Application of Qiwei Dangao Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions
In the swine production sector, medicated feed premixture manufacturing incorporating Qiwei Dangao Powder veterinary-grade API constitutes the highest-volume downstream application for this botanical hepatoprotective ingredient, driven by the continued regulatory de-emphasis on antibiotic growth promoters under Chinese Ministry of Agriculture and Rural Affairs Announcement No. 246 and the corresponding demand for non-antibiotic liver-support formulations in grower-finisher herds affected by mycotoxicosis-associated hepatobiliary dysfunction, biliary stasis, and icteric syndromes. The API powder, characterized by a bulk density of 0.38–0.52 g/cm³ and a particle size specification requiring ≥95% passage through an 80-mesh (≤180 μm) screen per Chinese Veterinary Pharmacopoeia (CVP) 2020 general chapter requirements, is introduced into horizontal ribbon mixer systems (L/D ratio ≥2.0:1, shaft speed 20–50 rpm, tip speed 0.5–1.2 m/s, fill ratio 50–65% of gross volume) at blending concentrations of 5.0–10.0 wt% for concentrated intermediary premises or at 0.15–0.30 wt% (equivalent to 1.5–3.0 kg per metric tonne) for direct therapeutic incorporation into complete finishing rations. The blending sequence requires a minimum 20-minute main blend cycle following active ingredient addition, with a post-lubricant blend interval of 3 minutes when magnesium stearate at 0.5 wt% is employed as flow modifier; blend uniformity validation under Chinese Ministry of Agriculture and Rural Affairs Order No. 41 (GMP for Veterinary Premix Manufacturing) mandates a coefficient of variation not exceeding 7.0% across no fewer than 10 sampling points distributed spatially throughout the batch. Environmental control during premix production is maintained at 18–25°C with relative humidity below 60% to prevent hygroscopic agglomeration of the botanical powder on mixer walls, shaft seals, and discharge gates—a failure mode documented on production-scale ribbon mixers when RH exceeds 65% for processing intervals longer than 90 minutes. Finished premixture moisture is controlled below 5.0 wt% (USP <731> loss on drying), total viable aerobic count below 10⁵ CFU/g per ISO 22283:2016, and heavy metal burden below the Chinese National Feed Hygiene Standard GB 13078-2017 limits (lead ≤5 mg/kg, arsenic ≤2 mg/kg). Terminal finished product formats emerging from this pathway include 5% and 10% concentrated veterinary premises in 25 kg multi-wall kraft paper bags with 60 μm interior polyethylene liners, 1 kg and 5 kg foil-laminated pouches for smallholder distribution, and 20 kg woven polypropylene bags with inner PE liner for export containerized transport under monitored atmosphere with embedded temperature loggers.

What Governs Dissolution Rate-Limiting Steps in Avian Hepatoprotective Water-Soluble Granule Systems?

