Application of Nainiu Fanchu Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Terminal Sterilisation Envelope and Endotoxin Control for Parenteral Presentation

Terminal sterilisation of parenteral presentations represents the highest specification-demanding route among the seven listed dosage forms. Nainiu Fanchu Powder Veterinary Grade API destined for sterile injectable manufacturing is subjected to a multi-stage sequence beginning with dissolution in Water for Injection (WFI) conforming to Ph. Eur. 5.1.1 at a concentration range of 50–200 mg/mL. The resulting bulk solution is adjusted to pH 6.0–7.5 using 0.1 M hydrochloric acid or 0.1 M sodium hydroxide, both of pharmacopoeial grade, before clarification through a 0.45 µm polypropylene pre-filter followed by sterilising-grade filtration through a validated 0.22 µm PVDF membrane cartridge with a bubble point of at least 1.2 bar per ASTM F838-20. Endotoxin burden in the finished sterile solution is maintained below 0.5 EU/mg per USP <85>, with LAL testing executed on every sub-lot. Terminal sterilisation is conducted in a saturated steam autoclave at 121°C for 15 minutes (F0 ≥ 8, Ph. Eur. 5.1.1 method B), unless thermal degradation profiling mandates aseptic filtration alone, in which case Grade A (ISO 5) laminar flow filling with concurrent media fill validation is required. Particulate matter in the finished container is controlled to USP <788> limits (NMT 6000 particles ≥ 10 µm and NMT 600 particles ≥ 25 µm per container). Container-closure integrity is verified by dye ingress testing per USP <1207>. Storage of the finished sterile solution in Type I amber borosilicate glass vials at 15–25°C is specified to mitigate photodegradation of the aromatic constituents. Batch sizes for sterile injectable production typically range from 50 L to 500 L, processed in 316L stainless steel compounding vessels equipped with bottom-mounted magnetic stirrers to avoid dead-leg contamination. The vial filling line operates at speeds of 2000–8000 units/hour depending on fill volume (50 mL or 100 mL), with in-process weight checks performed every 15 minutes targeting a fill volume tolerance of ± 3%. Lyophilisation is not routinely employed for this API class; however, when enhanced shelf-life beyond 24 months is specified, a lyophilisation cycle at −40°C primary drying and +25°C secondary drying (vacuum ≤ 0.1 mbar) produces a cake residue of NMT 2% moisture, enabling a 36-month expiry assignment with supporting ICH Q1E data. The selection of rubber stopper composition is restricted to bromobutyl formulations with low extractable profiles to prevent interaction with alkaline hydrolysable groups. Finished product types are sterile injectable solutions in 50 mL and 100 mL Type I glass vials with bromobutyl rubber closures and aluminium flip-off caps.

At the feed mill scale, incorporation of Nainiu Fanchu Powder Veterinary Grade API into complete dairy TMR premixes follows a step-up blending protocol specified under FAMI-QS and GMP+ BA2 feed safety assurance schemes, with the final premix registered under EU Regulation 1831/2003 where applicable. The undiluted API powder is first blended with a mineral or botanical carrier—limestone flour, rice hulls, or dried sugar beet pulp—at a ratio of 1:10, followed by a second dilution to 1:100 in a 500 kg ribbon mixer, and finally incorporated into the production-scale 2-tonne horizontal ribbon mixer at a target inclusion of 0.05–0.2% w/w in the complete feed. Homogeneity testing per EU Regulation 152/2009 method B requires a coefficient of variation (CV) below 5% across 10 sampling points, with the sampling plan including three points from the mixer discharge gate, three from the bagging line, and four from stored bagged product to account for segregation during handling. Bulk density of the API powder measured by a 1 L cylinder method (ISO 697:1981) falls within 0.45–0.65 g/cm³, and when blended with low-density botanical carriers, electrostatic charge accumulation is mitigated by the addition of 1–2% refined vegetable oil to the ribbon mixer during the final 2 minutes of mixing. Carryover control is mandatory under FAMI-QS; residual active substance in subsequent non-medicated batches is maintained below 2% of the target dose, verified by rinsate swab testing at the mixer wall, discharge auger, and bagging spout. Particle size distribution of the API powder is specified at D50 150–300 µm and D90 below 500 µm by laser diffraction (ISO 13320:2020), with sieve cuts above 800 µm rejected due to segregation tendency in TMR mixing wagons. The ribbon mixer is typically operated at 40–60 RPM with a mixing time of 8–12 minutes; over-mixing beyond 15 minutes increases the risk of particle attrition and may elevate fine particle fraction, which in turn increases dust generation and electrostatic adhesion to mixer walls. The bagging line utilises a horizontal form-fill-seal machine with nitrogen flushing to displace oxygen headspace and reduce oxidative stress on unsaturated carrier oils. Each batch of premix requires a Certificate of Analysis including active substance assay, moisture content, aerobic plate count (NMT 10⁵ CFU/g per USP <2021>), and Salmonella absence (per ISO 6579-1:2017). Storage stability in multilayer paper bags with LDPE inner liner is confirmed at 25°C / 60% RH for 24 months per VICH GL21, with moisture ingress limited to NMT 0.5% w/w per year. The finished premix format is a 20 kg bag for commercial dairy farms and a 500 kg bulk bag for feed mill integration. Inclusion rate at the farm level for TMR preparation is determined by the registered feeding guidelines and the target daily dose per adult dairy cow, typically 50–150 g of premix per cow per day delivered via the mixing wagon.

