Application of Qiwei Banlangen Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

For sterile aqueous injectable preparations based on Qiwei Banlangen Powder, the primary processing obstacle is not active ingredient solubility but the removal of cold-insoluble mucilage and high-molecular-weight polysaccharide fractions that can blind sterilising-grade membranes. A production-scale batch is usually reconstituted in Water for Injection heated to 80–85 °C, held for 30–45 min under vacuum-assisted high-shear mixing to fully wet the botanical matrix, then transferred to a jacketed settling vessel and cooled to 4–8 °C for a minimum of 12 h. The resulting flocculated polysaccharide layer is separated with a disc-stack centrifuge operating between 8,000×g and 12,000×g, followed by depth filtration and a 0.45 µm pre-filter. Final bioburden reduction filtration through a 0.22 µm polyethersulfone cartridge is installed immediately before the filling line. This sequence is necessary to meet USP <788> particulate matter thresholds for small-volume parenterals; filter fouling is routinely observed when the cold-settling step is shortened below 10 h, with transmembrane pressure rising above 1.0 bar before 50 L/m² throughput on a 0.45 µm membrane.

Formulation addition ratios screened at pilot scale fall between 1.0% w/v and 5.0% w/v dry API, with the lower concentration preferred when isotonicity must be achieved without large corrections of sodium chloride. Published data for this specific multi-herb injection configuration is limited; therefore the ratio is confirmed by marker retention studies and particulate recovery rather than by reference to a harmonised monograph value. After pH adjustment to 6.5–7.5 with phosphate buffer, the solution is sparged with nitrogen and filled under ISO 14644-1:2015 Class 5 conditions; terminal steam sterilisation is performed at 121 °C±2 °C for 15 min when marker stability permits, or at 115 °C±2 °C for 30 min for heat-sensitive batches. Compliance testing includes Ph. Eur. 2.6.1 for sterility, Ph. Eur. 2.6.14 or USP <85> for bacterial endotoxins, and USP <788> for particulate matter. Terminal finished product type is a sterile injectable solution in 10 mL or 20 mL amber Type I borosilicate vials sealed with bromobutyl rubber closures, intended for intramuscular administration in swine and cattle where authorised.

What Limits Dissolution Rate in Poultry Drinking-Water Soluble Powders?

The limiting factor for a water-soluble powder containing Qiwei Banlangen Powder is not active content but the particle architecture left after drying. When the dried powder is presented as a directly filled blend, fines below 100 µm can float on the drinking-water surface and create hydrophobic microaggregates that block nipple drinkers. Production-scale batches therefore use wet granulation: the API and a hydrophilic carrier such as lactose or sorbitol are blended, then wetted with purified water or a 5% w/v povidone K30 binder solution in a high-shear granulator at impeller speed 150–250 rpm and chopper speed 1,500–2,500 rpm. The wet mass is extruded through a 1.0 mm screen and dried in a fluid-bed dryer with inlet air at 55–65 °C until loss on drying is ≤5.0%; dried granules are sized to 250–500 µm using a vibratory sieve. This particle-size window keeps the wetting time below 3 min when 100 g of powder is dispersed in 10 L of water at 25 °C with gentle stirring; published data for this specific botanical powder is limited, but the target is routinely verified by a beaker wettability test before release.

Formulation addition ratio for field administration in poultry barns is typically screened between 1.0 g/L and 3.0 g/L of drinking water for short-duration programmes, though the registered target-species dose must be confirmed against the approved dossier. The API content in the finished powder is adjusted so that one 100 g sachet treats 100–300 L of water; therefore the API inclusion rate in the dry powder falls in the range of 30–60% w/w depending on the carrier system. Compliance standards include VICH GL18 for stability evaluation, Ph. Eur. 2.6.12 for microbial quality of non-sterile products, and USP <921> water determination for residual moisture. Terminal finished product type is a water-soluble powder packed in 100 g or 500 g aluminium-foil sachets or 1 kg HDPE jars, designed for administration through header tanks or proportional dosing pumps.

In feed-mill premix handling, a multi-component botanical API exposes a different failure mode than pharmaceutical water-soluble processing: segregation in ribbon blending and pneumatic conveying after the API pre-blend is added to the carrier. The raw Qiwei Banlangen Powder is first pre-diluted with ground corn cob, rice hulls, or soybean hulls at a ratio of 1:5 to 1:10 by weight to prevent electrostatic segregation of the fine botanical fraction. Pre-blend particle size is reduced through a cone mill or hammer mill fitted with a 0.5–1.0 mm screen, producing a carrier-active mixture with a coefficient of variation ≤5% across ten sampling points after 12–15 min in a paddle ribbon mixer. In production-scale feed mills, the pre-blend is metered into the final premix by loss-in-weight feeder at an accuracy of ±1%; batch records commonly show that bulk density differences between the botanical powder and the carrier cause unacceptable assay variability if the premix is pneumatically conveyed over distances longer than 20 m, and therefore the pre-blend is added after the conveying line rather than before it.

