Application of Mahuang Zhike Pingchuan Oral Solution Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

The liquid extract corresponding to Mahuang Zhike Pingchuan Oral Solution exhibits pH-dependent solubility of ephedrine alkaloids in potable water matrices. In commercial poultry operations with nipple-drinker delivery infrastructure, the stock solution is metered through a proportional injector set to deliver 1.0 mL of extract per liter of drinking water when total alkaloid content is normalized to 1.0 mg/mL L-ephedrine equivalent. Metering accuracy must conform to the volumetric displacement tolerance specified in ISO 8655-5 for piston-operated pipettes or to manufacturer-listed injection pump repeatability of ±1.5% across a 4–8 bar line pressure range. Water hardness exceeding 250 mg/L CaCO₃ reduces extraction-medium clarity and may precipitate polysaccharide residues from the herbal matrix; chelation with EDTA at 0.05% w/v is required in such matrices before extract addition. Chlorinated supply water containing more than 2.0 mg/L free chlorine oxidizes the secondary amine groups on ephedrine and pseudoephedrine within 6 hours at ambient temperature (22–25°C), forming N-oxide derivatives with diminished bronchodilatory activity. Addition of sodium thiosulfate at 0.01% w/v in the stock solution neutralizes residual chlorine prior to line injection. The diluted medicated water remains stable for 24 hours at pH 4.0–5.5; beyond 48 hours, microbial proliferation in residual organic matter accelerates alkaloid degradation and necessitates system flushing with 0.1% peracetic acid solution per farm biosecurity standard operating procedures. Compatibility with acidified drinking water systems is restricted: the addition of organic acids below pH 3.8 causes protonation of the secondary amine functional group and reduces voluntary water intake in weanling pigs, a palatability effect documented in production farm records and consistent with the organoleptic profile of ephedrine hydrochloride. In nipple-drinker lines with biofilm accumulation, cross-contamination between medicated and unmedicated lines is managed by dedicated water circuits or by routing the stock solution through a positive-displacement dosing pump with a minimum turndown ratio of 1:1,000. Batch-to-batch variation in extract total solids content between 45% w/w and 65% w/w dictates a pre-dilution viscosity adjustment; extract viscosity above 200 mPa·s at 25°C per Ph. Eur. 2.2.10 requires the addition of 10–15% v/v propylene glycol to maintain consistent pump draw-down from the stock reservoir.

At What Compression Force Does Thermal Degradation of Ephedrine Alkaloids Occur During Veterinary Tablet Manufacture?

Because the native liquid contains 55–65% w/w moisture, direct compression is not feasible, and a co-processed excipient matrix consisting of microcrystalline cellulose PH102, lactose monohydrate, and crospovidone at a 6:3:1 ratio serves as the adsorption substrate for wet granulation. The extract is sprayed onto the blend at 25–30% w/w of total dry granulation mass in a top-spray fluid bed granulator fitted with a 0.8 mm two-fluid nozzle and operated at inlet air temperature 55–60°C. Exceeding 65°C inlet air temperature initiates volatilization of free ephedrine base, which has a reported vapor pressure threshold that becomes measurable above 60°C at atmospheric pressure; published data for this specific configuration is limited, but production batch records indicate an alkaloid assay loss of 4–7% when bed temperature exceeds 68°C during the 45-minute spray cycle. The dried granulate is milled through a 1.25 mm conical screen and blended with magnesium stearate at 0.75% w/w for 5 minutes in a bin blender rotating at 12 rpm. Compression force between 45 kN and 65 kN on a 16-station rotary tablet press yields cores with hardness 60–90 N, friability below 0.8% per Ph. Eur. 2.9.7, and disintegration time under 15 minutes per Ph. Eur. 2.9.1 in purified water at 37°C. Tablet weight variation is controlled per Ph. Eur. 2.9.5 with an acceptance value limit of 15, and content uniformity for ephedrine alkaloids is verified by HPLC with a relative standard deviation below 6.0% across 10 individually assayed units. The compression process generates localized die-wall temperatures between 35°C and 45°C under continuous production speeds of 45,000–65,000 tablets per hour; at these temperatures, ephedrine hydrochloride remains chemically stable, but prolonged residence time in the feed frame above 15 minutes permits moisture uptake in the granulate that increases tablet weight variation to 3.5% from the baseline 1.8%. Immediate-release cores coated with an HPMC-based film at 3% weight gain reduce moisture uptake from ambient air at 60% relative humidity by 80% over 72 hours, per dynamic vapor sorption comparison. Tooling specifications require D-tooling punches with a cup depth of 1.2 mm and embossment depth not exceeding 0.15 mm to prevent capping at compression forces above 60 kN. Compression of the granulate at relative humidity exceeding 60% RH without prior conditioning in a dehumidified suite leads to picking and sticking on punch faces, a failure mode observed on production equipment when the facility HVAC dew point drifts above 12°C.