When drinking-water delivery is selected as the clinical intervention route for avian hepatobiliary disease management, the conversion of Qiwei Dangao Powder veterinary-grade API into water-soluble granule dosage forms for poultry operations requires precise control of dissolution kinetics within line-distribution drinking systems, where water residence time in nipple drinker pipelines ranges from 4 to 24 hours and solution stability under barn temperature swings (18–35°C) determines clinical efficacy in broiler, layer, and breeder flocks presenting with hepatitis-associated mortality elevations or egg production depression of 10–30% from baseline. The formulation pathway employs wet granulation in a high-shear granulator (Diosna P-250, impeller speed 250–300 rpm, chopper speed 1500–2000 rpm, granulation time 5–8 minutes) followed by fluidized-bed drying (Glatt GPCG-60, inlet air temperature 55–65°C, product temperature 35–42°C, final moisture content ≤3.0 wt% by USP <731>). The granule formulation specifies the veterinary-grade API at 40–60 wt%, dextrin USP-NF at 25–35 wt% as water-soluble filler, sodium bicarbonate at 3–5 wt% as effervescent dissolution accelerator, povidone K-30 at 2–3 wt% as binder delivered via 10 wt% aqueous solution at spray rate 80–120 mL/min, and colloidal silicon dioxide at 0.5–1.0 wt% as post-granulation flow conditioner. Regulatory compliance is anchored to CVP 2020 general chapter requirements for water-soluble veterinary powders, USP <711> Dissolution Method II (paddle apparatus, 50 rpm, 900 mL deionized water at 37 ± 0.5°C, sampling at 15, 30, and 45-minute intervals with acceptance criterion of ≥80% dissolution at 30 minutes), and a dissolution medium tolerance test in hard water (500 ppm CaCO₃ equivalent) verifying ≥90% solubility without turbidity or precipitation within 30 minutes. Terminal finished products include 100 g and 500 g aluminum foil composite pouches sealed under nitrogen atmosphere, 25 kg fiber drums with double PE liner for industrial poultry integrators, and 1 kg HDPE jars with child-resistant closures for veterinary clinic dispensing; the drinking-water dosing regimen for finished granules is 0.5–1.0 g/L of drinking water for 5–7 consecutive days, reconstituted as fresh solution every 24 hours to minimize microbial proliferation in water delivery infrastructure.

Compression Dynamics, Disintegration Profiling, and Binder Selection for Ruminant Oral Bolus Tablets

For ruminant production systems managed under extensive grazing regimes, the formulation of Qiwei Dangao Powder veterinary-grade API into large-format compressed tablets addresses the operational constraint of individual-animal dosing in cattle, buffalo, sheep, and goat herds where feed-based medication is impractical due to grazing management patterns, feed refusal by individual animals, or the requirement for precise therapeutic dose delivery during acute hepatitis episodes. The direct-compression pathway is preferred over wet granulation to preserve heat-labile botanical constituents—specifically the iridoid glycosides present in the Gentianaceae and Fructus Gardeniae fractions, degradation of which accelerates above 60°C during aqueous processing, with kinetic stability studies indicating a first-order degradation constant k of 0.012–0.025 h⁻¹ at 70°C in aqueous slurry conditions. The tablet formulation specifies API content at 50–60 wt% (equivalent to 3.0–6.0 g active botanical powder per 10 g bolus), microcrystalline cellulose PH-102 at 25–30 wt% as compressible filler, croscarmellose sodium USP-NF at 3–5 wt% as superdisintegrant, pregelatinized starch at 5–8 wt% as binder, and magnesium stearate (vegetable source) at 0.5–1.0 wt% as lubricant added in the final 3-minute blend cycle. Rotary tablet press operation (ZP-35B, 35-station, B-tooling, compression force 15–25 kN, pre-compression 3–5 kN, turret speed 25–35 rpm) produces bolus tablets of 5.0–10.0 g weight with hardness 60–100 N (USP <1217> Tablet Breaking Force), friability ≤0.5% after 100 revolutions (USP <1216>), and disintegration time ≤30 minutes in water at 37 ± 0.5°C per USP <701>. Process compliance is governed by CVP 2020 monograph specifications for veterinary tablet preparations, China GMP (2010 revision) for non-sterile solid dosage forms, and EU GMP Part II alignment for export-market validation. Terminal finished products include 10 g cattle bolus tablets in 50-count and 100-count HDPE jars with induction-sealed foil liners, 2 g and 5 g sheep/goat tablets in 10-count aluminum-PVC blister packs, and bulk 500-count fiber drums for veterinary hospital repackaging operations.Encapsulation operations for companion animal hepatobiliary support formulations utilizing Qiwei Dangao Powder veterinary-grade API commence with a powder flow characterization sequence that is mandatory for achieving fill weight uniformity within ±5% per USP <905> Uniformity of Dosage Units; the API blend must demonstrate a Carr compressibility index below 20 and a Hausner ratio below 1.25 following a 10-minute low-shear V-blender premixing cycle with lactose monohydrate NF (diluent, 35–45 wt%), microcrystalline cellulose PH-101 (compression aid, 5–10 wt%), and colloidal silicon dioxide (glidant, 0.5–1.0 wt%, charged in the final 2 minutes). The API content per capsule is specified at 40–60 wt%, corresponding to 80–300 mg of active botanical powder in finished capsule sizes selected by species: Size 3 capsules (0.30 mL fill volume) for feline patients at 200–250 mg total fill weight, and Size 0 capsules (0.68 mL fill volume) for canine patients at 500–680 mg total fill weight. Encapsulation is performed on a Bosch GKF-1500 intermittent-motion machine at 20,000–30,000 capsules/hour with automatic weight verification, operating in an environment controlled to 18–25°C and RH 35–50%, with the hygroscopic botanical powder transferred to the machine hopper only after moisture content verification at ≤4.0 wt% by USP <731> loss-on-drying. Regulatory frameworks applicable to this downstream pathway include FDA 21 CFR Part 211 (Current Good Manufacturing Practice for Finished Pharmaceuticals) for US-market veterinary products, FDA 21 CFR Part 514 for new animal drug application filing where applicable, USP <2040> for gelatin capsule disintegration (acceptance: complete disintegration of 6 test capsules within 15 minutes in simulated gastric fluid TS), dissolution testing per USP <711> Method I (basket, 100 rpm), and stability evaluation per VICH GL3 (Zone II conditions: 30°C/65% RH long-term, 40°C/75% RH accelerated). Terminal finished products include 10-count and 20-count PVC/aluminum foil blisters with child-resistant outer cartons, 30-count and 60-count amber HDPE jars with desiccant canisters, and veterinary hospital repack dispenser strips.

