Compaction of the Liwei Powder veterinary API into tablets is not governed solely by active content. The first measurable bottleneck is the flow function coefficient of the blended matrix, because the API is typically finer than 100 µm and may carry a residual moisture content that shifts with storage humidity. A direct compression route is possible only when the API fraction is low enough to be carried by a free-flowing excipient such as spray-dried lactose monohydrate or microcrystalline cellulose; if the compressibility index determined under USP <616> exceeds 25%, the powder is likely cohesive and wet granulation should be selected instead. Wet granulation with povidone K30 at 3–10% w/w in purified water is a standard starting platform, but the endpoint must be controlled by impeller torque or power draw on the high-shear mixer rather than by visual appearance. After drying, loss on drying should be maintained between 1.5% and 2.5% for most non-hygroscopic formulations; over-drying below 1.0% increases granule friability and generates fines that cause weight variation at the tablet press. The dried granule is milled through a 1.0 mm conical screen, and oversized material is recycled only after hardness testing confirms that the granules are not overwetted. Lubrication with magnesium stearate between 0.25% and 1.0% w/w is usually sufficient for veterinary tablet compression; levels above 1.5% w/w should be avoided because the hydrophobic film suppresses tablet tensile strength and slows disintegration. Tablet weight and hardness are not universal endpoints; a target hardness of 40–80 N for a standard concave veterinary tablet is a reasonable release specification only when it is correlated with friability below 1.0% under USP <1216> and disintegration under Ph. Eur. 2.9.1. Uniformity of dosage units must meet USP <905> with an acceptance value of ≤15.0 in the first stage; process validation should also record compaction force, precompression force, and press speed from an instrumented rotary press. Because the exact particle-size and polymorph specification of the Liwei Powder veterinary API is product-specific, the acceptable compaction force range cannot be stated independently of the applicant’s pharmaceutical development report, but a compaction simulator study should bracket the proposed commercial press speed.
The injectable route eliminates much of the excipient-loading freedom available in oral solid dosage forms because the Liwei Powder veterinary API must first dissolve completely in an aqueous or non-aqueous vehicle and then be sterilized without unacceptable potency loss. Staged dissolution in Water for Injection at 20–25 °C is preferable; if the API has pH-dependent solubility, adjustment with dilute hydrochloric acid or sodium hydroxide should be completed before final volume adjustment to avoid precipitation at the filter or during terminal sterilization. The bulk solution should be prefiltered through a 0.45 µm membrane to reduce bioburden and then sterile filtered through a 0.22 µm membrane that has been validated for bacterial retention under ASTM F838-20. Filter integrity testing by bubble point or diffusive flow must be performed before and after filtration, and the minimum bubble point must match the membrane manufacturer’s validated specification for that lot. Terminal sterilization at 121 °C for 15 min is the default preference if the API can withstand a validated F0 value above 8 min without exceeding degradation thresholds; otherwise aseptic processing in an ISO 5 environment under ISO 14644-1:2015 is mandatory. Endotoxin testing under USP <85> or Ph. Eur. 2.6.14 is not optional, because the API powder itself may introduce pyrogenic material that filtration cannot remove. Particulate matter in the finished injectable must meet USP <788> limits, and hold-time validation should include bioburden counts at 0 h, 8 h, and 24 h after bulk solution preparation to demonstrate that the filtered solution does not support microbial proliferation before filling. Container closure integrity testing under USP <1207> should be included for vials sealed with elastomeric closures, especially when the formulation contains a non-aqueous cosolvent that can interact with rubber stoppers.
Low-fill-weight capsule operations shift the control burden away from powder flow and toward statistical distribution of a high-potency API across hundreds of thousands of dose units. If the labeled API content is below 1 mg per capsule, direct blending of the Liwei Powder veterinary API with the full excipient mass will often fail to meet uniformity requirements because the fine API adheres to blender surfaces and electrostatic charge drives segregation. The API should first be preblended with lactose monohydrate or mannitol at a ratio of at least 1:10 before high-shear mixing; stepwise geometric dilution reduces the chance that an undispersed API cluster remains in the final blend. Blend uniformity testing should follow USP <905> or Ph. Eur. 2.9.40, and the acceptance value of the first 10 dosage units should be ≤15.0. For process validation, sampling at 10 or more locations with a blend RSD ≤5.0% is a stricter but practical internal control for high-potency veterinary capsules. Capsule fill weight variation must comply with Ph. Eur. 2.9.5; for an average powder fill of 200 mg, the single-capsule limit is ±10% because the average mass falls between 80 mg and 250 mg. Empty capsule shell moisture of 13–16% can transfer to a hygroscopic API and cause sticking, brittle shells, or potency loss in the filled product; the filling suite should therefore be maintained at 35–45% RH and 20–25 °C. Static charge on polished steel contact parts is reduced by proper grounding and controlled humidity; if magnesium stearate would unacceptably slow dissolution, up to 5% sodium stearyl fumarate may be evaluated as an alternative lubricant, but its compatibility with the Liwei Powder veterinary API must be confirmed in a forced-degradation study rather than assumed from tablet experience. Low-fill-weight capsules often require a dosator nozzle with vacuum hold to prevent powder spillage during transfer, and the empty capsule body should be checked for electrostatic retention after the powder bed remains static for more than 8 h.
