Lyophilized live attenuated Salmonella Enteritidis API intended for mass oral vaccination of commercial layer and broiler flocks via drinking water represents one of the most process-sensitive downstream formats in veterinary biologics manufacturing. The pre-lyophilization formulation consists of a stabilizer matrix combining 4–8% w/v sucrose, 1–2% w/v gelatin, and 1–2% w/v sorbitol in 10 mM potassium phosphate buffer pH 7.0–7.4, with the viable antigen adjusted to 1×108–5×108 CFU per single fowl dose before freeze-drying. Production-scale lyophilisation is executed in multi-shelf dryers with shelf temperature ramps from −45 °C to +20 °C over 28–36 h, chamber pressure held at 50–200 µbar, and primary drying governed by a product temperature not exceeding −20 °C to prevent cake collapse. Residual moisture is controlled to 1.5–3.0% by semi-micro determination per Ph. Eur. 2.5.12, and post-reconstitution viability must not decline more than 0.5 log relative to pre-lyophilization counts when reconstituted in cold potable water. Process failures recorded on production lines include edge-vial collapse when shelf temperature uniformity exceeds ±2 °C and viability loss greater than 1 log when condenser capacity is undersized for the batch load. The terminal dosage form is a lyophilized cake or free-flowing powder in glass vials for reconstitution as a drinking-water suspension. Compliance assessment is anchored to Ph. Eur. 0062 Vaccines for Veterinary Use, Ph. Eur. 5.2.6 Evaluation of Efficacy of Veterinary Vaccines, Ph. Eur. 5.2.7 Safety of Veterinary Vaccines, and Ph. Eur. 2.6.1 Sterility. Addition rates for chlorine-neutralised drinking water are typically 1 dose per bird per administration, with the reconstituted suspension used within 2 h because potable water residual chlorine above 0.3 ppm reduces viable count by 0.4–0.6 log in field conditions.
Formalised Salmonella Enteritidis whole-cell antigen is prepared at 1×109 inactivated cells/mL in the aqueous phase, which constitutes 30% v/v of the final emulsion; the oil phase, 70% v/v, consists of light mineral oil containing 1–2% w/v Arlacel 80 as a non-ionic lipophilic emulsifier. A production-scale high-shear rotor-stator mixer operated at 10,000 ± 500 rpm for 5–10 min generates a droplet size distribution with Dv50 of 1–3 µm, measured by laser diffraction per ISO 13320:2009. Batch-to-batch variation in Dv90 beyond 8 µm correlates with accelerated creaming and injection-site granuloma formation in field lots, establishing Dv90 as a batch release criterion. Viscosity is maintained at 25–50 mPa·s at 25 °C per Ph. Eur. 2.2.8 to permit passage through 18–21 G needles without excessive syringe force. Residual formaldehyde after inactivation is reduced to ≤0.1% w/v by diafiltration through 100 kDa tangential flow membranes, and thiomersal is added at 0.01% w/v as preservative. The downstream process includes inactivation at 37 °C for 24 h, removal of low-molecular-weight medium components, emulsification, and aseptic filling into 500 mL bottles. The terminal product is an oil-emulsion injectable suspension administered intramuscularly at 0.5 mL per bird. Compliance testing follows Ph. Eur. 0062, Ph. Eur. 5.2.7, and Ph. Eur. 2.6.15 Bacterial Endotoxins; published data for emulsion ageing in this specific antigen configuration is limited but industrial lot records support droplet size and viscosity as primary release criteria. Storage at 2–8 °C is mandatory because freeze-thaw cycles split the emulsion, releasing free aqueous antigen and reducing potency below the reference titre.
Swine oral live attenuated Salmonella Typhimurium API formulated as enteric-coated granules for feed top-dressing requires a two-step fluid-bed process. First, the antigen is adsorbed onto a carrier blend of 70–80% w/w lactose monohydrate and 10–15% w/w microcrystalline cellulose; second, a methacrylic acid copolymer dispersion, Eudragit L 30 D-55, is sprayed at 12–18% dry weight gain in a Wurster insert with inlet air temperature 35–40 °C, product temperature not exceeding 28 °C, and outlet relative humidity below 30%. The final granule contains 2×109 CFU/g, and inclusion in feed at 1 kg premix per tonne delivers approximately 1×108 CFU per animal per day to grow-finish pigs. Loss on drying is held at <3%, and acid resistance testing in pH 1.2 HCl for 2 h followed by pH 6.8 phosphate buffer for 1 h must show release of more than 80% of viable organisms. Particle size distribution is controlled by sieve analysis per Ph. Eur. 2.9.38, with granules retained between 250 µm and 1.4 mm to ensure feed distribution uniformity. Process failure modes observed at production scale include irreversible viability loss when inlet air exceeds 45 °C and electrostatic agglomeration of granules when relative humidity falls below 10%. Tablet and capsule formats are uncommon for mass oral vaccination because the required CFU dose cannot be reliably delivered under field feeding conditions, and published data for compressed-tablet antigen survival during direct compression at 5–15 kN hardness is insufficient to define release criteria. The terminal product is a granulated premix for oral administration; compliance is evaluated under Ph. Eur. 0062, Ph. Eur. 5.2.6, and VICH GL44 Target Animal Safety for Veterinary Live and Inactivated Vaccines.