Inclusion rates in complete feed are screened between 0.05% and 0.20% by weight, corresponding to 500–2,000 g/tonne in finished feed for swine and poultry; higher rates are generally reserved for short-term interventions and must be justified by target-species registration data. The terminal premix powder is filled into 1 kg, 5 kg, or 25 kg multi-wall paper bags with a polyethylene liner, and a desiccant pouch is inserted when the powder is shipped to humid climates because the botanical powder is hygroscopic above 60% relative humidity. Compliance anchors are ISO 6496:2005 for moisture and volatile matter, ISO 6497:2005 for sampling, EC 183/2005 for feed hygiene, and 21 CFR 507 for animal food CGMP in the United States market. Terminal finished product type is a dry feed premix powder intended for inclusion in complete compound feed at the feed mill, not for direct oral administration.

Controlling Binder Level and Drying Capacity During Low-Shear Wet Granulation

Dose accuracy in a graduated drenching gun or individual sachet, rather than dispersal behaviour in water, defines the granule size and binder level for oral granules. In a low-shear planetary mixer, the API is combined with microcrystalline cellulose and lactose, then wetted with a binder phase prepared from pregelatinised starch or hypromellose at 5–10% w/v; binder addition is targeted at 8–12% w/w of the dry mass. Overwetting above 12% w/w produces elongated, strong granules but extends tray-drying time beyond 18 h at 45–55 °C, while underwriting below 8% w/w creates a friable mass that generates fines above 15% after sieving. The dried granulate is passed through a 1.0 mm oscillating granulator and the 500–1,000 µm fraction is collected for final packaging; fines below 500 µm are recycled into the next wet mass at a maximum rate of 20% to avoid accumulation of heat-degraded botanical fines.

Formulation addition ratio of Qiwei Banlangen Powder in the granule core falls within 40–60% w/w, with the remainder composed of filler, binder, and a lubricant such as sodium stearyl fumarate at 1–2% w/w if flow into the auger filler becomes marginal. Batch-to-batch variance in the bulk powder tapped density between 0.30 g/cm³ and 0.60 g/cm³ is the main cause of fill-weight drift on single-auger dosing machines; therefore the granule density is normalised by compaction before dry granulation or by load-cell verification on the filling line. Compliance testing follows Ph. Eur. 2.9.12 for sieve analysis, USP <921> for water determination, and VICH GL18 for stability classification. Terminal finished product type is an oral granule packed in 20 g, 100 g, or 1 kg foil-lined sachets, suspended in water or mixed into a small amount of feed immediately before administration.

When a high-volume oral drench is preferred for swine or calves, the stability-critical factor shifts from particulate control to preservative efficacy and microbial challenge at dilute pH. The API is hydrated in purified water at 40–50 °C for 30–45 min with continuous agitation, then cooled before addition of a buffering system. Citric acid and sodium citrate are used to hold the final pH between 4.0 and 5.5; in multi-dose packaging, potassium sorbate or sodium benzoate is added at 0.1–0.2% w/w and must be validated by a USP <51> antimicrobial effectiveness test against the target microbial panel. Published data for this specific botanical oral solution is limited, so preservative load is not extrapolated from oral solutions of single chemical actives; the lowest effective concentration is determined by challenge testing on three consecutive production batches.

Formulation addition ratios for the oral drench are screened from 2.0% w/v to 5.0% w/v dry API, with the higher bound limited by viscosity rise and settling after 7 days of accelerated storage at 40 °C. Glycerol at 10–15% v/v is included as a viscosity-modifying and sweetening carrier; the solution is passed through a 100 µm nylon bag filter or cartridge before filling into amber HDPE bottles. Terminal finished product type is a multi-dose oral solution in 500 mL, 1 L, or 5 L containers with tamper-evident caps and graduated dosing cups. Compliance testing includes Ph. Eur. 2.6.12 for microbial examination of non-sterile products and USP <51> for preservative effectiveness.