Granulation for oral powders and granules intended for in-feed or drinking-water reconstitution introduces a different excipient set than the tablet line because the objective is not a hard compact but a free-flowing, low-dust granule that can be dispersed rapidly in warm water. A fluid-bed granulator with a 0.8–1.2 mm top-spray nozzle and inlet air temperature of 60–70 °C typically produces granules with a geometric mean diameter between 150 µm and 500 µm; fines below 45 µm should be kept under 10% w/w to limit dust generation during sachet filling. Moisture content after drying should be below 2.0% for moisture-sensitive APIs, and desiccant may be required in the primary package if the product is stored above 60% RH. Homogeneity in multi-dose containers is tested by USP <905> or Ph. Eur. 2.9.5, but the sampling plan for a 500 g multi-dose jar should include top, middle, and bottom probe samples because segregation can occur even when the sachet content uniformity passes. Dust containment is measured by industrial hygiene sampling during bulk transfer and sachet filling; a downflow booth or isolator with a confirmed inward air velocity is required if the API has an occupational exposure limit below 10 µg/m³. The finished granule should be challenged by a pourability test and a dissolution test in water at 25 °C and 5 °C, because the product may be added to cold drinking water in field conditions.
Medicated premix manufacturing treats the Liwei Powder veterinary API not as a finished dose form but as an intermediate that must survive dilution into feed at ratios typically between 0.5 kg and 10 kg per tonne. The carrier choice must minimize bulk density mismatch and electrostatic adhesion; calcium carbonate, rice hulls, and wheat middlings have different particle size distributions and oil absorption capacities, and a carrier with extreme fines will increase dust rather than improve blend homogeneity. Mixing sequence should add the API to a small carrier fraction first, screen the concentrate through a 1.0 mm sieve, and then add the remaining carrier in two or three segments to reduce dead zones in the ribbon mixer or double-cone blender. Homogeneity is evaluated under 21 CFR 558.5 or EU Regulation 2019/4; sampling must include at least 10 locations across the mixer, and the assay variability should be reported as RSD rather than as a single mean value. Carryover testing must simulate a worst-case subsequent batch of non-medicated feed, because the flush step may not remove API trapped in elevator boots, mixer discharge gates, or dust collection returns. The carryover limit for highly active veterinary APIs is often set at 1% of the lowest approved inclusion rate, but the exact acceptance criterion is regulatory-file-dependent and cannot be assumed from a single washout study. If the API has a narrow therapeutic margin in target species, a dedicated premix line or a validated sequencing protocol with ground corn flush and assay verification is required.

Water-soluble oral solutions for drinking water administration require the Liwei Powder veterinary API to remain in true solution at the lowest expected tap-water temperature, typically 5–10 °C in winter barns. The formulation pH should be selected from a solubility curve that covers at least pH 3.5–6.5, and a buffer system such as citrate or phosphate should be used only after confirming that the buffer salt does not reduce solubility through a common-ion effect. Dissolved oxygen can degrade oxidizable APIs after dilution in field water systems; nitrogen sparging of the bulk solution to ≤0.5 mg/L dissolved oxygen before filling is a useful control if oxidative degradation is confirmed by forced-degradation data. Preservatives such as sodium benzoate at 0.1–0.2% w/v and potassium sorbate at 0.05–0.1% w/v may be added for multi-dose containers opened repeatedly, but preservatives are not a substitute for microbial limits testing under Ph. Eur. 5.1.4 or USP <1111>. Compatibility with automatic proportioners requires a concentrate that does not precipitate when diluted at 1:50 or 1:100; a precipitation zone can appear at intermediate pH even when the API is soluble at both the concentrate and final dilution pH. Photostability testing under ICH Q1B should determine whether amber polyethylene terephthalate bottles are required; a loss of more than 5% potency after exposure to 1.2 million lux-hours and 200 Wh/m² UV light is a useful decision threshold for opaque packaging.

