Application of Ruyi Jinhuang Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Before direct compression of Ruyi Jinhuang Powder veterinary grade API into companion-animal tablets, particle-size classification and moisture-rheology testing determine whether the material will blind the tablet press. A d50 of 60–80 µm and a Carr index of 25–35 are typical for a dry powder that requires roller compaction rather than direct compression; production-scale tablet presses with 8–16 kN punch force are acceptable only when loss on drying is ≤ 2.0% because residual moisture above that value produces picking and sticking on 18-station rotary tooling. The regulatory baseline for these oral solids is 21 CFR Part 210–211 current good manufacturing practice, with release testing according to Ph. Eur. 2.9.7 for friability, Ph. Eur. 2.9.3 for dissolution, and USP <905> for uniformity of dosage units; residual solvent evaluation follows VICH GL18. The formulation addition ratio is derived from the certificate of analysis dry-matter assay rather than fixed arbitrarily. At an API dry-matter assay of ≥ 95.0% w/w and a target label claim of 200 mg herbal extract per 1000 mg finished tablet, the nominal API charge is 210.5 mg per tablet, corresponding to 21.1% w/w in the compression blend; batches at 18.0% w/w and 24.0% w/w are normally bracketed only after homogeneity data demonstrate CV ≤ 5.0%. Downstream production proceeds through conical milling at 0.8 mm, roller compaction with hydraulic pressure of 60–120 bar, and dry granulation through a 0.8–1.5 mm screen before tablet compression or capsule filling. Finished dosage types include uncoated and aqueous-film-coated tablets and hard gelatin or hydroxypropyl methylcellulose capsules in sealed aluminum blisters with desiccant.

What Limits Drinking Water Solubility in Poultry and Swine Dosage Forms?

Water-soluble oral powders and stock solutions for poultry and swine are constrained by the water-extractable fraction of the API and by the ionic composition of farm drinking water. Solubility screening is performed in purified water and in hard water adjusted to 200 mg/L calcium carbonate equivalents at 25 °C; turbidity is measured by Ph. Eur. 2.2.2 and must remain below the reference suspension for the lowest intended dilution. The applicable regulatory framework is Directive 2001/82/EC as amended and Regulation (EU) 2019/6 for veterinary medicinal products; non-sterile oral solutions follow microbiological quality requirements under Ph. Eur. 5.1.4. The formulation addition ratio for water-soluble concentrates is developed as a dilution factor rather than a single mass-per-dose value; a concentrate containing 10.0% w/w API is dispensed at 1.0 L per 1000 L drinking water, yielding a nominal concentration of 100 mg/L, but the farm dose is adjusted by measured daily water consumption and body weight. Downstream production uses high-shear dispersal of the API with 0.5% w/w colloidal silicon dioxide carrier, followed by spray drying at inlet temperature 160–180 °C and outlet temperature 75–85 °C to a moisture content ≤ 3.5%. Terminal finished product types include sachet-packed oral powders, bulk water-soluble powders in polyethylene-lined kraft bags, and liquid stock solutions for proportioner medicators.

In medicated feed premix and granule manufacturing for swine and poultry, the production line is treated as a high-dilution carrier system rather than a finished pharmaceutical granulation. The API is first pre-blended with maltodextrin or calcium carbonate at 5.0% w/w active content; this intermediate premix is then incorporated into final feed at 0.5–1.0% w/w according to the veterinarian's prescription and the target daily intake. Compliance is anchored to Regulation (EU) 2019/4 on medicated feed and ISO 22000 food-safety management; bulk and tapped density are determined by Ph. Eur. 2.9.34, and homogeneity is monitored through HACCP critical control points. Downstream production uses a ribbon blender with 500–2000 kg capacity and a mixing time of 10–15 min at 20–30 rpm; marker compound uniformity across 10 sampling points must remain at CV ≤ 5.0% before transfer. Following blending, the product may be granulated in a fluid bed dryer at 55–65 °C or extruded through a 0.6 mm die for crumble top dressing. Terminal finished product types include medicated premix, oral granules in bulk containers, and feed top dressing for automatic dispensing lines.