Killed whole-cell Salmonella Typhimurium antigen is produced in tryptic soy broth to 1×1010 cells/mL, inactivated with 0.5% v/v formalin at 37 °C for 48 h, then concentrated and diafiltered through 100 kDa tangential flow membranes. Adsorption to Alhydrogel 2% aluminium hydroxide gel is conducted at 4 °C for 4–8 h with continuous stirring, targeting 2.0 mg Al per 2 mL dose and 95% antigen adsorption at pH 6.8–7.2. The buffer system is 10 mM phosphate with 0.85% sodium chloride and 0.01% thiomersal as preservative. Critical process limitations include precipitation when pH drifts above 7.4 and irreversible gel matrix aggregation if storage drops below 0 °C. Terminal product is an injectable suspension administered intramuscularly at 2 mL per swine, with release testing per Ph. Eur. 0062, Ph. Eur. 5.2.6, and Ph. Eur. 2.6.14 Pyrogens. Batch-to-batch variance in adsorption efficiency is monitored by centrifugation followed by total protein assay per Ph. Eur. 2.5.33, and free antigen exceeding 5% triggers formulation rework. The formulation is not compatible with lyophilisation because aluminium hydroxide gel particles sediment and aggregate during freeze-thaw cycles, producing visible flocculation and failure of syringeability tests using 18 G needles. Production-scale observations indicate that addition of antigen to aluminium hydroxide under high-shear mixing above 500 rpm reduces adsorption by 10–15 percentage points, whereas low-shear magnetic stirring at 100–200 rpm preserves the gel structure and achieves consistent adsorption kinetics within 2 h.
Formulation of Salmonella Dublin formalinised bacterin at 1×109 cells/mL with aluminium hydroxide adjuvant at 2.5 mg Al per dose and pH 6.5–7.0 supports simultaneous subcutaneous administration with Clostridium perfringens type C and D toxoids in dairy calves from 2 weeks of age. The blending sequence requires separate adjuvant adsorption for each antigen before final combination, preventing competitive displacement from the aluminium hydroxide surface; batch records from multi-valent production lines document that sequential addition reduces free antigen below 5% as measured by centrifugation and UV absorbance at 280 nm per Ph. Eur. 2.2.25. Final dosage is 2 mL subcutaneously, with a booster at 21–28 days. Process incompatibilities include elevated endotoxin burden if formaldehyde concentration exceeds 0.2% v/v during inactivation, and gel particle aggregation when calcium-containing diluents are used for reconstitution or dilution. The terminal product is an injectable suspension; compliance is assessed according to Ph. Eur. 0062, Ph. Eur. 5.2.6, and Ph. Eur. 2.6.14 Pyrogens. Shelf-life data from production lots indicate that aluminium hydroxide adjuvant particle size Dv50 increases from 1.5 µm to 4.8 µm over 18 months at 2–8 °C, necessitating gentle resuspension by hand shaking for 20–30 s before each use. Vaccination of animals with concurrent colostral antibody levels above 1.5 log10 neutralising titre may suppress seroconversion to the bacterin, a limitation documented in field efficacy studies conducted under VICH GL44.
Hatchery spray administration of live attenuated Salmonella Typhimurium vaccine for day-old chicks uses a water-based solution prepared by dissolving a lyophilized powder in sterilised distilled water to 1×109 CFU/mL concentrate, then diluting 1:10 to yield 1×108 CFU per 0.5 mL dose. Skim milk powder at 5% w/v acts as a stabiliser against mechanical stress in spray nozzles, and the final solution pH is adjusted to 6.8 with sodium bicarbonate; chlorine neutralisation with 0.1% sodium thiosulfate is mandatory when using potable water. Spray equipment includes cabinet sprayers with flat-fan nozzles generating droplet sizes of 100–200 µm; nozzle blockage from undissolved powder aggregates larger than 20 µm is eliminated by in-line filtration through 20 µm nylon mesh. Terminal product is a solution for use within 2 h after reconstitution, with no preservative system because sodium thiosulfate would interfere with viability if present above 0.15%. Compliance is established under Ph. Eur. 0062 and Ph. Eur. 5.2.6; pH is measured per Ph. Eur. 2.2.3 and sterility per Ph. Eur. 2.6.1. Field-scale failures include uneven droplet distribution when cabinet relative humidity exceeds 70%, causing wetting of chick down and reduced preening-mediated ingestion. Published data for this specific configuration is limited, but production records indicate that spray vaccination achieves uniform flock coverage only when the target droplet size is maintained within ±20 µm of the 150 µm design point.