If High Active Loading Is Required in a Direct Compression Tablet Process

Direct compression of Qiwei Banlangen Powder into veterinary tablets becomes feasible only when the botanical powder is pre-processed to improve compactability and when the active loading is kept below 50% w/w of the core. The API is first milled and passed through a 500 µm screen; pre-lubricated blends are then prepared with microcrystalline cellulose at 30–50% w/w, spray-dried lactose or anhydrous dibasic calcium phosphate at 10–20% w/w, croscarmellose sodium at 2–4% w/w as disintegrant, and colloidal silicon dioxide at 0.5–1.0% w/w as glidant. Magnesium stearate is added last at 0.5–1.0% w/w and mixed for no longer than 3–5 min to prevent compaction defects. Compression is performed on a rotary tablet press at 8–15 kN compression force, with target hardness between 70 N and 120 N and friability ≤1.0% according to Ph. Eur. 2.9.7.

    40%10–14 kN85–110 NAcceptable
    50%12–15 kN100–120 NEdge capping above 50%

Formulation addition ratio of the API is screened at 30–50% w/w; at loadings above 50% w/w, the mixture shows elastic recovery after ejection and capping at the edge of the tablet, a failure recorded on production presses when turret speed exceeds 30 rpm without pre-compression. The finished tablets are produced in 250 mg or 500 mg active-powder strengths, with a total core weight of 700–1,000 mg for the 500 mg strength. Disintegration testing follows USP <701> with water at 37 °C; the target is ≤15 min for immediate-release veterinary tablets. Terminal finished product type is an oral tablet for sheep, goats, or calves, packed in aluminium/PVC blisters or HDPE bottles with induction-sealed caps. Compliance anchors include Ph. Eur. 2.9.8 for resistance to crushing, USP <905> for content uniformity, and USP <701> for disintegration.

Capsule Filling Constraints for Multi-Herb Powder Blends with High Hygroscopicity

For volumetric capsule filling of Qiwei Banlangen Powder, the primary constraints are hygroscopicity and shear-resistant bridging in the powder hopper. The API is dried to loss on drying ≤5.0% before blending, and the blend is prepared with colloidal silicon dioxide at 0.5–1.0% w/w plus a filler such as microcrystalline cellulose or pregelatinised starch to bring the active loading to 60–75% w/w. If the tapped density of the final blend falls below 0.35 g/cm³, the blend is dry-granulated with a roller compactor and then milled to a granule size between 200 µm and 800 µm to improve flow into a dosator-type encapsulator. Published data for this specific botanical capsule configuration is limited; therefore the fill weight and capsule size are confirmed by content uniformity on three validation batches rather than by comparison with a compendial standard.

Formulation addition ratio is calculated to deliver 300–500 mg of the API powder per capsule, filled into size 0 or size 1 hard gelatin or hypromellose capsules. The encapsulation room is maintained at ≤40% relative humidity; filled capsules are packaged in HDPE bottles with a desiccant canister and induction-sealed closure to limit moisture uptake. Terminal finished product type is an oral capsule for companion animal veterinary practice where such use is authorised by the target-region dossier. Compliance testing includes USP <905> for uniformity of dosage units, USP <701> for disintegration, and USP <921> for water determination.

    More Introduction

Qiwei Banlangen Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions is supplied under article code QWB-VAPI-100 as a brownish-yellow to brownish-green fine powder derived from the root of Isatis indigotica. The material is milled to a controlled particle-size distribution, with representative release targets of d50 ≤ 75 µm and d90 ≤ 150 µm by laser diffraction under ISO 13320:2020. The powder passes through an 80-mesh sieve at not less than 95% by weight. It is packaged in 25 kg net lots in double-layer polyethylene-lined fiber drums with an aluminum foil barrier layer. The same article code may be released for tablets, injectable solutions, capsules, powders, granules, premixes, and oral solutions; therefore the batch-specific certificate of analysis must be reviewed against the target dosage form. The powder is not sterile as supplied. Injectable lots receive additional bioburden and endotoxin controls, and the finished-product manufacturer is responsible for validated sterilizing filtration or terminal heat treatment where feasible.

For nonsterile dosage forms, QWB-VAPI-100 is incorporated at a concentration defined by the registered veterinary medicinal product specification and the marker content of the batch. The powder is hygroscopic and should be pre-dried at 60°C to 65°C under vacuum if loss on drying exceeds 5.0% after storage. This drying step should not exceed 4 h because prolonged heating reduces the (R,S)-goitrin marker. These statements describe material handling characteristics, not finished product release criteria.

