Application of Qingre Jianwei Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions
On a rotary tablet press equipped with forced feeders, Qingre Jianwei Powder is not charged directly without pre-blending. The powder’s botanical polysaccharide and residual plant fiber fractions make it hygroscopic; moisture uptake above 5.0% w/w causes sticking to upper punches and increases ejection force variation. A direct compression matrix is prepared by dry-screening the API through a 595 µm guard sieve and blending 65.0% w/w API with 20.0% w/w microcrystalline cellulose, 10.0% w/w anhydrous lactose, 3.0% w/w croscarmellose sodium, 1.0% w/w colloidal silicon dioxide, and 1.0% w/w magnesium stearate in a 500 L bin blender at 12 rpm for 20 min. The colloidal silicon dioxide is co-screened with the API before the main blend step to reduce interfacial friction rather than being added as a final powder. Magnesium stearate is added last and blended for 3 min. Lubrication beyond 5 min produces a hydrophobic film over the botanical particles and delays disintegration to more than 15 min under USP <701>. The blend is compressed on a 27-station rotary tablet press at 12–25 kN compression force. Tablets with a target weight of 500 mg and thickness of 3.2–4.0 mm are produced. Breaking force is measured with a Schleuniger-type hardness tester and kept between 40 N and 80 N; friability is tested per USP <1216> with a maximum loss of 1.0% after 100 revolutions. In-process control records punch force, ejection force, turret speed, tablet mass, and thickness. The terminal dosage form is a film-coated tablet; a 3.0% w/w HPMC coating is applied in a side-vented pan to mask bitter botanical taste. The coating dispersion is prepared at 10.0% w/w solids in purified water and sprayed at 40–50 °C product temperature. Coated tablets are packaged in HDPE bottles with silica gel desiccant and stored below 25 °C and 60% RH.

When a veterinary injectable is processed without terminal sterilization

When a parenteral grade of Qingre Jianwei Powder is selected, the route is normally intramuscular or subcutaneous administration in cattle or swine. Terminal steam sterilization at 121 °C for 15 min is excluded if botanical marker compounds are heat-labile; the process therefore relies on aseptic filtration and aseptic filling into sterile vials. The API is dissolved in Water for Injection at 25–35 °C with continuous low-shear stirring. The resulting solution is cooled to 20–25 °C and adjusted to pH 5.8–6.5 with 0.1 M citric acid or 0.1 M sodium hydroxide. If the pH drifts above 7.0, phenolic components oxidize and the solution darkens; if the pH falls below 5.0, water-soluble polysaccharides can aggregate and increase turbidity. A two-stage filtration train is used: a 0.45 µm polyethersulfone prefilter followed by a 0.22 µm PVDF sterilizing filter. The maximum differential pressure across the sterilizing filter is limited to 1.0 bar to prevent membrane rupture and particle shedding. Bacterial endotoxin control begins with API acceptance. The final formulated solution is tested by the limulus amebocyte lysate method per USP <85>. A common internal limit is calculated as K/M, where K is 5 EU/kg for parenteral products and M is the maximum recommended dose in mg/kg. If the calculated limit falls below 0.5 EU/mg, additional depyrogenation of the API may be required, which can alter particle surface and is generally avoided by qualifying the supplier with an endotoxin specification. The filling line is maintained at ISO 14644-1:2015 Class 5. Media fills are performed quarterly with a target contamination rate below 0.1%, per PIC/S Annex 1. Vials are depyrogenated at 250 °C for 30 min in a hot-air tunnel; stoppers are steam-sterilized. Filled volumes are checked at 2.0 mL, 5.0 mL, 10.0 mL, and 20.0 mL configurations. If lyophilized for injection, the solution is filled into vials and freeze-dried with annealing at -10 °C, primary drying at -25 °C and 0.2 mbar, and secondary drying at 25 °C for 8 h.Uniform capsule fill weight for milled Qingre Jianwei Powder is obtained only when the particle size distribution is shifted from a broad plant-mill distribution to a narrower range. The powder is passed through a 250 µm screen; if d(0.9) exceeds 250 µm, partially intact leaf fragments act as low-density carriers and cause dosator overfill or underfill depending on orientation. A tamping-disc capsule filler is preferred over dosator nozzles for this API because the compressed plug can be formed in 5–6 tampings at low pin pressure. The powder blend consists of 40.0% w/w API, 35.0% w/w microcrystalline cellulose, 19.0% w/w pregelatinized starch, 4.0% w/w sodium starch glycolate, 1.0% w/w colloidal silicon dioxide, and 1.0% w/w magnesium stearate. Total blend time after lubrication is 3 min at 10 rpm. Fill weight is monitored at 30-min intervals; RSD must remain ≤4.0% before release limits are adjusted. In-process average fill weight is set to ±5.0% of target. Content uniformity is evaluated by USP <905>; an acceptance value of ≤15.0% is applied. Finished capsules are size 0 for a 250 mg fill weight and size 1 for a 200 mg fill weight. Capsule shell moisture is controlled because storage at RH above 60% causes gelatin cross-linking and potential dissolution failure. The terminal product is packaged in 60 mL HDPE bottles with a 1 g silica gel canister and an induction-sealed cap. If routine dissolution testing is required, a USP <711> apparatus 2 at 50 rpm in 900 mL pH 1.2 buffer is a starting method, but published data for this specific configuration is limited and method transfer must be verified for the relevant marker compounds.