Sterile Parenteral Solutions Require Endotoxin and Particulate Limits Above Dry Oral Grades

Injectable solutions incorporating Ruyi Jinhuang Powder veterinary grade API are processed under aseptic or terminal-sterilization conditions because the finished injection must meet endotoxin and particulate limits absent from oral preparations. A published monograph for this specific herbal API in injectable form is limited; therefore, formulation development is based on the certificate of analysis for bacterial endotoxins and on pharmacopoeial requirements for the finished injection. Compliance is governed by Ph. Eur. 2.6.14 for bacterial endotoxins, USP <788> for sub-visible particulate matter, Ph. Eur. 5.1.1 for sterilization methods, and EU GMP Annex 1 for sterile product manufacture. The formulation addition ratio is expressed as weight by volume; a sterile aqueous formulation may contain 0.5–2.0% w/v extract equivalent after assay adjustment, but injectable strengths must be validated by recovery studies because polysaccharide precipitation can occur if the pH falls outside 4.5–6.5. Downstream production proceeds from chilled purified water into a high-shear homogenizer at 10,000 rpm for 10 min, then through a 0.45 µm prefilter and a 0.22 µm sterilizing filter; the solution is filled into amber glass vials under nitrogen and terminally autoclaved at 121 °C for 15 min where thermal stability permits. Terminal finished product types include single-dose and multi-dose injectable solutions for large animals, with vial sizes of 50 mL, 100 mL, and 250 mL.

When Aquaculture Immersion Baths Use Herbal APIs, Dissolved Oxygen and Water Hardness Are First-Order Variables

Aquaculture applications of Ruyi Jinhuang Powder veterinary grade API appear either as short-term immersion baths for fish and crustaceans or as medicated pellet feed. In immersion systems, the powder is first pre-dispersed in aerated freshwater; dissolved oxygen is maintained above 5.0 mg/L because fine botanical solids increase biological oxygen demand. The applicable compliance framework is ISO 22000 for feed-safety management and farm-level HACCP; where the product is intended for medicated feed in aquaculture, Regulation (EU) 2019/4 and national residue-control programs may also apply, although published data for this specific configuration is limited. For immersion baths, the working dilution range is commonly evaluated at 10–30 mg/L extract equivalent; for feed inclusion, pilot trials assess 2–5 g/kg of finished feed at the extrusion stage. Downstream processing for medicated pellets uses a twin-screw extruder with L/D ratio 25:1, barrel temperatures of 85–105 °C, and post-extrusion vacuum coating of the heat-labile API after pellets cool below 45 °C; final pellet moisture is controlled at 8–10%. Terminal finished product types include floating or sinking medicated pellets, water-stable crumbles, and immersion concentrate sachets.

Ruminant Oral Drench Suspension and Feed Top Dressing Homogeneity

For ruminant administration, Ruyi Jinhuang Powder veterinary grade API is formulated as an oral drench suspension or as a feed top dressing rather than a simple dry powder because rumen passage and saliva dilution can separate fine particles from coarse carrier fractions. The compliance baseline for oral liquid suspensions includes USP <905> uniformity of dosage units and Ph. Eur. 2.9.36 uniformity of mass of delivered doses; for feed top dressing, Regulation (EU) 2019/4 and ISO 22000 apply. The formulation addition ratio for an oral drench is expressed as suspension strength, typically 5.0% w/v extract equivalent, with sodium carboxymethyl cellulose at 0.3–0.5% w/v to prevent sedimentation; feed top dressing is applied at 1.0–2.0% w/w of the daily concentrate allocation. Production of the drench requires high-shear dispersion at 3000–5000 rpm for 10–15 min, followed by homogenization at 200–300 bar and filling under clean area class D; the final suspension is tested for pourability, viscosity, and sedimentation ratio. Terminal finished product types include ready-to-use oral drench bottles, calibrated dosing containers, and top-dressing sachets for on-farm mixing.

