than 0.5% residue on a 75 µm screen. Release controls include Ph. Eur. 5.1.4 microbial quality of non-sterile products for oral use, Ph. Eur. 2.9.5 uniformity of mass of single-dose preparations, and Regulation (EU) 2019/6 for veterinary medicinal products. Terminal finished products are 100 g, 500 g, and 1 kg laminated sachets for poultry, turkeys, and swine. If storage temperature exceeds 30°C and RH exceeds 60%, bulk powder should be consumed within 30 days because hygroscopic carrier fractions increase moisture above 1.5% and reduce dispersion through nipple drinker systems.
Spirit Veterinary Grade API is incorporated into a 5.0%–10.0% w/w active premix on a stepwise dilution basis to limit assay variance in finished feed. The first stage blends the API with calcium carbonate or corncob meal in a 250 kg horizontal ribbon mixer at 60–70% fill for 10 min; the intermediate is then diluted 1:10 with additional carrier and mixed for 15 min. Final in-feed inclusion is calculated to deliver 100–400 ppm active ingredient, typically 1.0–4.0 kg premix per metric ton of finished feed. During production-scale runs, segregation has been observed when carrier particle-size difference exceeds 300 µm between active granules and carrier; the premix specification therefore requires RSD ≤5.0% for assay of ten stratified samples per batch. Medicated feed production is governed by Regulation (EU) 2019/4 and FDA 21 CFR Part 225/226; batch records document sequential load order, mixer fill, mixing time, discharge, and carryover flushing. Terminal finished types are 20 kg paper bags premix and pelletized medicated feed in 25 kg bags.
| Stage | API content | Mixer fill / time | Acceptance |
|---|---|---|---|
| Primary blend | 10.0% w/w | 60–70% fill / 10 min | No visual streaking |
| Secondary dilution | 1.0–10.0% w/w | 60–70% fill / 15 min | RSD ≤5.0% |
| Finished feed | 100–400 ppm | Batch mixer 12–18 min | RSD ≤5.0% |
| Carryover flush | ≤2.5% of previous batch | Flush with 10 kg carrier | Assay at LOQ |
If relative humidity in the feed mill exceeds 65%, the API premix should be consumed within 7 days because moisture uptake above 0.8% increases caking and changes flow through the dosing screw. Direct addition into molasses-containing feeds is acceptable only after the pellet cooler discharge temperature falls below 45°C; otherwise, heat-labile fractions of the API may degrade and hard pellets can mask homogeneity defects.
Substitution of water-soluble powder with a liquid oral solution is justified when dosing accuracy is prioritized over transport weight. Spirit Veterinary Grade API is dissolved at 5.0%–10.0% w/v in a vehicle composed of propylene glycol 10.0–20.0% v/v, sodium benzoate 0.1% w/v, and purified water. The batch is stirred at 300 rpm for 30 min, pH-adjusted to 3.8–4.5 with citric acid, and filtered through a 10 µm polypropylene cartridge before filling into 1 L and 5 L HDPE containers with calibrated dosing pumps. Compliance includes USP <51> antimicrobial effectiveness testing, Ph. Eur. 5.1.4 microbial quality of non-sterile products for oral use, and VICH GL18 residual solvent limits. Terminal finished product is an oral solution for swine and calves, administered via drinking water or drenching. The solution should not be diluted in hard water with total hardness above 200 mg/L CaCO₃ unless a chelator is added, because precipitation of the active acid salt may occur and reduce delivery through nipple drinkers.
For capsule filling operations that require low-dust granulation, Spirit Veterinary Grade API is dry-granulated by roller compaction rather than wet granulation when moisture sensitivity is a constraint. The API load in capsule fills is 15.0%–50.0% w/w, with roller-compacted ribbons milled through an oscillating granulator fitted with a 1.0 mm screen. The granules are blended with lactose monohydrate and croscarmellose sodium in a 100 L bin blender at 15 rpm for 15 min, then filled into size 0, 1, or 2 hard gelatin or HPMC capsules at 60–80% of maximum fill volume. Loss-on-drying is controlled to ≤2.0% before encapsulation. Dissolution testing uses USP <711> apparatus II at 50 rpm, with acceptance of ≥80% released at 60 min. Terminal finished products are capsules for companion animals and equine, in 50 mg, 100 mg, and 250 mg strengths, packed in 10-capsule or 30-capsule aluminum blister strips. Compliance includes USP <905> uniformity of dosage units, Ph. Eur. 2.9.3 uniformity of mass, ICH Q3D elemental impurities, and FDA 21 CFR 211. Wet granulation should be avoided unless API-excipient contact tests confirm no hydrate formation or binding; published data for this specific configuration on high-shear wet granulation is limited.