Liwei Powder Veterinary Grade API, model LW-VG-API-P, is released as a crystalline powder for veterinary medicinal product manufacture. The same lot-controlled API can be allocated to tablets, injections, capsules, powders, granules, premixes, and solutions without changing the primary stability data package. Release acceptance criteria include assay by HPLC 98.0–102.0% on dried basis, loss on drying ≤0.5%, residue on ignition ≤0.1%, total related substances ≤1.0%, heavy metals ≤10 ppm, and residual solvents within VICH GL18 Option 1 limits. Particle size is measured by laser diffraction according to ISO 13320-1:2020; the standard powder is released with D90 ≤150 µm, while parenteral-grade material is micronized to D90 ≤10 µm. The product is supplied in 25 kg polyethylene-lined fiber drums with lot-specific certificates of analysis and stability statements. The material is not intended for human use and is not sterilized at the API stage.
For each batch, analytical release includes identity by Fourier-transform infrared spectroscopy and X-ray powder diffraction to confirm polymorph consistency. Assay and related substances are determined by HPLC-UV using a 250 mm × 4.6 mm, 5 µm C18 column with a phosphate buffer/acetonitrile gradient. Residual solvents are determined by headspace gas chromatography with flame ionization detection; water content is determined by Karl Fischer titration according to Ph. Eur. 2.5.12. Particle size distribution is measured by laser diffraction using a wet or dry dispersion unit; for the standard grade, the dry dispersion pressure is set at 1.0–2.0 bar to avoid particle attrition during measurement. Elemental impurities are determined by inductively coupled plasma mass spectrometry and controlled according to USP <232>/<233>. The certificate of analysis therefore includes the same data set expected for a pharmaceutical-grade substance, not a feed additive.
In veterinary supply chains, commodity feed-grade powders may contain the same nominal chemical entity but are not manufactured under a pharmaceutical quality system. Their particle size distribution is often broad, with span values above 3.0, which increases segregation during transfer and reduces uniformity in direct compression and premix dilution. The Liwei standard grade is released with span 1.8–2.2 and a controlled fines fraction below 75 µm of 20–35%. Batch documentation includes residual solvent concentrations for Class 2 solvents according to VICH GL18, elemental impurities according to USP <232>/<233>, and a defined polymorph identity by X-ray powder diffraction. Technical-grade material may not include these data. In addition, the product carries a bioburden limit of ≤100 CFU/g for oral dosage forms and ≤10 CFU/g for parenteral grade; feed-grade powder is not routinely tested for bioburden. These differences reduce the technical transfer burden when a veterinary manufacturer must qualify a single API source across multiple dosage forms.
Before lot release, powder flow is evaluated using a tapped density analyzer and a laser diffraction instrument. For direct compression, the acceptance window is bulk density 0.45–0.65 g/cm³, tapped density 0.60–0.85 g/cm³, Carr index 20–30, and Hausner ratio 1.2–1.4. When the Carr index approaches 30, wet granulation in a 50 L high-shear granulator is used to densify the material; granule D50 150–250 µm after extrusion through a 1.0 mm screen is treated as the target. For capsule filling on a 100,000 capsule/hour continuous machine, plug formation requires a stable density under compression; fines below 75 µm are held between 20% and 35% of the distribution to prevent both segregation and poor flow. If the fines fraction exceeds 35%, the powder may require dry granulation or addition of a glidant.
For premix and oral powder applications, the product is sieved through an 850 µm screen and mixed geometrically with feed carrier. A stratified sampling plan with 10 sampling points is recommended for blend homogeneity; active recovery at each point should be 90–110% of label claim. Direct addition of micronized parenteral grade to dry premix is not recommended because electrostatic agglomeration can produce local high-potency pockets. When solution dosage forms are prepared, particle size influences dissolution rate but not the final chemical assay; if the formulation requires rapid reconstitution, the micronized grade with wetting agent is used.
Thermal processing is not assigned above 80 °C unless forced degradation data support it. Hot-melt granulation on a twin-screw extruder with 40:1 L/D ratio requires barrel temperatures below the API melting onset; if the powder is processed at 60–80 °C, residence time is limited to 2–5 min to prevent polymorph conversion. For aqueous wet granulation, water addition is controlled by torque rather than time; the granulation endpoint is reached when impeller torque increases by 15–25% from dry mix baseline.