What Determines Capping Propensity During Direct Compression of Ruminant GI Function Modulators?

Direct compression of Nainiu Fanchu Powder Veterinary Grade API into oral tablets for cattle is constrained by the powder's viscoelastic recovery characteristics, which induce capping when the compression force exceeds the formulation's plastic deformation threshold. Formulation development studies conducted on single-station instrumented tablet presses (Korsch XP 1, 10 kN load cell) demonstrate that the API alone exhibits a compactibility below 60 N at a compression pressure of 120 MPa when tested per USP <1062>, necessitating the inclusion of microcrystalline cellulose (MCC grade PH102) at 30–45% w/w as a plastic binder, with lactose monohydrate (200 mesh) serving as a brittle filler at 15–25% w/w. Magnesium stearate is restricted to 0.5–1.0% w/w because higher levels significantly delay disintegration (USP <701>) beyond the 15-minute pharmacopoeial limit. The final tablet formulation typically contains 45–65% w/w API, producing a 1000 mg tablet with hardness 60–120 N (tested per Ph. Eur. 2.9.8), friability below 1% per USP <1216>, and disintegration not exceeding 15 minutes in 37°C distilled water. Film coating with hydroxypropyl methylcellulose (HPMC, 15 cP grade) at 2–3% weight gain provides a moisture barrier for tablets stored at RH > 60% and masks the bitter botanical constituents. Pre-compression force is set at 2–4 kN with main compression at 8–15 kN on a rotary press (FETTE 2090, 30 stations, 100,000 tablets/hour). The powder blend is subjected to lubrication for exactly 3–5 minutes; excess lubrication beyond 10 minutes produces measurable reductions in tensile strength due to shear-induced coating of MCC surfaces. Direct compression is selected as the default manufacturing route because the API powder's particle morphology permits adequate die filling when flow aid (colloidal silicon dioxide 0.25–0.5% w/w) is incorporated, though environments with RH below 30% may necessitate 1% w/w water addition to the pre-blend to suppress static charge during hopper discharge. Finished product types are 500 mg and 1000 mg uncoated or film-coated tablets packed in blister strips of 10 units.

Establishing Granulation Endpoint Parameters for Fluid-Bed Drying at Variable Ambient Humidity