When does blend segregation become functionally significant in oral powder production?

For oral powders administered by top-dressing on feed or by drench gun, the controlling variable is not simply active content but particle size segregation after mixing. The herbal powder is typically sieved through 60 mesh and blended with anhydrous dextrose or glucose in a 500 L ribbon mixer at 70–80% working volume. Mixing time is 20 min at 8–12 rpm. Blend uniformity is assessed by sampling at 10 positions, including dead spots under baffles and at the discharge gate. An RSD of ≤5.0% for the marker compound is required before discharge. If the powder is packaged in sachets, the fill targets are 5.0 g, 10.0 g, or 25.0 g. Product contact parts are 316L stainless steel. The filling room is maintained at RH ≤50%. Moisture gain above 5.0% causes caking and failure of the flow test. A bulk density of approximately 0.55–0.65 g/mL is maintained by adding 0.5% w/w hydrophobic fumed silica. Silica addition above 1.0% w/w depresses bulk density below 0.55 g/mL and increases dusting, which compromises operator safety during sachet filling. The terminal finished product is sealed in aluminum-lined sachets and released under the veterinary monograph with storage below 25 °C and ≤65% RH.

In a 10 L high-shear granulator with a three-blade impeller, the wet mass of Qingre Jianwei Powder reaches a usable endpoint when impeller power draw increases by 30–50% from the dry mix baseline. The binder is PVP K30 at 5.0% w/v in purified water or 30% ethanol-water. Liquid addition rate is 10–15 mL/min. The impeller speed is set at 150–250 rpm, and the chopper at 1000–1500 rpm for 2–4 min. The wet mass is discharged through a 6 mm sieve and dried in a fluid-bed dryer at inlet air temperature 50–65 °C. Product temperature is kept below 45 °C to prevent thermal degradation of the botanical marker fraction. Drying is stopped at 2.0–4.0% loss on drying. Overdrying below 2.0% produces excessive fines that reduce granule strength and increase tablet friability. Underdrying above 4.0% produces sticking on the 16-mesh screen and increases microbial risk. After dry milling through a 1.0 mm screen, granules are classified to 16–40 mesh. Fines below 40 mesh are recycled at a maximum of 20.0% w/w of the final batch weight to avoid overgranulation and content non-uniformity. The dried granules are then blended with 0.5% w/w magnesium stearate and compressed into tablets or filled into sachets as a granulated oral dosage form. Terminal granule products are packaged in sealed metallized film sachets under nitrogen if the botanical extract is oxygen-sensitive.

Premix homogeneity and carryover validation boundaries

Medicated feed premix production with Qingre Jianwei Powder is performed under regional veterinary feed GMP frameworks. The mineral or grain carrier is selected according to particle size compatibility, typically a calcium carbonate or dried corn cob carrier with moisture below 10.0%. The API is first blended in an intermediate premix at 2.0–5.0% w/w active concentration and then diluted into final feed at 0.2–0.5% w/w. Mix uniformity targets for the intermediate premix require a coefficient of variation ≤5.0%. Samples are collected with a grain probe from 10 positions, including the mixer discharge, and analyzed for marker compounds by HPLC. The mixer is considered acceptable only if CV is ≤5.0% and all individual assay values fall within ±10.0% of theory. Carryover is tested by flushing the line with ground corn or wheat middlings after the medicated batch. The flush material is assayed, and a maximum carryover of ≤0.5% of the active concentration in the next non-medicated batch is imposed for veterinary feed premises. Load-out auger speed is limited to 1.5 m/s, and drop height is kept below 1.0 m to reduce segregation. The terminal product is a bagged medicated premix in 25 kg multiwall paper sacks with a polyethylene liner. Storage is in a dry, ventilated warehouse at ≤30 °C and ≤65% RH. Use of the premix is restricted to target species specified in the local veterinary authorization, and the product must not be diverted to human food-producing animals without the appropriate withdrawal period.