Spirit Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions is supplied as a form-specific veterinary active pharmaceutical ingredient platform covering six release designations: Spirit VT-101 for tablet compression, Spirit VC-101 for capsule filling, Spirit VP-101 for oral powders and granules, Spirit VX-101 for medicated feed premix, Spirit VS-101 for solution compounding, and Spirit VI-101 for injectable solution preparation. These designations represent the same chemical entity under different physical, microbial, and packaging controls and are not separate salts or polymorphs unless the approved registration dossier explicitly specifies a salt form. Manufacture is conducted under EU GMP Part II for active substances used in veterinary medicinal products. Release documentation supports EU Regulation 2019/6 and relevant national dossiers. The product is not sterile and is not a finished dosage form; terminal sterilisation, aseptic filtration, or preservative addition remains the responsibility of the marketing authorisation holder.
Every batch is released against a certificate of analysis covering assay, related substances, residual solvents, loss on drying, residue on ignition, and grade-specific physical or microbial properties. The assay acceptance envelope is 98.0–102.0% on dried basis. Total related substances are controlled at ≤0.5%, and any unspecified impurity is controlled at ≤0.10%. Residual solvents are controlled according to ICH Q3C; Class 1 solvents are absent at the method limit, and Class 2 solvents are below the permitted daily exposure calculated for the target species. Loss on drying is ≤0.5% for Spirit VI-101 and ≤1.0% for the oral, premix, and solution designations. Residue on ignition is ≤0.1%. Heavy metals are controlled at ≤10 ppm only where the relevant active substance monograph retains the compendial limit test; otherwise the elemental impurity risk assessment is documented under ICH Q3D principles.
Technical-grade feed additives and non-GMP bulk actives are not interchangeable with Spirit Veterinary Grade API. The platform is released against pharmacopoeial impurity and residual solvent limits; technical-grade materials often carry only assay, moisture, and heavy metal data and may contain process-related impurities above the safety thresholds applied to veterinary medicinal products. Documentation includes an active substance master file or veterinary master file, zone-specific stability data, and batch-to-batch physical property data required for downstream process validation. In contrast to a single-grade veterinary API, the form-specific designations avoid site-level milling, sieving, and reconditioning operations that can introduce cross-contamination risk and shift particle size distribution.
Compared with a human-grade API of the same chemical entity, Spirit Veterinary Grade API may use the same compendial assay and impurity methods, but it adds acceptance criteria for target-animal feed carryover, dusting potential, and packaging suitability for veterinary clinics and feed mills. Human-grade API documentation does not normally address withdrawal periods, milk or egg residues, or compatibility with medicated feed carriers. Compared with a finished veterinary premix or oral powder, the API contains no excipients, carriers, or preservatives; blend uniformity and final dose accuracy must therefore be demonstrated by the marketing authorisation holder using the form-specific grade.
The table below lists the model designations and the primary downstream controls associated with each dosage-form route. The physical specifications are not target values for all active moieties; they are the release envelope for a neutral, crystalline active substance with adequate flow. For low-melting-point or hygroscopic actives, the envelope is adjusted in the registration commitment.
| Designation | Dosage form | Critical physical or microbial control | Method or standard |

| Spirit VC-101 | Capsule filling | Particle size D90 ≤100 µm for high-speed encapsulation | ISO 13320 |
| Spirit VP-101 | Oral powder and granule | Loss on drying ≤1.0%; sieve fraction 0.125–1.0 mm | Ph. Eur. 2.2.32 |
| Spirit VX-101 | Medicated feed premix | Bulk density/tapped density; carrier moisture <12%; carrier pH <8.0 | Ph. Eur. 2.9.36 |
| Spirit VS-101 | Oral or topical solution | Clarity after reconstitution ≤3 NTU at 10% w/v, 25 °C | Validated nephelometry |
| Spirit VI-101 | Injectable solution preparation | Endotoxin <0.5 EU/mg; bioburden ≤10¹ CFU/g | Ph. Eur. 2.6.14; Ph. Eur. 2.6.12 |
Spirit VI-101 is released under more restrictive particulate and microbial controls than the oral designations. Total aerobic microbial count is ≤10¹ CFU/g, and combined yeast and mould count is ≤10¹ CFU/g. The material is not sterile, but bioburden reduction, moisture control, and particle size control support subsequent terminal sterilisation or aseptic filtration through a 0.22 µm membrane. Sub-visible particulate burden is assessed after reconstitution in particle-free water using light obscuration; the acceptance limit is applied to the finished solution rather than to the dry powder alone. In production-scale filtration studies, the filtration rate is influenced by sub-10 µm particle fines and by solution viscosity; dissolution time and clarity are therefore reported for each batch. Storage of opened containers at relative humidity above 60% without desiccant is not recommended because moisture uptake can increase aggregation and shift reconstitution time.