When wet granulation is employed because the API powder's poor flowability (Carr index above 25 per USP <1174>) prevents direct compression or uniform capsule filling, the process sequence utilises a 250 L high-shear granulator (GEA Aeromatic-Fielder PMA series) with granulating liquid consisting of purified water or 5% w/v polyvinylpyrrolidone (PVP K29/32) solution at a liquid-to-solid ratio of 0.2–0.3, impeller speed ramped from 150 to 300 RPM, and chopper engaged at 1500 RPM during the final 2 minutes. Granulation endpoint is determined by power consumption inflection (GLATT Real-Time Moisture Analyser) correlated with a target moisture of 8–12% w/w. The wet mass is discharged through a 3 mm sieve and transferred to a fluid-bed dryer equipped with a 30 µm filter bag; inlet air temperature is maintained at 50–60°C, with drying continued until final moisture reaches 2–4% w/w (measured by loss-on-drying at 105°C per USP <731>). The dried granules are sized through a 14–60 mesh sieve stack (1.40 mm to 0.25 mm), rejecting oversized material for dry milling and undersized material for re-granulation. When ambient relative humidity exceeds 60%, the granulation endpoint requires adjustment: the liquid-to-solid ratio is reduced by 10% and the drying time extended by 20% to compensate for the increased moisture load in the inlet air. The API addition ratio in the final granule formulation is 30–50% w/w, with sucrose and lactose monohydrate as bulking agents and povidone as binder. The finished granules are packed in 100 g or 500 g sachets made from aluminium foil laminate with a water vapour transmission rate below 0.5 g/m²/day (ASTM F1249-20) to maintain API stability at 25°C / 60% RH for 24 months per VICH GL21. Compliance for the granule presentation is anchored to USP <795> for nonsterile compounding, Ph. Eur. 2.9.12 for flowability, and VICH GL11 for impurity profiling. Published data on the compressibility of granules produced with this specific API at scale remains limited beyond the 250 L batch size threshold.

The direct oral powder presentation for drench administration is manufactured by low-shear tumble blending in a 500 L V-blender at 15 RPM for 20 minutes, combining Nainiu Fanchu Powder Veterinary Grade API at 10–25% w/w with sodium bicarbonate (USP grade) as alkalising diluent and sucrose (NF grade) as palatability agent. The mixture is passed through a 60 mesh (250 µm) sieve to break up soft agglomerates, then packaged in 250 g and 1 kg HDPE jars fitted with 33 mm polypropylene caps and a 2 g silica gel desiccant canister to maintain moisture below 0.5% w/w during storage. Reconstitution instructions specify dispersing the powder in 5–10 L of warm water (35–40°C) per adult cow dose immediately before administration via stomach tube or dose syringe. Compliance for the powder presentation references USP <795> for nonsterile compounding and VICH GL11 for impurity limits, with terminal finished product types being 250 g and 1 kg HDPE jars. Published data on the bioavailability equivalence of this presentation relative to the granule form is limited.

When High-RH Processing Constrains Capsule Fill Weight Uniformity

Capsule filling of the API powder for individual animal dosing employs a semi-automatic encapsulator (CAP8, 8000 capsules/hour) with size 0, 1, or 2 HPMC or gelatin capsules, targeting a fill weight of 250–500 mg. The dry blend comprises 50–70% w/w API with lactose monohydrate and 0.5% magnesium stearate. Fill weight uniformity per USP <1163> requires acceptance value below 15. When ambient RH exceeds 60%, the API powder's hygroscopic constituents absorb moisture, reducing flow and causing fill weight variance; pre-drying at 50°C for 2 hours in a tray dryer restores acceptable flow characteristics. Disintegration testing per USP <2040> confirms capsule rupture within 30 minutes. Finished product types are size 0, 1, and 2 capsules in HDPE bottle counts of 100 and 500.

Preparation of the oral solution presentation requires dissolving Nainiu Fanchu Powder Veterinary Grade API in purified water at a concentration of 5–15% w/v with the addition of sodium benzoate (0.1–0.2% w/v) and potassium sorbate (0.05–0.1% w/v) as antimicrobial preservatives, conforming to antimicrobial effectiveness testing per USP <51>. The solution pH is adjusted to 5.5–6.5 with citric acid monohydrate, and viscosity is confirmed below 100 cP at 25°C (Brookfield LV viscometer, spindle 1, 60 RPM) to ensure compatibility with automatic drenching equipment. The packaged product in 500 mL or 1 L amber PET bottles is stable for 18 months at 15–25°C when stored upright in the dark; inversion or horizontal storage during transport may result in particulate settling, and the product should be re-shaken before use. Compliance for the oral solution references USP <795> for nonsterile compounding, VICH GL18 for residual solvents, and Ph. Eur. 5.1.3 for pharmaceutical water quality. The downstream production process utilises a 500 L stainless steel mixing tank with bottom-mounted propeller agitator at 120 RPM, followed by 100 µm in-line straining before filling on a 2-head volumetric bottling line operating at 3000 bottles/hour. Finished product types are 500 mL and 1 L amber PET bottles with tamper-evident dosing cup closures.