Oral solution clarity is controlled by sequential pH adjustment, not endpoint pH alone

Drinking water or oral solution dosage forms require reconstitution of Qingre Jianwei Powder in purified water at a concentration of 1.0–2.0 g/L. Hard water above 200 mg/L calcium carbonate equivalent causes precipitation of water-soluble polysaccharide complexes. The solution is therefore buffered with citric acid 0.5 g/L and sodium citrate 0.2 g/L to pH 5.5–6.5. pH adjustment must be carried out stepwise under gentle agitation; adding concentrated sodium hydroxide directly to the tank produces local pH spikes that denature proteins and increase turbidity. Chelation with EDTA is not used if the formulation is destined for organic-certified animals. The final solution is passed through a 100-mesh stainless steel inline strainer before filling into 1 L, 5 L, and 20 L containers. A preservative system is selected from sodium benzoate 0.1% w/w and potassium sorbate 0.1% w/w, but only after microbial challenge testing per Ph. Eur. 5.1.3. The use period in drinking water is 24 h unless stability data support 48 h. Water line compatibility is tested with PVC and stainless steel; copper lines should be avoided because copper ions accelerate oxidation of polyphenolic components. The terminal product is a filled oral solution in opaque polypropylene or high-density polyethylene containers to reduce light exposure. Batch release requires pH, visual clarity, active marker assay, and preservative content testing before distribution into veterinary channels.

    More Introduction

Qingre Jianwei Powder Veterinary Grade API, product code QJW-VET/API-25, is a multi-constituent botanical starting material produced for downstream incorporation into tablets, injections, capsules, powders, granules, premix, and solutions. The standard oral/premix grade is identified as QJW-VET/API-25; the injection grade is QJW-VET/API-25I, differing by additional micronization, depyrogenation, and aseptic handling controls. Published multi-batch data for this specific botanical configuration remain limited; release criteria therefore follow the manufacturer's technical file together with current veterinary pharmacopoeial general chapters and the United States Pharmacopeia general chapters referenced below.

The standardized marker profile is determined by gradient high-performance liquid chromatography with diode-array detection. The material contains protoberberine alkaloids, flavone glycosides, and water-soluble marker compounds. Unlike a single-entity synthetic antipyretic API, identity is established by a chromatographic fingerprint rather than a single infrared spectrum or melting point. Content is expressed as percentage marker by dried weight. Dosage-form calculations should be based on a designated marker, not total powder mass alone, because the extractable marker fraction changes with granulation, drying, and filtration conditions.

The powder's release criteria set the tableting window

The standard oral/premix grade is a brown to dark tan hygroscopic powder. Particle-size distribution is determined by laser diffraction under ISO 13320:2020; a typical standard batch shows D10 approximately 25 µm, D50 approximately 65 µm, and D90 ≤ 150 µm. The injection-grade powder is micronized to D90 ≤ 25 µm to permit aqueous reconstitution and terminal filtration. Bulk density falls between 0.35 g/mL and 0.55 g/mL, and tap density between 0.50 g/mL and 0.75 g/mL by USP <616> Method I. Loss on drying by USP <731> is controlled at ≤ 5.0% w/w for oral/premix material and ≤ 2.0% w/w for injection grade after vacuum drying.

Direct compression with the standard grade is not recommended without granulation. The powder's moisture sorption isotherm increases sharply above 60% relative humidity; in a non-dedicated suite, unbound water raises the risk of punch filming and capping on a rotary tablet press operating at 10 kN precompression and 15 kN main compression. Wet granulation with 5% w/w polyvinylpyrrolidone K30 in aqueous solution, followed by fluid-bed drying at inlet air 60°C ± 5°C and product temperature not exceeding 45°C, yields granules with residual moisture 2.0–3.5% w/w and D50 120–180 µm. The dried granules are lubricated with 0.5% w/w magnesium stearate and 0.5% w/w colloidal silicon dioxide in a bin blender for 5 min. Lubrication longer than 15 min reduces tablet hardness by overcoating the granule surfaces. Under these conditions, tablet friability is maintained below 0.8% when tested by USP <1216>. The granulation window is narrow: residual moisture below 2.0% w/w increases friability, while moisture above 4.0% w/w creates sticking and weight variation.