Assay variability in commercial Banlangen powders is influenced by geographical origin, harvest year, and drying history. The (R,S)-goitrin marker is thermally sensitive; extraction temperatures above 70°C lower marker recovery, so the manufacturing route holds extraction at 60°C to 65°C under reduced pressure. Indirubin and indigotin are monitored as secondary markers because their quantitative relationship to activity is not established across all target species. Total polysaccharides are measured by phenol–sulfuric acid and expressed as glucose equivalents; a single marker assay is not sufficient for supplier qualification. The full chromatographic fingerprint, polysaccharide profile, and loss on drying should be compared across batches. Published data for this specific veterinary-grade configuration is limited; therefore incoming acceptance should include at least two marker compounds and a bioburden screen until site-specific historical data exist.

Residual solvents are controlled under ICH Q3C for the extraction solvent used. If ethanol is present in the extraction step, the powder is dried under reduced pressure until ethanol content is below 5000 ppm or the registered monograph limit, whichever is lower. Pesticide residues are controlled by USP <561> and Ph. Eur. 2.8.13 multi-residue procedures. Aflatoxins B1, B2, G1, and G2 are controlled below 2 μg/kg total and below 1 μg/kg for B1 when the powder is used in dry premix or granule operations. Drying technology should be specified on the purchase order because spray-dried material may have lower bulk density and more amorphous character than vacuum-dried material, and this changes direct compression behavior.

What limits direct use of crude Isatis root powder in injectable preparations?

Crude Isatis root powder contains lignified vascular fragments, soil-borne bacterial spores, and an uncontrolled particle-size tail that fails the particulate matter requirements of USP <788> after reconstitution. For injectable finished products, the veterinary API is manufactured as a clarified extract, dissolved or dispersed in water for injection, and filtered through a 0.22 µm PVDF or PES membrane. A 0.45 µm prefilter is placed upstream to protect the sterilizing filter from pigment aggregates and polysaccharide gels. Terminal moist-heat sterilization at 121°C for 15 min may degrade (R,S)-goitrin; heat-sensitive injectable formulations therefore rely on aseptic filtration. The filtered solution is filled into depyrogenated Type I glass vials under Grade A laminar flow. The finished solution is tested for subvisible particles according to USP <788>, for sterility according to USP <71>, and for bacterial endotoxin according to USP <85>. Endotoxin control is dose-justified: at a veterinary dose of 5 mg/kg, a lot with 0.50 EU/mg delivers 2.5 EU/kg, which may be acceptable or excessive depending on the regional monograph; the limit must be evaluated under VICH GL18 and EU Regulation 2019/6. For high-dose injectable products above 10 mg/kg, an endotoxin limit below 0.20 EU/mg is often required unless administration is limited to oral or intramammary routes.

Filter capacity is a practical bottleneck in injectable manufacturing. Dissolving the powder at 5.0 mg/mL to 50 mg/mL in water for injection at 35°C with high-shear mixing at 8000 rpm for 15 min reduces visible pigment aggregates. After cooling to 20°C, the solution is passed through the prefilters and sterilizing filter with a maximum transmembrane pressure of 1.0 bar. Batches with high total polysaccharide content can require a filter area of approximately 0.2 m2 per 10 L; this value is configuration-dependent and must be confirmed by a filterability study. Clarification by centrifugation at 3000×g for 15 min before filtration reduces subvisible particles that may otherwise pass the membrane and cause light obscuration failures.

When aqueous solubility and pH fall outside the expected range

At 2.0% w/v in distilled water at 25°C, the unadjusted pH of QWB-VAPI-100 is typically 5.0–6.5. If the solution pH is driven below 4.0, precipitation of indirubin-type components increases and filterability decreases. If the pH exceeds 8.0, oxidative darkening and (R,S)-goitrin loss accelerate. Oral solutions are therefore hydrated first and then buffered, commonly with citrate–phosphate to pH 5.5–6.5. The powder has low intrinsic buffering capacity, so acids or bases should not be added to the dry powder because local pH excursions can damage the marker fraction. In premix manufacture, calcium carbonate and sodium bicarbonate are not recommended as carriers, because alkaline microenvironments reduce (R,S)-goitrin stability during storage. Lactose monohydrate is preferred over dextrose for premixes that are extruded or pelletized above 65°C, because dextrose can participate in browning reactions that bind the API and reduce water extractability. For solution products, polysaccharide-derived viscosity increases with concentration; above 10.0% w/v, the product may behave as a non-Newtonian liquid and should not be processed through narrow-bore filling needles without a viscosity validation.