When Bacterial Endotoxin Limits and Particulate Load Dictate Parenteral Formulation Viability of Botanical APIs

Sterile parenteral manufacturing of Qiwei Dangao Powder veterinary-grade API is constrained at the outset by the bacterial endotoxin load inherent to crude botanical powders—typically 10³–10⁵ EU/g before purification—and the particulate burden generated during aqueous decoction extraction, including suspended fiber fragments, resinous agglomerates, and tannin-protein complexes that must be removed to satisfy USP <788> particulate matter requirements. The injectable formulation pathway commences with double extraction of the crude powder at 95–100°C for 60 minutes per cycle using purified water USP at a 1:8–1:10 drug-to-solvent ratio, followed by vacuum concentration in a falling-film or rotary evaporator at ≤70°C under −0.08 MPa, ethanol precipitation at 60% v/v final concentration with 24-hour cold settling (2–8°C), removal of precipitated polysaccharides and proteins by plate filtration, and serial adsorption with pharmaceutical-grade activated charcoal (0.5–1.0 wt%, 30 minutes at 70°C) to reduce pyrogen burden. The clarified extract is then adjusted to a concentration equivalent to 0.5–1.0 g crude botanical powder per mL injection solution, with pH adjustment to 5.5–7.0 (USP buffer system), osmotic pressure adjustment to 280–320 mOsm/kg using sodium chloride USP, and the addition of 0.1–0.2 wt% sodium metabisulfite as antioxidant in multidose vial presentations. Terminal sterilization by moist heat at 121°C for 15 minutes (F₀ ≥12) is acceptable only when validated to preserve marker iridoid glycoside content within 90–110% of labeling; alternative aseptic processing through 0.22 μm PVDF membrane filters (Sartorius Sartopore 2, filtration area 0.8 m² per 100 L batch) under ISO Class 5 (Grade A) unidirectional airflow is used when heat-labile constituent degradation exceeds this threshold. Compliance is governed by EU GMP Annex 1 (sterile medicinal products), USP <71> sterility testing (membrane filtration method, fluid thioglycollate and soybean-casein digest media, 14-day incubation), USP <85> bacterial endotoxin testing (turbidimetric method, limit ≤2.5 EU/mg of active botanical equivalent), USP <788> particulate matter in injections (light obscuration particle count: ≤6000 particles ≥10 μm and ≤600 particles ≥25 μm per container for small-volume parenterals), and container-closure integrity testing per USP <1207>. Terminal products are 10 mL Type I borosilicate glass vials sealed with bromobutyl rubber stoppers and aluminum flip-off caps in 10-vial cartons, 5 mL amber glass ampoules for single-dose administration in neonatal calves, and 20 mL multidose vials for equine practice. Published comparative pharmacokinetic data for the injectable presentation of this specific botanical formula versus the oral premix route remains limited, and the manufacturing operation requires dedicated facilities with Grade C background areas and Grade A filling zones—a capital expenditure threshold that materially constrains contract manufacturer entry into this product category.