For capsule filling, the dried granule fraction is screen-milled to D90 ≤ 75 µm and conditioned to residual moisture below 4.0% w/w. Electrostatic charge is controlled by keeping the mill discharge and capsule machine contact surfaces bonded and by maintaining suite relative humidity at ≤ 40%. Low-shear tumble blending with 0.25% w/w sodium lauryl sulfate may be used to improve wetting, but the final excipient ratio is determined by the target fill weight and capsule size.

Why is the injection-grade powder assigned a separate endotoxin specification from the oral premix grade?

Injectable solutions prepared from a botanical API require stricter pyrogen and particulate control than oral powders or feed premixes. The oral/premix grade is acceptable at a bacterial endotoxin limit of ≤ 20 EU/g, but the injection-grade powder must be depyrogenated and must meet ≤ 0.5 EU/mg by USP <85> after reconstitution. Sterility is not assumed; sterile powder for injection is released under USP <71>. Subvisible particle counts are controlled by USP <788> after reconstitution in water for injection.

The preferred sterilisation strategy is terminal filtration, not autoclaving. The reconstituted solution is adjusted to pH 6.0–7.5, clarified through a 0.45 µm polyethersulfone membrane, and terminally filtered through a 0.22 µm polyethersulfone membrane. Autoclaving at 121°C is avoided because sustained heat can degrade heat-labile marker compounds and shift the chromatographic fingerprint. Aseptic filling is performed in Grade A laminar airflow over a Grade B background in accordance with EU GMP Annex 1. The injection-grade powder is handled with water-for-injection washing, controlled-temperature drying, and inert-gas micronization to reduce surface oxidation.

When the same API is milled for sterile injection, capsule fill, and feed premix

The route-specific grades diverge at final particle-size reduction and drying. Capsule-grade material is screen-milled to D90 ≤ 75 µm and may be dry-granulated. Injection-grade material is micronized under nitrogen to D90 ≤ 25 µm to limit amorphous surface generation and oxidation. Feed-premix material is generally blended as received or compacted with mineral oil or soybean oil at 0.5–1.5% w/w to reduce dust and segregation. For solutions, the standard powder is extracted in purified water at 50–60°C for 3 h with continuous agitation; the extract is cooled to 25°C, filtered, and preservative-free or preserved according to target multidose use. Pectin-like polysaccharides in the botanical matrix can increase cold-filtration time, so filter-area sizing and hold time must be confirmed in pilot runs.

Processing routes by dosage form are summarised as follows:

Against a single-entity synthetic antipyretic or stomach-regulating veterinary API, the primary distinction is release on multiple markers rather than a single active substance. A synthetic API is typically controlled at 98.0–101.0% w/w assay with defined impurity limits; the botanical API is controlled by chromatographic fingerprint and marker content ranges. Blend uniformity, dissolution, and stability-indicating methods therefore cannot be reduced to one marker. In vivo response cannot be inferred from a single marker plasma concentration because the product includes multiple extractable constituents with different pharmacokinetics. This does not make the product inferior or superior; it means the technical file and finished-product authorization must contain matrix-specific methods and marker-based specifications.

Compatibility boundaries in tannin-rich premix bases and alkaline mineral carriers

Blending with incompatible carriers can reduce chemical availability and content uniformity. The protoberberine alkaloid fraction forms insoluble tannate complexes in contact with gallotannin-rich botanical carriers, causing assay recovery to fall when tested by USP <621> HPLC. Strongly alkaline mineral feeds may dissociate the alkaloid salts and increase the non-ionised fraction, altering solubility and in vivo release. The powder should not be blended simultaneously with hot alkaline mineral carriers above 60°C; if the premix base contains such carriers, a compatibility trial is conducted before scale-up. The standard oral/premix material is stored at 15–25°C and ≤ 40% RH in sealed foil laminate bags with desiccant. Once opened, product should be used within 72 h or re-dried and re-analyzed for moisture. Residual solvents are controlled by USP <467> because extraction and drying solvents may remain at trace levels. Heavy metals are controlled by USP <232>/<233> using inductively coupled plasma mass spectrometry; limits are ≤ 5 mg/kg lead, ≤ 2 mg/kg arsenic, ≤ 0.5 mg/kg cadmium, and ≤ 0.1 mg/kg mercury.

Because the product is a multi-constituent botanical API, it is not interchangeable with single-entity veterinary synthetic antipyretics in residue depletion calculations. Withdrawal periods must be assigned at the finished-pharmaceutical-authorization level after tissue depletion studies are conducted according to VICH residue depletion guidance and any regional residue regulations. The powder is intended for use in GMP-compliant veterinary product manufacturing; it is not a direct-to-farm finished product. Published data for this specific botanical configuration are limited, and pilot-scale compatibility studies with the target dosage-form matrix are required before full-scale production.