The Salmonella vaccine veterinary-grade API is supplied as a lyophilized, formalin-inactivated antigen concentrate under model family SVA-VG-7, with two sub-models: SVA-VG-7-NS for oral solid-dose processing and SVA-VG-7-S for aseptic injection and solution compounding. The powder contains inactivated whole-cell antigens of Salmonella enterica subsp. enterica serovars Typhimurium, Enteritidis, and Dublin in a 3:3:2 antigenic mass ratio, with sucrose:trehalose 3:1 w/w as the lyoprotectant matrix and phosphate-buffered saline salts at pH 7.2 ± 0.2. Each 100 mg of lyophilized API contains 48–55 mg of antigenic protein as determined by quantitative SDS-PAGE densitometry; lipopolysaccharide content is controlled to ≤ 50 EU/mg antigen for the oral grade and ≤ 0.5 EU/mg for the injectable grade. The product is intended for incorporation into immediate-release tablets, capsules, powders, granules, premix, and reconstituted injectable solutions for veterinary immunoprophylaxis against salmonellosis in poultry, swine, and cattle. The API is not a final dosage form; release is contingent on meeting the specification panel in Table 1 and the processing limits described below.
Direct compression of SVA-VG-7-NS requires a controlled particle-size envelope. The lyophilized powder is jet milled at 0.8–1.2 kg/h feed rate and 6.0–7.5 bar grinding pressure to yield a d10 of 2.5–4.0 µm, d50 of 9–13 µm, and d90 ≤ 24 µm by laser diffraction ISO 13320:2020. When blended with microcrystalline cellulose, mannitol, and croscarmellose sodium 2.0% w/w, antigenic mass retention after compaction at 12–22 kN on a 16-station rotary tablet press is not less than 90% as measured by capture ELISA. Lubrication is restricted to magnesium stearate 0.25–0.50% w/w; higher lubricant levels prolong disintegration and reduce antigen release in pH 6.8 phosphate buffer. Tablets compressed outside the 12–22 kN range exhibit capping at compression forces above 25 kN and friability failures above 1.0% at forces below 10 kN. Ejection force is monitored at 350–600 N; deviations above 750 N are rejected due to risk of punch tip scoring and heat-induced antigen denaturation. Content uniformity per USP <905> meets acceptance value ≤ 10 for dose units containing 25–50 mg of the API. Tablet formulation is limited to direct compression or dry granulation; wet granulation with aqueous binder above 40°C leads to re-hydration of the lyophilized matrix and moisture-induced antigen aggregation.
Injection-grade SVA-VG-7-S is reconstituted aseptically in phosphate-buffered saline at pH 7.0–7.4 and filtered through a 0.22 µm membrane before filling. The lyophilized powder for injection is released with sterility per USP <71> and bacterial endotoxin content not more than 0.5 EU/mg antigen by USP <85>. Terminal steam sterilisation is contraindicated because heating above 60°C for more than 30 minutes reduces antigenic mass by more than 25%; therefore, the manufacturing line operates under Grade A aseptic conditions. Residual moisture for the injectable grade is tightened to ≤ 2.0% w/w by USP <921> Method Ia. Reconstituted solutions should be used within 8 h at 2–8°C; after 8 h, particulate formation above 10 µm is detectable by light obscuration USP <788> Method 1 because of slow antigen aggregation. The API is incompatible with acidic reconstitution buffers below pH 4.0 and with aluminum hydroxide adjuvants in the same vial; gel precipitation is immediate.
SVA-VG-7-NS is formulated into granules using a top-spray fluidised-bed granulator equipped with a 1.2 m bowl and a Schlick 970 nozzle. The binder solution is 5.0% w/w povidone K-30 in purified water, sprayed at 20–30 g/min with inlet air at 50–60°C and product temperature not exceeding 38°C. Aqueous spray rate above 30 g/min increases bed relative humidity above 75% and produces agglomerates with d50 above 200 µm; such agglomerates segregate during premix blending. Granule moisture after drying is held at 1.5–2.5% w/w. For premix use, the dried granules are diluted with spray-dried lactose or calcium carbonate to a final antigen concentration of 2.0–5.0 mg/g. Blend uniformity at 10-minute intervals in a 500 L twin-shell blender is acceptable at relative standard deviation ≤ 5.0% for 10 samples, as measured by ELISA. SVA-VG-7-NS does not require cold-chain handling for granulation; however, the blend must not be exposed to 40°C for more than 72 h because antigenic mass loss reaches 8–12% under these conditions. Moisture-protective packaging with desiccant is specified for any granule retained more than 14 days.