Concentrated Oral Drench Manufacture and Sedimentation Stability Profiles for Neonatal Ruminant Administration

In neonatal ruminant practice, the clinical requirement for forced oral administration of hepatoprotective botanical preparations arises when icteric mucous membranes, biliary stasis, or acute toxic hepatitis preclude voluntary feed or water intake, and the concentrated oral drench manufacturing pathway for Qiwei Dangao Powder veterinary-grade API addresses this therapeutic gap through a liquid suspension format designed for direct buccal dosing via syringe or drench gun. The suspension formulation specifies the crude API at 0.20–1.0 g equivalent per mL (20–100 wt/vol%), with xanthan gum USP-NF at 0.1–0.3 wt% as thixotropic suspending agent providing a yield stress of 1.5–8.0 Pa to minimize sedimentation, microcrystalline cellulose/carboxymethylcellulose sodium (Avicel RC-591) at 0.5–1.0 wt% as co-structural network former, polysorbate 80 at 0.05–0.1 wt% as wetting agent, sodium benzoate at 0.3 wt% plus potassium sorbate at 0.15 wt% as preservative system requiring pH adjustment to 4.5–5.5 for optimal antimicrobial efficacy (benzoic acid pKa 4.19), and citric acid/sodium citrate buffer at 0.05 M to maintain the target pH window. The production sequence consists of high-shear dispersion (Silverson L5M-A, 10,000 rpm, 5–10 minutes) for complete powder wetting, colloid mill passage (IKA MK-2000, rotor-stator gap 50–100 μm, 2–3 passes) for particle size reduction to D90 ≤50 μm (laser diffraction, Malvern Mastersizer 3000), vacuum deaeration at −0.05 to −0.08 MPa for 30 minutes to reduce entrained air, and automated piston filling into 100 mL, 500 mL, or 1 L HDPE bottles with graduated oral dosing pumps (1 mL graduation) or 10 mL dosing syringes. Regulatory compliance integrates CVP 2020 general chapter requirements for oral suspensions, USP <51> antimicrobial preservative effectiveness testing (category IV products), USP <61> microbial enumeration with USP <62> specified microorganism testing (absence of Escherichia coli per 1 mL), VICH GL3 stability requirements, and EU GMP Part I for non-sterile liquid manufacturing. The prescribed dosing regimen for finished drench products is 1–2 mL per 10 kg body weight administered twice daily for 3–5 consecutive days, delivered via oral dosing syringe or drench gun into the buccal cavity. Terminal packaged formats include 100 mL presentations for small ruminant flock operations, 500 mL and 1 L bottles for dairy calf intensive operations, and 5 L HDPE jugs for veterinary hospital dispensing.Across caged layer operations and segregated swine starter facilities, direct oral granule administration of Qiwei Dangao Powder veterinary-grade API via dry granulation roller compaction or extrusion-spheronization constitutes an alternative delivery strategy for situations where drinking-water medication is contraindicated by water system maintenance procedures, where feed premix incorporation is operationally infeasible during short-duration therapeutic windows, or where individual animal dosing requires a palatable, free-flowing granular physical form suitable for top-dressing onto feed or direct oral administration. The dry granulation route, selected when the API powder exhibits poor flow characteristics (Carr index ≥25) or when moisture-sensitive constituents preclude aqueous granulation, employs roller compaction (Alexanderwerk WP 120, roll pressure 50–100 bar, gap 2–4 mm) followed by milling (rotary granulator with 1.0–1.5 mm screen) to produce granules with a target particle size distribution of 0.5–1.5 mm (minimum 85 wt%), final moisture ≤4.0 wt%, and bulk density 0.50–0.65 g/cm³. The formulation for dry granulation specifies API content at 50–70 wt%, with mannitol USP at 20–30 wt% as non-hygroscopic filler, pregelatinized starch at 5–10 wt% as binder, and magnesium stearate at 0.5–1.0 wt% as lubricant. Alternatively, extrusion-spheronization (Fuji Paudal EXD-60 extruder, 0.8–1.2 mm die, spheronizer speed 600–1000 rpm, spheronization time 3–5 minutes) followed by fluidized-bed drying at 55–60°C inlet temperature yields spherical granules with narrower particle size distribution (0.8–1.2 mm) and superior flow properties; this route requires microcrystalline cellulose PH-101 at 20–30 wt% as spheronization aid and water as granulating fluid at 35–45 wt% of dry blend. The regulatory framework for this pathway includes CVP 2020 bulk granule specifications, China GMP for non-sterile solid dosage forms, and particle size analysis per USP <786>. Terminal finished products include 500 g and 1 kg HDPE jars with measuring scoops calibrated to deliver 5 g per level scoop, 5 kg and 10 kg foil-lined bags for farm-level administration, and 25 kg drums for further processing into finished feed by integrator mills. The administration rate for finished granules is 50–100 mg/kg body weight per day for 5–7 days, administered by top-dressing onto individual feed portions in controlled feeding operations.