    More Introduction

Ruyi Jinhuang Powder Veterinary Grade API (Model RJH-VAPI-2401) is supplied as a milled multi-herb powder for seven veterinary dosage-form routes: tablets, injectable solutions, capsules, oral powders, granules, premixes, and drinking-water solutions. The grade designation RJH-VAPI-2401 encodes the botanical formula RJH, the veterinary API segment, and the current process edition 2024-01. The material is controlled against two retention marker compounds, berberine hydrochloride and aloe-emodin, rather than a single isolated chemical entity. This release strategy is used because the API retains non-volatile extractive matter, micro-fiber, and a small amount of acid-insoluble ash from the botanical source; the full extractive profile is considered relevant to veterinary formulation performance.

Packaging comprises a 25 kg HDPE drum with double LDPE liners, net weight tolerance ±0.2 kg. Storage is specified at ≤25°C and ≤60% RH; under these conditions the manufacturer assigns a 24-month retest interval from date of manufacture. The product is not sterile and is not depyrogenated; it is intended as an API for further processing only. Compared with common food-grade or unstandardized crude powders, RJH-VAPI-2401 differs in particle size control, marker content, and elemental impurity documentation. The release specification requires not less than 95.0% passing a 75 µm sieve and marker assay of not less than 0.35% w/w berberine hydrochloride and 0.10% w/w aloe-emodin. Total ash is limited to ≤9.0%, acid-insoluble ash to ≤2.0%, and elemental impurities to Pb ≤5 ppm, Cd ≤2 ppm, As ≤3 ppm, and Hg ≤1 ppm by USP <232>/<233> methodology.

Differentiation from purified extract powders and single-chemical veterinary APIs is operational as well as compositional. A purified alkaloid fraction may offer better flowability but does not reproduce the co-occurring tannin and polysaccharide behavior encountered in oral and injectable products. RJH-VAPI-2401 therefore requires dosage-form-specific particle size reduction, endotoxin control, and pH management; these requirements are not present with a single-marker synthetic chemical API. In addition, the API is supplied with a chromatographic fingerprint for batch-to-batch consistency, rather than only a single assay value. This provides traceability in multi-herb supply chains but increases the analytical burden at incoming quality control.

What Limits Direct Compression Use of This Powder in Veterinary Tablets?

The powder is not a free-flowing direct-compression grade. In pilot-scale evaluation using a 45-station rotary tablet press, untreated API at compression force from 8 kN to 15 kN produced capping and lamination at press speeds above 35 rpm. Root-cause data from the same runs showed Hausner ratio 1.45–1.55 and Carr index 31–35%, both above the direct-compression usability threshold of Carr index ≤25% described in USP <1174>. The lignocellulosic fraction exhibits elastic recovery after compression, which reduces tensile strength at ejection.

Wet granulation is therefore the recommended processing route for tablet batches. In a production-scale train, the API is blended with lactose monohydrate and partly pregelatinized starch in a 600 L high-shear granulator at impeller 150 rpm and chopper 1800 rpm. Purified water is added to 9–12% w/w. The wet mass is screened through a 1.2 mm screen and dried in a fluid-bed dryer at inlet air 55–65°C to a final granule moisture of 2.5–4.0% w/w. Final compression on a 45-station rotary press at 60–80 rpm uses compression force 15–22 kN; ejection force is monitored and controlled below 1500 N. Under these conditions, 500 mg tablets achieve hardness 80–120 N and friability ≤0.8% by USP <1216>. Direct compression should be considered only if the API is co-processed with silicified microcrystalline cellulose and fumed silica; even then, blend flow must be confirmed by shear cell testing and ejection force signature on the actual production machine.

Hard capsule production with this API requires an intermediate granulation step unless a high-shear blended powder with glidant is accepted. On a production-scale tamping pin capsule filler running 60,000 capsules/h, direct powder fill of untreated API into size 1 hard gelatin capsules produced weight variation above 5.0% and frequent powder bridging, because the powder flow index remained below the acceptable mass-flow regime. Dry granulation on a roller compactor with roll force 18–25 kN/cm and screen size 0.8 mm, followed by blending with 0.5% w/w fumed silica and 1.0% w/w magnesium stearate, reduced weight variation to ≤2.0%. For oral powders and granules, the API is blended in a 2,000 kg ribbon mixer at 60% fill and 12 rpm for 10 min; a carrier of lactose monohydrate or corncob granules with particle size 150–300 µm is preferred. Final sachet filling uses volumetric auger fillers; fill weight variation is maintained at ±5.0% for 20 g sachets.