Parenteral Compatibility Matrix and Terminal Sterilization Boundary for Ephedra Alkaloid Solutions

Prior to formulation, the crude extract undergoes cascade filtration through 0.45 μm depth media followed by 0.22 μm polyethersulfone membrane cartridges with an upstream-to-downstream membrane surface area ratio of 1:0.4. The filtrate is assayed for bacterial endotoxins per Ph. Eur. 2.6.14 and must register below 0.5 EU/mg of dry extract solids before release into the parenteral compounding suite. Terminal sterilization by autoclaving at 121°C for 15 minutes is contraindicated for ephedrine-containing parenteral products because the secondary amine undergoes Maillard-type condensation with residual reducing sugars from the herbal matrix, producing chromophoric adducts that elevate solution absorbance at 420 nm by 0.15–0.35 AU and reduce total alkaloid assay by 6–9% in accelerated degradation studies. Filtration-based aseptic processing at ambient temperature (18–25°C) with a 0.22 μm sterilizing-grade filter is the recognized route, provided the pre-filtration bioburden does not exceed 10 CFU/100 mL per 21 CFR 610.12 and the filter integrity test by bubble point method conforms to the membrane manufacturer's stated minimum pressure threshold. The parenteral vehicle comprises water for injection, sodium chloride for isotonicity adjustment to 280–320 mOsmol/kg per Ph. Eur. 2.2.35, and citrate buffer at 10 mM to maintain pH 4.5–5.0, at which range ephedrine hydrochloride exhibits maximum chemical stability as determined by forced-degradation kinetics. Antioxidant selection is restricted to sodium metabisulfite at 0.05% w/v; combination with EDTA disodium at 0.01% w/v chelates trace transition metals that otherwise catalyze oxidative deamination of the side-chain amine. Acceleration data at 40°C/75% RH over 6 months indicate total alkaloid retention of 97.3% in clear Type I borosilicate vials, whereas amber vials provide no additional photoprotection benefit beyond 6 months when stored per ICH Q1B light-exposure conditions at 1.2 million lux-hours and 200 W·h/m² near-ultraviolet energy. Particulate matter in the finished injectable is controlled per Ph. Eur. 2.9.19: no more than 25 particles per container at ≥10 μm and no more than 3 particles per container at ≥25 μm. Sterility assurance level of 10⁻⁶ is confirmed per Ph. Eur. 2.6.1 membrane filtration method using a sterility test volume of 20 mL per container from a minimum of 20 filled units. Freeze-thaw cycling between -20°C and 25°C for 3 cycles produces no visible precipitation when the formulation contains 5% v/v propylene glycol as a co-solvent; without the co-solvent, aqueous formulations form needle-like crystals of pseudoephedrine hydrochloride below 4°C, a physical instability that limits cold-chain transport without glycol addition.