Evaluating blend uniformity and segregation risk in premix operations

QWB-VAPI-100 has a bulk density of 0.40–0.65 g/mL and a tapped density typically 0.55–0.80 g/mL. The Carr Index may exceed 25 when the powder is stored at high humidity, indicating poor flow. In premix blending, 0.5% w/w colloidal silicon dioxide is used as a glidant when the API is first blended with a carrier such as lactose monohydrate. A ribbon blender of 500 kg working capacity is charged with the carrier, the glidant, and the API; a preliminary mixing time of 10 min at 25 rpm is suitable for many formulations, but published data for this specific configuration is limited. The mixing time must be confirmed by a site-specific blend uniformity study. Samples are taken from 10 locations and assayed for (R,S)-goitrin; a relative standard deviation below 5.0% is a typical release criterion. Segregation can occur during transfer if the blend is pneumatically conveyed over long distances or dropped from a height greater than 1.5 m. In these cases, a densified granulated premix is preferred over a simple dry blend.

Direct compression of QWB-VAPI-100 without prior granulation is feasible when the API content is below 20.0% w/w. Above this level, the powder’s high elastic recovery reduces compact hardness and increases capping risk. On a rotary press with 8 mm flat-faced punches, a blend of 15.0% API, 84.0% microcrystalline cellulose PH102, and 1.0% magnesium stearate yields tablets with hardness 60–80 N and friability below 1.0% after 100 rotations in a USP <1216> friabilator. For higher API loads, wet granulation with a 5.0% w/w hydroxypropyl cellulose binder in a high-shear granulator operating at impeller speed 300 rpm and chopper speed 1500 rpm is used, followed by fluid-bed drying at inlet air 60°C to a product temperature of 35–40°C. The granulated material is dried to loss on drying ≤ 3.0% before lubrication. Capsule filling on a dosator-type machine is preferred over tamping-pin systems because the extract powder is cohesive and can develop static charges at relative humidity below 30% RH.

Microbial risk control for nonsterile powders and granules is tied to water activity. During wet granulation, water added during binder preparation raises water activity above 0.60; the wet mass should be dried immediately and not held longer than 12 h at room temperature. Fluid-bed dryers with product temperature 35–40°C and final moisture ≤ 3.0% reduce water activity below 0.60. In premix operations, extended storage at > 60% RH may increase total aerobic microbial count; re-testing is recommended after 6 months in tropical climates.

Difference from unstandardized Banlangen extracts and synthetic antivirals

Unstandardized root powder differs from QWB-VAPI-100 in particle size, microbial load, and marker content. Crude powder is not suitable for injectable or highly dispersed liquid dosage forms. Fluid extracts may contain ethanol or glycerin, which complicates dry blending and increases residual solvent control requirements. QWB-VAPI-100 is a dry extract with controlled residual solvent, a defined particle-size upper limit, and a release assay for (R,S)-goitrin. Compared with synthetic single-entity antiviral agents, the veterinary-grade Banlangen powder is multicomponent: the marker fraction includes (R,S)-goitrin, indirubin, indigotin, and water-soluble polysaccharides. No single marker can fully describe the activity of the product, and batch-to-batch equivalence should be supported by chromatographic fingerprint similarity and polysaccharide content, not by marker assay alone.

Stability, packaging, and nonsterile processing boundaries

QWB-VAPI-100 is hygroscopic and should be stored in the original sealed container at ≤ 25°C and ≤ 60% RH. After opening, the remaining powder should be re-sealed under nitrogen if the local relative humidity exceeds 60%. Pre-drying is required when loss on drying exceeds 5.0%; use a vacuum dryer at 60°C to 65°C for not more than 4 h to minimize marker loss. The powder contains pigmented indirubin and indigotin components; equipment contact surfaces require cleaning validation with HPLC detection of indirubin to avoid cross-contamination. Strong oxidizing agents can degrade the sulfur-containing marker (R,S)-goitrin, so peroxide-based cleaning or blending agents should not contact the API unless validated. The product should not be blended with strong acids or alkali metal hydroxides in the same processing train without a documented cleaning boundary. For granules, a fluid-bed top-spray process with 5.0% povidone K30 binder solution at spray rate 20–30 g/min per kg batch, inlet air 60–70°C, and product temperature 35–40°C is used. The dried granules are sieved to 16–40 mesh; fines below 60 mesh may be recycled, but the recycle fraction should not exceed 20% of the batch because recycled fines shift particle-size distribution and reduce flow.

The product is intended for veterinary use and is not a sterile API. For food-producing species, withdrawal period decisions require residue data and should follow EU Regulation 2019/6 or the applicable regional veterinary medicinal product framework. Pharmacopoeial compliance for the finished dosage form must be established separately from the API release specification. The manufacturer should maintain a stability program based on ICH Q1A(R2)/Q1E principles with the actual packaging configuration.