    More Introduction

Nainiu Fanchu Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions is released as a free-flowing active pharmaceutical ingredient powder for multi-dose-form veterinary manufacturing. The model designation is Veterinary Grade API Powder, multi-dosage-form; the exact batch designation and specification version appear on the certificate of analysis. This material is differentiated from technical-grade, feed-grade, and agricultural premix powders by tighter controls on assay, related substances, residue on ignition, heavy metals, residual solvents, microbial bioburden, and, when the downstream dosage form is an injection, bacterial endotoxins. Non-pharmaceutical grades may be sold on visual appearance and a simple ingredient declaration, but they do not support reproducible tablet compression, sterile filtration, or parenteral safety because they are not tested against Ph. Eur., VICH, or manufacturing-authorization release limits. The veterinary-grade powder is intended solely for veterinary medicinal products and must not be used in food-producing animals unless the finished product carries a licensed withdrawal period. All statements in this technical introduction refer to the product as a starting material, not to a finished dosage form.

The principal difference from other powder products lies in the release specification and intended use. A feed premix ingredient may contain carriers, anticaking agents, and mineral diluents that are acceptable in feed but not in an injection or tablet; a technical-grade powder may have broad particle-size distribution and unknown bioburden. Nainiu Fanchu Powder Veterinary Grade API is released without unapproved carriers unless specifically declared on the label. The powder may be used as the active ingredient in all seven listed dosage forms, but the formulator must assign a dedicated grade to each manufacturing route: oral solid, oral liquid, premix, and parenteral grade are not automatically interchangeable. This is a critical difference from commodity powders, where the same lot may be marketed for multiple uses without route-specific testing.

Physical and Chemical Identity of the Veterinary-Grade API

Release testing combines identity, purity, water content, residue, particle size, and microbial safety. Where the product-specific monograph identifies a marker substance, assay is performed by high-performance liquid chromatography with UV detection; where no single chemical marker is specified, total active fraction may be quantified by a validated spectrophotometric or gravimetric method. The powder is also evaluated for polymorphic or extractive consistency because alterations during drying or milling can change dissolution and flow. Batch-to-batch variance is controlled by comparing the release data with the previous five batches; a shift in water content above 0.5 percentage points triggers review before acceptance.

The loss-on-drying limit is not a stability limit but a release criterion; residual moisture above 5.0% in this powder can increase the risk of granule capping and microbial growth. When the API is used in a parenteral formulation, the incoming endotoxin level must be confirmed on each batch because downstream sterile filtration does not remove endotoxin. Storage in sealed aluminium-laminated bags at ≤25 °C and ≤60% RH is required; repeated opening under high humidity can increase water content above the release limit within 48 h. The retest date on the CoA is based on long-term stability data; if no long-term data are available, the supplier should provide a conservative retest interval.

Tablet and capsule processing begins with particle-size adjustment. In a rotary tablet press running at 40–60 rpm, the granular intermediate should exhibit a bulk density of 0.45–0.65 g/mL and a Carr index below 20% to maintain weight variation within pharmacopoeial limits. Direct compression is generally limited to low-dose tablets with active content below 25 mg per unit because larger loads of the powder may reduce flow and compressibility. For higher-dose tablets, wet granulation with purified water or 2%–5% povidone K30 binder is used; fluid-bed drying at inlet air 55 °C–65 °C is maintained until the final granule moisture is below 2.0%. Lubrication with magnesium stearate at 0.5%–1.0% w/w is added for tablets but can delay disintegration if the blending time exceeds 5 min. Tablet hardness is commonly adjusted to 6–12 kp for veterinary bolus formulations, but the hardness target must be derived from the finished product’s disintegration and dissolution profile. Capsule filling on dosator-type machines is supported by tapped densities of 0.55–0.70 g/mL; when the powder’s angle of repose exceeds 40°, addition of colloidal silicon dioxide at 0.5%–1.0% w/w reduces interparticle friction. Dissolution of the finished tablet is tested according to Ph. Eur. 2.9.3, and friability is tested according to Ph. Eur. 2.9.7; a friability value above 1.0% indicates that tableting conditions require adjustment. Published data for this exact powder in production-scale tableting is limited, so the stated ranges should be verified with the finished product marketing authorization.