When Capsule Fill Exceeds 620 mg and Tapped Density Falls Below 0.45 g/mL

Achieving reproducible fill weight in capsule production depends on conversion of the liquid extract into either a semi-solid thixotropic mass for liquid-filled hard capsules or a dry free-flowing granulate for powder-filled operations. For liquid-filled hard gelatin capsules, the extract is concentrated under vacuum at 40–45°C to a specific gravity of 1.18–1.25 g/mL, then homogenized with PEG 400 and glycerol at a 7:2:1 ratio to achieve a fill viscosity of 800–1,200 mPa·s at 40°C measurable per Ph. Eur. 2.2.10 rotational viscometry. Capsule sealing by LEMS banding or hypromellose spray sealing prevents cross-contamination and moisture ingress; shell brittleness at relative humidity below 35% dictates storage in desiccated bins conditioned to 45% RH. The fill weight target of 480 mg per size 0 capsule requires bulk density of 0.52–0.60 g/mL and Carr's index not exceeding 18% per Ph. Eur. 2.9.34; powder bed collapse during transfer from the hopper to the dosing disc is minimized by maintaining the powder in the hopper at a fill level between 30% and 70% of total capacity, as both under-filling and over-filling alter the aerated-to-tapped density ratio and introduce fill weight drift beyond ±5% of target. Fill weight uniformity is verified per Ph. Eur. 2.9.5 with an acceptance value of 15, and the mass variation at 60 capsules per hour sampling intervals is monitored by in-process check-weighing on a 1 mg resolution balance with automatic feedback to the dosing disc pressure setting. The powder-filled formulation contains colloidal silicon dioxide at 20–30% w/w of extract dry solids as a moisture-scavenging carrier; without this carrier, the granulate absorbs atmospheric moisture within 30 minutes at 55% RH and forms soft agglomerates that obstruct the tamping pin assembly, a process interruption documented on intermittent-motion capsule fillers operating at 45,000 capsules per hour. Desiccant pouches containing 1 g silica gel per 60 capsules maintain internal headspace moisture below 3.0% w/w after 24 months at 25°C/60% RH in HDPE bottles with tamper-evident induction seals. For gelatin shells, dissolution testing per Ph. Eur. 2.9.3 in 900 mL of 0.1 M hydrochloric acid at 37°C with paddle apparatus at 50 rpm requires not less than 80% of the labelled ephedrine content released within 30 minutes; hypromellose shells show equivalent release but require an additional preconditioning step in pH 1.2 buffer without enzyme for 15 minutes to prevent shell surface gelation from retarding early drug diffusion.

The incorporation of the liquid extract into dry feed powder systems across feed mills processing 10–50 tonnes per hour requires adsorption onto a hydrophilic carrier to prevent caking and segregation in bulk silos. The extract is first diluted 1:1 v/w with propylene glycol to reduce surface tension to below 45 mN/m, then sprayed onto pre-screened wheat bran or defatted rice bran carrier at a ratio of 1 part extract to 4 parts carrier in a twin-shaft paddle mixer operating at 60 rpm for 12 minutes. The resultant adsorbed intermediate comprises 18–22% moisture and must be dried in a vacuum tray dryer at 45°C and 25 kPa absolute pressure until final moisture reaches 6–8% w/w, determined by loss-on-drying per AOAC 930.15. Drying above 50°C in forced-air convection causes surface crusting and internal moisture entrapment, yielding a biphasic particle with friable exterior and sticky core that resists uniform blending and produces stratified layers in the finished feed. The dried intermediate is passed through a 500 μm sieve and blended into complete feed matrices at a finished dosage of 0.5–1.0 kg of extract equivalent per tonne of feed. Blend uniformity testing per FDA CVM guidance for medicated feed mixing requires a coefficient of variation below 5.0% across 10 sampling points taken at 5-minute intervals during continuous mixer discharge; mixers with a paddle-to-wall clearance exceeding 25 mm exhibit residual pockets of unmixed active premix near the dead zones at the discharge end, a failure mode resolved by installation of a 10 mm clearance paddle set. Near-infrared spectroscopy at 1,450 nm and 1,940 nm bands provides inline moisture and alkaloid content verification during the final blend stage, with a calibration root mean square error of prediction below 0.8% w/w for moisture and below 3.5% relative for ephedrine content. Segregation potential is minimized by maintaining the finished feed at a particle size distribution where D₁₀ ≥ 200 μm and D₉₀ ≤ 1,800 μm, verified by laser diffraction per ISO 13320:2020. Contamination carryover between batches is limited to 0.5% of the active concentration per current veterinary GMP expectations for medicated feed lines, achievable by a 5-minute flushing sequence with untreated carrier at 100% mixer capacity and verified by assaying the flush material for ephedrine residue below 10 μg/g.

Granulation Endpoint Parameters Measured on a 300-L High-Shear Mixer for Ephedra Extract Dispersions