For Spirit VX-101 in medicated feed premix, the largest processing risk is segregation during blending and pneumatic transfer. Uniformity of content is assessed after blending at the intended inclusion rate; the batch is released for commercial use only when the relative standard deviation of active content is ≤5.0% across sampling points. Bulk density and tapped density are controlled because a density mismatch between API and carrier above approximately 0.20 g/cm³ can produce stratification in a double-cone blender. The carrier should be selected with a moisture content below 12% and a pH below 8.0 to avoid hydrolysis of the active moiety. Amine-based carriers or mineral carriers containing free alkaline earth oxides are incompatible with the product and may produce degradation-related impurities above the release limit.
Direct-compression tablet grade Spirit VT-101 is specified with a D50 in the range 50–150 µm and a D90 ≤150 µm. Tablet formulations should be designed to a target compression force that yields hardness in the range 50–120 N and friability ≤1.0% as measured by Ph. Eur. 2.9.7; the exact force depends on excipient choice and tablet geometry. In capsule filling, Spirit VC-101 is specified with a D90 ≤100 µm to maintain uniformity of weight during high-speed encapsulation. Powder flow is characterised by Hausner ratio; a Hausner ratio above 1.30 indicates poor flow and may require the addition of glidant.
For wet granulation of Spirit VP-101, the liquid-to-solid ratio is selected to achieve a wet mass consistency with impeller torque between 20 N·m and 40 N·m on a 25 L high-shear granulator. Endpoint is determined by impeller power consumption rather than time alone. Drying is performed with inlet air temperature not exceeding 60 °C to avoid thermal degradation. The dried granules are milled through a 1.0 mm screen and blended with extragranular disintegrant. The final granule fraction between 0.125 mm and 1.0 mm reduces dusting during transfer and supports reproducible die filling.
Method suitability is verified for each grade because finished veterinary matrices can interfere with direct API methods. Feed premix matrices containing alfalfa meal, mineral oil, or molasses require extraction cleanup before HPLC assay; the validated method should demonstrate recovery between 98.0% and 102.0% in the presence of matrix components. Stability-indicating methods separate the active peak from oxidative and hydrolytic degradation products; forced degradation studies include acid, base, peroxide, thermal, and photolytic conditions. Where the product is intended for food-producing species, residue depletion and withdrawal period data are supplied in the veterinary medicine dossier, not in the API certificate of analysis.
| Test | Acceptance criterion | Reference method or standard |
|---|---|---|
| Assay (dried basis) | 98.0–102.0% | Validated HPLC |
| Total related substances | ≤0.5% | Validated HPLC |
| Unspecified impurity | ≤0.10% | Validated HPLC |
| Loss on drying | ≤0.5% injection; ≤1.0% oral/premix | Ph. Eur. 2.2.32 |
| Residue on ignition | ≤0.1% | Compendial sulfated ash method |
| Total aerobic microbial count | ≤10² CFU/g oral; ≤10¹ CFU/g injection | Ph. Eur. 2.6.12 |
| Bacterial endotoxins (Spirit VI-101) | <0.5 EU/mg unless lower | Ph. Eur. 2.6.14 |
Stability data are generated using ICH Q1A conditions: long-term 25 °C/60% RH, intermediate 30 °C/65% RH, and accelerated 40 °C/75% RH. The retest period is assigned from long-term data and may be extended with ongoing stability batches. Packaging is high-density polyethylene with a heat-sealed aluminium foil overwrap; the double-bag configuration reduces moisture ingress during storage. For moisture-sensitive designations, desiccant is placed between the inner and outer bags. The container closure system is qualified for transport under tropical conditions because field failures in veterinary distribution often occur at humidity above 75% RH.
In dissolution testing, the difference between tablet and capsule grades is most visible. Spirit VT-101 is controlled for particle size and flow so that finished tablets meet a typical dissolution criterion of Q=80% at 30 minutes for a highly soluble active moiety in 0.1 N HCl. For poorly soluble actives, the particle size of Spirit VC-101 or Spirit VS-101 is reduced or a specified polymorph is selected; the dissolution method and acceptance criterion must be established in the finished product dossier. Bioequivalence or target animal efficacy data are not part of the API release and must be generated by the applicant.
Cleaning validation for the multi-product facility follows EU GMP Part II and includes swab and rinse limits derived from the permitted daily exposure of the most potent veterinary active. Carryover of a previous product into Spirit Veterinary Grade API is controlled below 10 ppm to maintain the impurity profile. Cross-contamination risk is further reduced by dedicated sieving and milling capability for each physical grade and by verification of retained solvent levels after cleaning.
For solution-grade Spirit VS-101, clarity after reconstitution is assessed using a validated nephelometric method; the acceptance value is ≤3 NTU for a 10% w/v solution in purified water at 25 °C. This criterion is not applied to Spirit VI-101 injection grade, where sub-visible particle counts are controlled instead. The oral powder and granule grades are sieved to pass 1.0 mm but retain a particle fraction above 0.125 mm to reduce dusting during feed mixing; this range may be adjusted following site-specific occupational exposure monitoring. The solution and parenteral grades are not intended for direct administration without dilution or vehicle preparation.