    More Introduction

Qiwei Dangao Powder Veterinary Grade API is a multi-component pharmaceutical powder specified for formulation into tablets, injections, capsules, powders, granules, premix, and solutions. The material is not controlled as a single chemical entity; release acceptance is therefore defined by a combination of pharmacopoeial general-chapter limits and a validated chromatographic marker profile, which is supplied as a lot-specific certificate of analysis. Commercial dossiers may reference model codes such as QDP-VG-API-100M, but these identifiers are manufacturer-specific and do not replace the full specification matrix. The powder is normally assigned a maximum loss on drying of 5.0% and a residue on ignition limit of 5.0% for routine oral-grade material, with tighter endotoxin and particulate requirements when the same API is designated for parenteral processing. Heavy-metal control is commonly set at 10 mg/kg or lower under the receiving manufacturer’s supplier qualification procedure. The particle-size target for direct compression and capsule filling is a laser-diffraction D90 below 150 µm; for granules and premix operations, a coarser sieve cut between 80 µm and 180 µm may be selected to reduce dusting and segregation. Table 1 lists representative specification parameters against the test methods used in veterinary API release.

Why Does Direct Compression Demand Tighter Particle-Size and Moisture Targets Than Wet Granulation?

Direct compression of Qiwei Dangao Powder at API loadings between 20% and 40% by weight is feasible only when the powder flow function satisfies a Hausner ratio below 1.35 and the median particle size is maintained near 100 µm. Rotary tablet presses such as a Korsch XL 400 or Fette 1200i with compression force set at 8–22 kN are used for 750 mg target tablets. At higher API loading, the brittle-ductile transition of the dried matrix causes capping when compaction pressure exceeds 25 kN; addition of microcrystalline cellulose at 30–50% and crospovidone at 2–5% improves tensile strength. If loss on drying exceeds 5.0%, sticking to upper punches becomes significant under continuous run conditions above 50,000 tablets/h. The granulated route uses a top-spray fluid-bed granulator with inlet air temperature 60–75°C and spray rate 10–20 g/min/kg for a binder solution of povidone K30 at 5% solids. Published data for this specific API in direct compression are limited; therefore, the compression profile should be confirmed by compaction simulator studies before registration stability batches.