Agglomeration of the liquid extract into free-flowing oral granules requires the addition of carbohydrate binders to produce dispersible particles for administration in swine and calves. The extract is pre-mixed with maltodextrin DE 10–12 and sucrose at a 5:3:2 ratio to yield a paste with moisture content 28–32% w/w. Granulation proceeds in a 300-L high-shear mixer fitted with a chopper blade and main impeller; impeller speed 180 rpm and chopper speed 1,500 rpm for the first 4 minutes, followed by 6 minutes at 220 rpm impeller and 2,000 rpm chopper, produce an endpoint granulate with mean particle diameter between 400 μm and 800 μm. Endpoint is determined by torque rheometry: a plateau in impeller torque between 18 N·m and 22 N·m corresponds to granulate moisture of 12–14% and indicates that liquid bridges have fully formed without over-wetting. Over-wetting beyond torque 24 N·m produces a paste-like mass that coats the mixer bowl and requires manual scraping, extending batch discharge time by 40 minutes and producing granulate with unacceptable lumps exceeding 2.5 mm in diameter. The wet mass is discharged through a 4 mm square-mesh screen and dried in a fluid bed dryer at inlet air temperature 50–55°C to final moisture 2.5–3.5% w/w; the drying endpoint is confirmed when outlet air temperature reaches 42°C and exhaust relative humidity falls below 15%. Granule hardness measured by a Pharmatron ST-50 stylus at 5 N probe force yields values between 1.5 N and 3.0 N, sufficiently robust for packaging yet friable enough for rapid dissolution in 38–40°C water within 90 seconds per dissolution testing adapted from Ph. Eur. 2.9.3 with a basket apparatus. The finished granules are filled into 250 g, 500 g, and 1 kg HDPE jars with 0.5 g silica gel canisters and sealed with aluminum-laminate induction liners. Storage at 30°C/65% RH for 24 months produces alkaloid assay retention of 98.5% relative to initial per ChP 2020 HPLC methodology with a C18 column, 0.1% phosphoric acid-acetonitrile mobile phase, and UV detection at 210 nm. Migration of moisture through the HDPE jar wall at 40°C/75% RH conditions produces granulate moisture increase of 0.8–1.2% after 6 months, which remains within specification but accelerates sucrose crystallization and increases granule hardness beyond 4.0 N, extending dissolution time to 150 seconds. Sieve analysis per ISO 3310-1 test sieves confirms that not more than 10% of granules pass through a 180 μm sieve, thereby limiting dust generation during farm-level scoop transfer.

Within a premix manufacturing line, the highest dilution ratios demand rigorous stepwise blending to achieve content uniformity in concentrated intermediate products intended for terminal feed mixing. The extract is adsorbed onto colloidal silicon dioxide at 10% w/w of the extract mass; the silica matrix immobilizes the aqueous component and produces a free-flowing powder with angle of repose not exceeding 35° per USP <1174>. The silica-adsorbed extract is then diluted in geometric progression: 1 part active premix to 9 parts ground limestone carrier with particle size between 200 mesh and 325 mesh in the first dilution stage, followed by 1 part of the 10% premix to 19 parts corn cob meal carrier in the second stage. Each dilution stage employs a double-cone blender running at 15 rpm for 20 minutes, validated by sampling 12 points per USP <905> uniformity requirements for premixes with acceptance value not exceeding 15. The final premix contains 1–2% extract equivalent and is intended for incorporation at 10–20 kg per tonne of finished feed. Critical process parameters include carrier particle size uniformity with D₅₀ between 150 μm and 300 μm, carrier surface oil content below 1.5% w/w, and ambient relative humidity below 55% during blending to prevent electrostatic attraction of fine silica particles to blender walls and subsequent demixing at the discharge valve. Static buildup on the blender interior exceeds 5 kV/m when relative humidity falls below 35%, producing adherence of up to 3% of the batch mass to the vessel wall and requiring ionization bar installation at the discharge port. The finished premix is packaged in multi-wall paper bags with an inner 0.1 mm linear low-density polyethylene liner, and labelled with a 9-month expiration date when stored below 30°C and 60% RH. Assay of the premix per FDA 21 CFR 225.158 sampling protocols requires ephedrine content within 90–110% of the declared label claim, with lot-to-lot relative standard deviation below 4.0% across 10 consecutive production batches. Carryover cross-contamination into subsequent non-medicated premix batches is limited to 0.25% of the active level, verified by flushing the blender with 5 kg of untreated corn cob meal for 10 minutes at 20 rpm and assaying the flush material for ephedrine residue below 5 μg/g by LC-MS/MS with a limit of detection of 0.1 μg/g.

