Lyophilized liposome veterinary API intended for injectable dosage forms is supplied as a sterile cake with residual moisture <3.0% and is reconstituted for aseptic downstream processing. In companion-animal medicine, the two largest injectable applications are liposomal amphotericin B for canine leishmaniasis and liposomal doxorubicin for canine lymphoma. The liposomal bilayer in these systems is typically hydrogenated soy phosphatidylcholine–cholesterol–DSPE-PEG 2000 at a 55:40:5 molar ratio, with a drug-to-lipid molar ratio between 0.2:1 and 0.5:1 depending on active solubility and cardiolipin content. The API is incorporated into the pre-lyophilization bulk solution at 20–50 g/L active concentration, equivalent to 2–5% w/v; total lipid concentration ranges from 12–30 mg/mL. During manufacturing, the lipid film is hydrated in 10 mM histidine buffer at pH 7.0 and dispersed with a rotor-stator at 10,000–15,000 rpm for 15–30 min, then reduced in size with a microfluidizer at 15,000–20,000 psi for 5–8 discrete passes. Extrusion through 200 nm and then 100 nm polycarbonate membranes narrows the vesicle distribution before aseptic filtration through a 0.22 µm PES membrane. Lyophilization is performed with trehalose at a lipid-to-sugar mass ratio of 1:5; the freezing ramp reaches -45°C within 2 h, primary drying is held at -25°C and 0.2 mbar for 48 h, and secondary drying is completed at 25°C for 12 h. Release testing follows USP <71>, USP <788> Method 1, USP <85>, Ph. Eur. 2.6.1, and Ph. Eur. 2.9.19; analytical methods are validated under VICH GL1 and VICH GL2, and stability data are generated under VICH GL3. Aseptic filling is conducted under EU GMP Annex 1. Terminal sterilization is generally avoided because autoclaving at 121°C for 15 min increases free drug leakage above 5% in several bilayer compositions. pH excursions below 5.0 or prolonged exposure above 60°C during downstream dilution can hydrolyze phosphoglyceride esters and must be avoided. Terminal finished dosage forms include single-dose lyophilized vials and sterile suspensions in USP Type I glass vials.

During direct compression of spray-dried liposome powders for veterinary tablets, compaction pressures above 14 kN on a rotary tablet press with flat-faced punches can collapse multilamellar vesicles and produce free drug leakage. The liposome API is added at 10–30% w/w of the final tablet blend for active loads between 5 mg and 50 mg per tablet, with tablet mass held between 150 mg and 500 mg. For hard gelatin capsule filling, the liposome powder addition ratio is 25–45% w/w of the fill weight, with fill weights between 200 mg and 600 mg. The spray-dried powder contains mannitol or trehalose as a cryoprotectant and must have residual moisture below 2.0% before blending. A V-blender is operated at 25 rpm for 15 min with microcrystalline cellulose and croscarmellose sodium; magnesium stearate is held below 1.0% w/w because higher hydrophobic lubricant levels accelerate liposome rupture under compression. Compression on a rotary press is maintained between 8 kN and 14 kN, with turret speed set from 20–40 rpm. Tablet hardness is controlled between 40 N and 80 N; disintegration should complete in <15 min according to USP <701>, and dissolution should reach Q = 80% at 45 min in pH 6.8 phosphate buffer using Apparatus II at 50 rpm according to USP <711>. Coating with hypromellose-based film is performed in a side-vented pan at 50–60°C inlet air and product temperature 38–42°C. Compliance for oral solid dosage forms requires USP <905> content uniformity, Ph. Eur. 2.9.3, VICH GL3, and 21 CFR 211 finished pharmaceutical GMP. Terminal finished product types include film-coated tablets packed in Alu-Alu blisters and hard gelatin capsules.
Replacement of calcium carbonate with rice hulls in a 1:9 medicated premix carrier lowers bulk density from 1.2 g/cm³ to 0.35 g/cm³ and alters the mixing endpoint. The liposome API is first pre-blended 1:10 with a portion of fine carrier, then step-diluted to 1:100 before final ribbon blending. The target addition ratio is 5–25 g/kg of premix, corresponding to final feed concentrations from 20–200 mg/kg depending on species, active potency, and withdrawal requirements. A 1200 L ribbon mixer is operated at 60–70% fill volume and 15–20 rpm for 10–15 min; longer mixing of low-density rice hull carriers separates fines and increases coefficient of variation above 5%. For pelleted medicated feed, conditioning is held at 60–70°C for 30–60 s and die temperature is kept at 70–80°C. Die temperatures above 85°C are not used because the gel-to-liquid crystalline transition of hydrogenated soy phosphatidylcholine occurs near 52–55°C, and extended thermal stress above this range causes bilayer leakage and free drug loss. Regulatory compliance for medicated feed premix production includes 21 CFR 225 CGMP for medicated feed, 21 CFR 558, EU Regulation 1831/2003, VICH GL18 residual solvent limits, and ISO 22000 food safety management. Terminal finished product types include 25 kg multi-wall paper bags of medicated premix, extruded granules, and top-dressed oral powders for swine or poultry rations.