A direct comparison against non-pharmaceutical Nainiu Fanchu powder grades clarifies the significance of veterinary-grade controls. Technical or feed grades may be produced with lower sieving precision and without endotoxin testing, which creates a high rejection risk in sterile injection lines. The veterinary grade is the only form suitable for tablets, injections, capsules, and oral solutions under GMP because its certificate of analysis contains the release data needed for batch record review. Batch-to-batch variance in particle size has direct consequences for content uniformity.

What Limits the Use of a Powder API in Injections and Oral Solutions?

For injectable formulations, the powder must be dissolved in water for injection or a co-solvent vehicle and sterile-filtered through a 0.22 µm polyethersulfone or polyvinylidene fluoride membrane. Adsorptive losses to the filter can be significant when the active substance is hydrophobic; a filter validation study is therefore required for batches below 10 L. The filtered solution must meet sub-visible particle limits according to Ph. Eur. 2.9.19: for parenteral solutions, not more than 25 particles per mL at ≥10 µm and not more than 3 particles per mL at ≥25 µm. Injectable solutions must also pass sterility testing according to Ph. Eur. 2.6.1 and bacterial endotoxin testing according to Ph. Eur. 2.6.14. Endotoxin is the main reason a non-injectable powder cannot be converted into an injectable product by simple sterile filtration, because filtration removes bacteria but not endotoxins. The veterinary-grade API assigned for parenteral use is released with a bacterial endotoxin limit of <0.5 EU/mg, which allows calculation of the maximum endotoxin load per dose. If the active substance requires a pH below 3.0 for complete dissolution, the finished injection may cause injection-site pain and should be buffered or reformulated. For aqueous injections, the solution pH should be adjusted with dilute hydrochloric acid or sodium hydroxide only if the active substance is stable at the chosen pH; forced degradation at pH 1.0 and pH 13.0 is used to identify degradation products. The powder’s buffer capacity is low, so the finished formulation rather than the API determines final pH. The oral solution route is less stringent; however, the preservative efficacy test of Ph. Eur. 5.1.3 still applies to multi-use oral drench containers. Solubility in propylene glycol/water mixtures at 20%–30% v/v propylene glycol is often used to maintain a clear oral solution, but published data for this exact powder in high-concentration oral vehicles is limited and should be generated during formulation development.

When Premix and Granule Manufacturing Demands Low Dust Generation

For direct powder dosing, granules, and feed premixes, the principal processing risk is airborne dust and segregation. The veterinary-grade powder can be densified into granules by dry roller compaction at roll pressure 20–40 kN to lower dust and improve flow. In a double-ribbon blender operating at 40%–60% of geometric capacity, a 20% active premix is first produced with a carrier such as ground corncob, lactose monohydrate, or maltodextrin. Ten-point sampling after 8–12 min of mixing should yield a coefficient of variation for active content of below 5.0%. The premix is then diluted to the final feed concentration, typically in the range of 100 g to 2 kg per tonne depending on the veterinary approval; each dilution step requires a documented blend uniformity study because particle-size mismatch between the API and carrier can produce segregation during transfer. Granulation with starch paste at 5%–8% solids improves particle-size distribution and reduces dust, but drying above 70 °C may darken the granules and should be avoided unless stability data show no active loss. Premix stability in finished feed can be measured by accelerated storage at 40 °C/75% RH for 3 months; active recovery below 90% requires reformulation with a protective carrier. Granule friability below 1.0% after tumbling is desirable for automatic dosing systems. If the powder is used in a soluble premix for drinking water, the particle size should permit dispersion without clogging nipple drinkers; a D90 below 150 µm is usually sufficient, but re-agglomeration in humid environments above 60% RH must be controlled by sealed storage and desiccant. The working area airborne dust limit should follow the occupational exposure band assigned to the active substance; local exhaust ventilation is required during open weighing of the bulk powder.