Repacking the Concentrated Extract into Metered Farm-Delivery Systems

Field repacking operations for cattle and swine herds involve calibrated dosing systems integrated into drinking water lines or milk replacer circuits. The extract as supplied typically contains 5–10% residual propylene glycol or ethanol from the manufacturing extraction process; field dilution into aqueous carrier systems at rates of 1:500 to 1:2,000 v/v yields final alkaloid concentrations of 0.5–2.0 μg/mL in drinking water. Automated proportioner systems require operating water pressure between 0.3 bar and 6.0 bar, with injection rates adjustable from 0.2% to 5.0% v/v. The extract is drawn from 20-L or 200-L HDPE drums through a pre-filter screen retaining particulate matter above 75 μm before entering the proportioner piston chamber. Compatibility testing per ISO 13485 material interaction protocols confirms that EPDM rubber seals in the dosing pump maintain swelling below 5% after 72 hours of continuous contact with the extract containing 5% propylene glycol; silicone rubber seals show superior chemical compatibility with swelling below 2% but require higher replacement frequency due to mechanical wear. The line residual after dosing is flushed with clean water for 15 minutes at 2.0 bar to prevent biofilm accumulation in low-flow zones; this flush volume corresponds to 3 line volumes in typical barn installations with 50 mm internal diameter piping over 120 m runs. Stability of diluted medicated water is governed by water temperature: at 10°C, 95% alkaloid retention is achieved after 48 hours; at 30°C, retention falls to 88% after the same interval due to accelerated oxidative deamination of the β-hydroxyl amine side chain. Photodegradation in transparent header tanks exposed to direct sunlight accounts for an additional 3–5% ephedrine loss per 8-hour light period at 40,000 lux, and covered tanks reduce this loss to below 1%. The extract must not be combined in the same stock solution with alkaline veterinary products such as sodium bicarbonate oral electrolytes; admixture at pH > 8.0 converts ephedrine hydrochloride to the free base form, which exhibits reduced aqueous solubility below 50 mg/mL and partitions into the headspace of the stock container, leading to irreversible vapor-phase loss through vented closures. Sequential administration with ionophore coccidiostats in poultry requires a 24-hour withdrawal window between medicated water cycles to prevent feed-water cross-contamination in automatic recirculation systems that return unused water to the central reservoir.

    More Introduction

Supplied as a clarified multi-component botanical concentrate designated MZKPC-VET-API-04, the veterinary-grade Mahuang Zhike Pingchuan oral-solution active pharmaceutical ingredient is standardized for two marker groups: total ephedrine-type alkaloids and amygdalin. Despite the originating oral-solution nomenclature, this material is not a finished oral solution and is not directly injectable in its shipping form. It functions as a blending intermediate for licensed veterinary manufacturers producing tablets, injections, capsules, powders, granules, premix, and liquid solutions. The production stream uses a counter-current extraction train at 60 ± 2 °C in 316L stainless steel vessels, followed by 0.45 µm cellulose acetate clarification and low-temperature vacuum concentration at 45–55 °C under −0.08 to −0.06 MPa. This thermal profile is lower than conventional atmospheric concentration and is specified to preserve amygdalin, which degrades rapidly in neutral or alkaline aqueous media above 70 °C. Batch-to-batch variation is controlled by bulking three separate extraction runs into a single homogenized lot, with the final lot volume limited to 500 L. Suffix designations -T, -I, -C, -P, -G, -PM, and -S correspond to validated downstream use in tablet granulation, injectable processing, capsule powders, dry powders, granules, premix carriers, and oral solutions respectively. A base non-sterile grade may be used for solid and oral liquid forms, but the -I grade cannot be replaced by a base grade when injectable manufacture is intended.

What release limits define the veterinary API grade?

The product is released against a two-tier specification. Non-sterile solid and oral liquid grades comply with standard microbial limits, while the injection-grade suffix -I is subjected to additional endotoxin and sub-visible particle controls. Alkaloid content is expressed as ephedrine hydrochloride equivalents using HPLC-UV at 207 nm; amygdalin is quantified by HPLC-DAD at 254 nm. The total alkaloid ratio between ephedrine and pseudoephedrine shall remain between 1:0.35 and 1:0.55. A narrow ratio is required because synthetic alkaloid spiking or substitution with ephedra herb of different geographical origin alters the ratio and changes bronchodilatory kinetics. The expanded release profile is given in Table 1.

Each lot is sampled from the top, middle, and bottom of the bulking vessel through sterile 316L sample ports. Homogeneity is confirmed by assay variance below 2.0% RSD across sampling points. If the ratio of ephedrine to pseudoephedrine falls outside 1:0.35–1:0.55, the lot is quarantined and not released for injection or premix use. Because no international harmonized monograph exists for this specific multi-component veterinary extract, the listed release limits derive from Chinese Veterinary Pharmacopoeia general chapters and are tightened for injectable processing routes.