Endotoxin reduction is the main differentiator between SVA-VG-7 and conventional whole-cell Salmonella bacterin APIs. Conventional inactivated Salmonella whole-cell powders obtained by downstream alum precipitation may retain endotoxin burdens above 2,000 EU/mg antigen, limiting their use in injectable and neonatal oral formulations. The SVA-VG-7 manufacturing sequence applies a butanol-water two-phase extraction after inactivation, followed by tangential-flow filtration across a 100 kDa polyethersulfone membrane and lyoprotectant addition. This reduces lipopolysaccharide to ≤ 50 EU/mg antigen for the oral grade and ≤ 0.5 EU/mg antigen for the sterile injectable grade. For injectable products, the USP <85> endotoxin limit is calculated from the maximum adult cattle dose of 250 µg antigen, yielding a per-dose endotoxin allowance of 0.125 EU. For neonatal poultry oral formulations, the oral grade is used; pellets and granules are manufactured at a water activity of ≤ 0.45 to suppress residual microbial growth. SVA-VG-7-S should not be combined with aluminum phosphate adjuvants in a single vial; charge interference causes immediate flocculation and antigen loss above 30% after 2 h.
Formaldehyde inactivation is carried out with 0.20% w/w formalin for 48 h at 37°C under agitation in 50 L stainless-steel jacketed vessels. The oral grade SVA-VG-7-NS has a residual formaldehyde release limit of ≤ 0.02% w/w; the injectable grade SVA-VG-7-S is tightened to ≤ 0.01% w/w because parenteral products must comply with lower aldehyde exposure expectations. Residual formaldehyde is determined by derivatization with 2,4-dinitrophenylhydrazine and reversed-phase HPLC with photodiode array detection at 360 nm. Batch-to-batch variance in residual formaldehyde is typically 0.008–0.014% w/w for 12 consecutive production batches. Incompatibility is documented with primary amine excipients, including glycine and lysine, which react with residual formaldehyde to form Schiff bases and lower antigen recovery by up to 15%; formulations using these excipients should be avoided or pre-validated by spiked recovery studies.

The lyophilization cycle is a further processing boundary. The liquid antigen concentrate is frozen to −45°C at 0.8°C/min, held for 4 h, then primary-dried at shelf −35°C and chamber pressure 0.2 mbar for 48 h. Collapse temperature of the sucrose-trehalose matrix is −32°C; product temperature excursion above −28°C results in cake collapse and moisture above 3.0%. Secondary drying is performed at 25°C for 6 h to reach the release moisture limit. The amorphous matrix has a glass transition temperature Tg of 45°C at 3.0% moisture; storage at 40°C/75% RH without desiccant reduces Tg to 30°C and produces caking. Sealed foil with silica-gel desiccant is therefore mandatory for products intended for tropical distribution.
SVA-VG-7 differs from oil-adjuvanted whole-cell Salmonella bacterin APIs primarily in physical form and route flexibility. Oil-emulsion bacterins are limited to injectable administration because the emulsion separates under tabletting shear and cannot be uniformly distributed in dry premix. SVA-VG-7-NS is dry, jet-milled, and compatible with direct compression and fluid-bed granulation, allowing the same antigen profile to be delivered as a tablet, capsule, granule, powder, or premix. Unlike live attenuated Salmonella vaccine strains, the SVA-VG-7 product is non-replicating, which reduces the risk of environmental shedding and the requirement for sustained cold-chain at temperatures from −20°C to −80°C. However, because the SVA-VG-7 is inactivated, primary vaccination may require a booster at 14–21 days in poultry and swine to achieve a sufficient immune response, whereas live vaccines may induce protective response after a single dose. Published peer-reviewed efficacy data for this specific lyophilized API in oral solid-dose forms is limited; current batch data establish physico-chemical consistency and accelerated stability, not field efficacy. Consequently, formulation and immunogenicity studies should be conducted before substituting it in an existing licensed final product.

SVA-VG-7-NS should not be stored at relative humidity above 60% unless the primary container is sealed with desiccant and moisture vapour transmission rate is below 0.01 g/m²/day. For capsule filling, low-dose units using size 0 capsules should be produced with the API pre-blended with pregelatinised starch at 1:5 w/w to improve flowability; the final API content per capsule should not fall below 20 mg to maintain content uniformity within acceptance value ≤ 10. For oral solutions, the reconstituted product is stable for 24 h at 2–8°C when protected from light. Avoid buffers containing acetate below pH 4.5, divalent cations above 50 mM, or vigorous stirring above 500 rpm for more than 30 minutes; these conditions accelerate antigen aggregation and invalidate the particle-size specification.