When the same powder is declared for injection formulation, the control strategy shifts from particle flow to particulate and endotoxin burden. A sterile injectable solution is prepared by reconstitution in Water for Injection at 5–20 mg/mL, followed by pH adjustment with 0.1 M sodium hydroxide or hydrochloric acid to a target pH between 6.0 and 7.5, depending on the extract’s zeta potential and active marker stability. The solution is prefiltered through a 0.45 µm polyethersulfone membrane and then sterile-filtered through a 0.22 µm membrane under aseptic conditions in an ISO 14644-1 ISO 7 cleanroom. Terminal sterilization may not be used if the marker compounds degrade at 121°C for 15 min; this limitation must be verified by forced degradation studies. Subvisible particulate matter is controlled to USP <788> limits for small-volume parenterals: not more than 6,000 particles per container at 10 µm and not more than 600 particles per container at 25 µm. Endotoxin acceptance for the reconstituted solution is set at 0.5 EU/mg of API. If insoluble residues exceed 0.5% w/w after reconstitution, filtration throughput at 0.22 µm drops sharply, and the dosage form may require reformulation as a suspension rather than a solution.

Premix Homogeneity and Segregation Risks Are Controlled by Median Particle Size Alignment

For medicated premix manufacture, the API is blended with a mineral or lignocellulose carrier in a ribbon mixer with volume 500–1,000 L. Active constituent migration between mixer discharge and bagging is minimized when the API median particle size is within 80–120 µm and the carrier particle size is 600–850 µm. Segregation tests using a validated sampling thief procedure should demonstrate relative standard deviation below 5.0% across 10 sampling points. A horizontal paddle mixer operating at 25 rpm for 15 min typically reaches blend uniformity for a 2% active premix; extending mix time beyond 25 min increases electrostatic adhesion to the vessel walls and lowers recoverable active concentration. The finished premix is then diluted into final feed at 0.5–1.0% inclusion. Unlike unrefined multi-component powders, the standardized particle-size distribution of this API reduces variation in feed assay from batch to batch.

Capsule filling with Qiwei Dangao Powder at API doses between 100 mg and 500 mg per unit is performed on dosator or tamping-pin machines. Powder blends containing 30% API and 70% lactose monohydrate plus 0.5% magnesium stearate are suitable for encapsulation when the Carr index is below 25. Granules for oral administration are produced by roller compaction with a roll force of 10–15 kN/cm and a roll gap of 1.5–2.5 mm, then milled through a 1.0 mm screen. The resulting sieve fraction between 250 µm and 850 µm is retained for filling. Powders for direct administration are sieved through an 850 µm screen and mixed with dextrose or sodium chloride as a diluent to provide a uniform dose per scoop. Oral solutions prepared from the powder require preservative compatibility testing because benzalkonium chloride at 0.01% may precipitate with acidic marker fractions.

When Oral Solutions Require Filtration and Preservative Stability, the Formulation Order Determines Intermediate Precipitation

Oral solution preparation from the powder involves dissolution in purified water at 40–50°C, followed by cooling to 20–25°C before addition of sorbitol or glycerol as viscosity modifiers. If the API is added to a premixed preservative solution, local concentration gradients can cause agglomeration of poorly soluble fractions; therefore, the powder is first dispersed in water with high-shear mixing at 3,000 rpm for 10 min. The solution is polished through a 10 µm polypropylene depth filter to remove insoluble cell fragments. The pH is then adjusted to 5.5–6.5 to balance marker stability and preservative activity. Samples held at 40°C and 75% RH for 6 months are monitored for appearance, pH shift, and marker content. Differences from synthetic solutions include increased filter fouling and the need for a validated antioxidant package such as 0.1% sodium metabisulfite; however, compatibility with sulfite-sensitive species must be assessed before a batch is released.