Particle Size, Moisture Sorption, and Blend Uniformity Parameters

Particle size distribution is measured by laser diffraction on a Malvern Mastersizer 3000 with dry dispersion at 2.0 bar. A typical batch shows D10 12 µm, D50 48 µm, and D90 97 µm. The milled API is packaged only after the D90 value remains ≤97 µm for three consecutive samples. Moisture sorption is evaluated by dynamic vapor sorption at 25°C; equilibrium moisture content is 7.2–8.4% w/w at 60% RH, rising to 11.0–12.5% w/w at 75% RH. Bulk handling above 65% RH is therefore restricted; drums should be closed within 30 min after sampling.

Blend uniformity in low-dose premixes is sensitive to API particle size. In a 500 kg double-cone blender at 60% fill and 14 rpm, a 1.0% active premix produced a blend uniformity acceptance value of 8 by USP <905> after 20 min when the API was milled to D90 ≤97 µm. The same formulation using unmilled API with D90 >180 µm produced an acceptance value above 15, indicating failure. Geometric dilution with 5.0 kg API added to 50 kg carrier before full-scale mixing is mandatory for active concentrations below 2.0%. The release specification and typical batch data are summarized in Table 1.

When Terminal Sterilization Is Required for Injectable Solutions

Injectable solutions prepared from RJH-VAPI-2401 require aqueous extraction rather than simple dissolution. The extraction liquor contains pectin-like polysaccharides, tannins, and calcium oxalate microcrystals that can foul sterilizing-grade membranes. At production scale, the clarified extract is first passed through a 0.45 µm PVDF prefilter and then a 0.22 µm PES membrane at a controlled feed pressure ≤1.5 bar. Two-stage filter capacity is typically 300–500 L/m² on 0.22 µm membrane before differential pressure increases by 0.5 bar, after which the membrane train is replaced. The acceptable pH window is 4.5–6.5. At pH above 8.0, accelerated stability at 40°C for 48 h produces visible particulate and a marker assay loss above 5.0%; at pH below 3.5, acid hydrolysis of glycosidic linkages increases free anthraquinone aglycones and changes the chromatographic fingerprint.

Terminal sterilization at 121°C for 15 min is acceptable only after thermal marker mapping on three consecutive production batches. If the assay loss exceeds 5.0%, aseptic filtration through 0.22 µm is substituted. Avoid combination with benzalkonium chloride in cationic preservative systems; turbidity has been observed due to anionic polyphenol interactions. Type I borosilicate glass vials with halogenobutyl closures are used because published sorption data for polycarbonate or PVC containers with this specific multi-herb extract is limited.

Premix production on farm-scale feed blending equipment requires the API to be diluted to a working concentration of 2.0–5.0% w/w in a non-hygroscopic carrier before final feed mixing. In a 1,500 kg twin-shaft paddle mixer at 20 rpm, a 10-min blend time after geometric dilution yields a coefficient of variation ≤5.0% by USP <905> stratified sampling at 10 points. The finished premix is packaged in aluminum foil-lined multi-wall bags with desiccant; moisture pickup under 25°C/60% RH remains below 0.8% w/w over 24 months. For drinking-water solutions, the formulator should use a buffered vehicle at pH 4.5–6.5 and a 0.2% w/w xanthan gum suspending agent; high-shear mixing at 1,500 rpm for 20 min produces a viscosity of 250–450 mPa·s and a 24-h sedimentation ratio not greater than 0.9. Hard water with calcium above 120 mg/L should be chelated or softened before use, because polyphenolic fractions form haze and sediment in the presence of divalent cations. Published data for this specific product in tropical zone IVb long-term storage is limited; local stability qualification is required before zone IVb distribution.