During parenteral development, controls are imposed that are not applicable to dry premix. The injection grade is released with bioburden ≤10 CFU/g by membrane filtration according to Ph. Eur. 2.6.12, bacterial endotoxins <0.25 EU/mg by Ph. Eur. 2.6.14, and subvisible particulate matter after reconstitution according to Ph. Eur. 2.9.19. The API is not sterile and not depyrogenated; the drug product must be terminally sterilized or aseptically filtered. The dry premix route does not require endotoxin control at that stringency, but using the same injection-grade lot in a premix can be wasteful and technically inappropriate because micronized material may agglomerate in low-shear ribbon or drum blenders. If transfer of the same lot across both routes is required, a terminal deagglomeration step through a 500 µm screen is used before premix blending.
For injectable solution manufacture, water for injection is used and the pH is selected from pH-solubility and pH-stability profiles. Holding times at room temperature should not exceed 8 h unless chemical stability data support longer periods. Contact surfaces should be 316L stainless steel with electropolished finish; the use of highly alkaline buffers is avoided unless specific pH-stability data are available. For dry premix, moisture uptake above 70% RH causes flow decline and possible clumping; storage of opened drums should therefore be limited to 48 h at ≤60% RH.
On a 16-station rotary tablet press with compression force 8–14 kN, direct compression formulations containing 20–35% w/w Liwei powder and microcrystalline cellulose with croscarmellose sodium are targeted to produce compact tensile strength ≥1.5 MPa and friability ≤0.5% according to Ph. Eur. 2.9.7. Published data for this specific formulation is limited; the 1.5 MPa value is therefore applied as an internal development gate rather than a compendial requirement. Encapsulation on a 100,000 capsule/hour machine requires Carr index 20–25; if Hausner ratio exceeds 1.4, the powder is dry granulated by roller compaction before encapsulation. In wet granulation, water addition of 5–10% w/w is typical, and drying at 40–50 °C to a granule moisture content ≤1.5% is used to control sticking and tablet picking.
Blend consistency on production-scale 600 L bin blenders is monitored by stratified sampling. The particle size span is a more sensitive indicator of segregation risk than average particle size alone; a shift in span from 1.8 to 2.5 can indicate milling screen wear. For this reason, the span value is trended on control charts with a rejection limit >2.2 for standard grade. In direct compression, tableting speed is kept below the point where weight variability exceeds 2.0% relative standard deviation.

Stability studies are conducted according to VICH GL3. The product is stored in the original sealed drums at 15–25 °C, protected from light and moisture. Long-term conditions 25 °C/60% RH and accelerated conditions 40 °C/75% RH are used for data generation; the retest period is assigned from real-time data. When relative humidity exceeds 60%, pre-drying at 40–50 °C for 2–4 h is required before direct compression. Moisture sorption is low below 60% RH but increases above 70% RH; drums should be closed immediately after sampling. The packaging configuration is 25 kg net weight in a double polyethylene liner inside a fiber drum with desiccant and tamper-evident seal.
Batch-to-batch consistency is controlled by releasing every lot against full specifications rather than skip-lot testing. The particle size span is a process consistency marker; shifts above 2.2 indicate milling screen wear or change in feed rate and trigger corrective action before release. For injectable grade, micronization performance is monitored by D50 3–5 µm and span ≤2.0. Residual solvent and elemental impurity results are lot-specific and included in the certificate of analysis. These data support multi-source qualification in veterinary manufacturing sites that require a single API vendor for oral, parenteral, and feed-route products.
In solution formulation, pre-screening for solubility across pH 1.2, 4.5, 6.8, and 7.4 is used to select the vehicle. If solubility is below 1 mg/mL, the formulation is adjusted with pH control or a solubilizer only after compatibility testing. The powder is pre-wetted before addition to the aqueous phase to reduce floating and lumping. For injectable solutions, the final filtration train includes a 0.45 µm clarifying filter and a 0.22 µm sterilizing filter; filter integrity is tested according to ASTM F838-20. The final solution is not released unless filter integrity passes and pH, assay, and particulate matter are within specification. Avoid contact with strong oxidizing agents and rubber stoppers that leach zinc; metal ions can accelerate degradation if the molecule is oxidation-sensitive.