Processing compatibility from sterile injectable lines to low-moisture premix carriers

The liquid API can be introduced at different unit operations, but the critical processing window depends on the dosage form. For tablet manufacture, the liquid is substituted for the granulating solvent in high-shear or fluid-bed granulation. The recommended addition rate is 80–150 g/kg of dry blend, supplemented with purified water to achieve a final granulation moisture of 28–34% w/w. High-shear granulator trials on a 65 L bowl with impeller speed 180–220 rpm and chopper speed 1,500 rpm produced granules with mean particle size 180–320 µm and compressibility suitable for 8–12 kN compression force on 12 mm round tooling. Drying should be carried out at inlet air 55 °C to 65 °C; bed temperature above 70 °C causes surface darkening and loss of amygdalin. For hard gelatin capsules, the liquid is preferably sprayed onto microcrystalline cellulose or mannitol rather than used directly as a fill fluid, because its water activity of 0.78–0.86 can soften gelatin shells at 75% RH. When filled into vegetable capsules, a 4-week stress storage at 40 °C/75% RH is used to check crosslinking and brittleness. For powder and granule formulations, the API is sprayed in a top-spray fluid-bed processor at liquid feed rate 12–18 g/min, atomizing air pressure 1.5–2.5 bar, and product temperature 30–35 °C. Carrier selection is critical; maltodextrin DE 10–15 mixed with colloidal silicon dioxide at 0.5–1.0% w/w provides better adsorption than calcium carbonate alone. For premix carriers, the liquid is adsorbed onto corn cob meal or rice hulls in a horizontal ribbon mixer operating at 20–30 rpm. Total liquid addition should not exceed 100 g/kg of carrier unless the carrier is subsequently dried; above this level, caking and bridging occur in vertical storage bins. For oral liquid manufacturing, the API is added to purified water at 25 ± 5 °C under low-shear agitation. Multi-dose solutions should be preserved with potassium sorbate at 0.10–0.20% w/v or sodium benzoate 0.10–0.15% w/v; benzoate salts can reduce pH by 0.2–0.4 units and should be added after pH adjustment. Injectable formulations require a dedicated low-endotoxin grade. The liquid must be diluted to final concentration, adjusted to pH 5.0–6.5, and passed through a 0.22 µm PVDF membrane. Terminal sterilization at 121 °C for 15 min is not validated for this multi-component botanical API because amygdalin degrades under prolonged heat; therefore aseptic filtration is the only validated sterilization method. Pre-filtration bioburden must be ≤10 CFU/100 mL, and filling must occur in an ISO 14644-1 Class 5 zone with Grade A air supply.

When the API replaces dry extracts in premix and granule manufacture

A comparison with spray-dried extract powder and synthetic alkaloid blend is made under production conditions. Spray-dried extracts require re-dissolution before wet granulation, and typical spray-drying inlet temperatures of 180–220 °C cause a 5–12% reduction in total alkaloids relative to the liquid concentrate, depending on the atomizer type. Synthetic ephedrine hydrochloride blends, in contrast, lack amygdalin and polyphenolic co-solutes, producing faster dissolution but different respiratory-product response; they are not interchangeable for compounded veterinary preparations where the Ma Huang–bitter apricot seed ratio determines expectorant action. The liquid API also carries no insoluble herb powder residue, which reduces die-wall wear on rotary tablet presses compared with raw herb powder formulations. The comparative data are summarized in Table 2.

Storage stability has been assessed in high-density polyethylene drums with foil-sealed caps under nitrogen headspace at 2–8 °C for 24 months; assay loss at 24 months was 3.4% for total alkaloids and 6.1% for amygdalin. The liquid should not be frozen; freezing causes precipitation of polysaccharides and may shift the alkaloid ratio after thawing. The extract is incompatible with strong alkalis, concentrated oxidizing agents, and polyvalent metal salts such as ferric chloride, which can form tannin–metal complexes. Because the extract contains ephedrine-type alkaloids, every shipment must include a certificate of analysis stating the total alkaloid content and a regulatory declaration for the destination jurisdiction. Published data for specific formulation platforms such as long-acting injectable suspensions using this exact multi-component extract are limited; veterinary product developers should commission platform-specific stability studies before scale-up. Batch-to-batch variance is monitored with a statistical process control chart on total alkaloids and pH. A lot is released only when all parameters in Table 1 fall within the specified ranges and when the electronic batch record shows no deviation from validated extraction and filtration procedures.