In closed nipple drinker lines fitted to commercial broiler houses, water flow above 1.5 L/min entrains air, and foam fractions above 5% v/v from poorly designed liposome powders cause airlocks in pressure regulators and uneven medication delivery. A water-soluble liposome API is therefore formulated to reconstitute at a final drinking-water concentration of 0.5–5 g/L, prepared from a stock solution of 10–20% w/v in 25–30°C water and metered at 1:100 to 1:200 through a proportioner. The dispersing medium is a citric acid–trisodium citrate buffer at pH 5.5–6.5; propeller stirring at 500–1,000 rpm for 10 min yields a translucent dispersion without high-shear homogenization. The reconstituted solution is passed through a 100 µm inline screen before the proportioner to remove aggregates that would obstruct nipple drinker orifices. Hard water above 200 mg/L as CaCO₃ increases liposome aggregation; additional citric acid chelation is required before stock solution dilution. Stability of the final diluted solution is 24 h at 25°C, after which free drug leakage exceeds acceptable limits if the pH has drifted above 7.0. Compliance for water-medication powders and solutions is based on USP <1231> water for pharmaceutical purposes, Ph. Eur. 2.2.38 conductivity testing, VICH GL3 stability testing, and 21 CFR 211 finished pharmaceutical GMP where the product is classified as a veterinary medicinal solution. Terminal finished product types include water-soluble sachets, oral solutions in HDPE jerrycans, and metering-pump concentrates for automatic drinking water administration.
A liposomal intramammary suspension containing 2–10% w/w active API in a medium-chain triglyceride–aluminum stearate vehicle displays non-Newtonian pseudoplastic flow; viscosity at 25°C is maintained between 50 mPa·s and 200 mPa·s at 10 s⁻¹ shear rate. The total lipid concentration is 5–25% w/w, and the finished syringe contains 10 mL of suspension. The liposome dispersion is sterile-filtered through a 0.22 µm membrane before incorporation into the vehicle; aseptic mixing is performed under vacuum at -0.8 bar with an anchor stirrer at 50–100 rpm to minimize gas entrapment. Terminal sterilization is not applied because gamma irradiation of hydrated liposomes increases free drug leakage and lipid peroxidation. Syringability through a 16 G cannula requires plunger force below 10 N at 10 mm/min; higher lipid loads above 25% w/w are not recommended because cold storage at 4°C increases viscosity beyond 400 mPa·s and causes incomplete evacuation from the syringe. Compliance includes USP <911> viscosity measurement, USP <71> sterility, VICH GL1 and VICH GL2 analytical validation, and 21 CFR 211 aseptic processing requirements. Terminal finished product types include disposable low-density polyethylene intramammary syringes fitted with 16–18 G cannulas.
Following storage at 40°C and 75% RH for 6 months, aqueous film-coated liposomal pellets show a measurable increase in disintegration time because of polymer coalescence and liposome surface rearrangement at the pellet core. The pellets contain 15–35% w/w liposome API and are coated with hypromellose–triethyl citrate to a 3–5% w/w weight gain. Extrusion-spheronization uses a screen aperture of 0.8 mm at 50–100 rpm extruder speed, followed by a spheronizer plate speed of 1,000–2,000 rpm for 5–10 min. The wet mass contains microcrystalline cellulose and lactose with water at 30–40% w/w; drying at 40–45°C for 2–4 h reduces moisture to <2.0% before Wurster coating. Inlet air is set at 50–60°C and product temperature is maintained at 38–42°C; talc is limited to 5% w/w of the coating dispersion to prevent sieve blinding during capsule filling. Coating inlet temperatures above 65°C are avoided because bilayer phase separation can produce surface pores and increase free drug release. Disintegration and dissolution are tested according to USP <701> and USP <711>, with stability protocols under VICH GL3. Terminal finished product types include enteric-coated pellets filled into hard gelatin or hypromellose capsules and multi-unit sprinkle sachets for small-animal oral administration.

Formulated as a multi-dose, route-flexible liposome concentrate, the veterinary-grade API identified by model code LVG-API-07 is released in three sub-designations: LVG-API-07-T for tablets, capsules, powders, granules, and premix; LVG-API-07-I for injections; and LVG-API-07-S for solutions. The composition uses hydrogenated soy phosphatidylcholine, cholesterol, and dipalmitoylphosphatidylglycerol at a weight ratio of 62:33:5 w/w and a molar drug-to-lipid ratio of 1:4.2 to 1:6.0. Release acceptance for Z-average hydrodynamic diameter is 80 nm to 130 nm by dynamic light scattering according to ISO 22412:2017, with polydispersity index <0.15 and zeta potential from -25 mV to -35 mV by ISO 13099-1:2012. Encapsulation efficiency is not less than 88% of label claim and free unencapsulated active ingredient is not more than 6%.
The injection sub-designation is aseptically processed, preservative-free, and endotoxin-controlled; the solution sub-designation may contain benzyl alcohol 1.5% v/v as a multi-dose preservative. All presentations are stored at 2 °C to 8 °C in double polyethylene bags inside aluminum foil pouches with desiccant, with a shelf life assigned at 24 months for lyophilized and spray-dried material.
All presentations are tested against the same core release criteria. Total phospholipid content is 82.0% to 90.0% w/w; lysophosphatidylcholine content is <3.0% w/w to limit membrane-disrupting hydrolysis products. Residual water is <2.0% w/w for lyophilized material and <1.0% w/w for spray-dried powder. Bacterial endotoxins are controlled to <0.25 EU/mg using USP <85> and Ph. Eur. 2.6.14. For injection grade, sterility is tested by USP <71> and Ph. Eur. 2.6.1; the product is sterile-filtered through 0.22 µm membranes and lyophilized in closed isolators. Residual solvents are controlled per USP <467> and ICH Q3C: chloroform <60 ppm, methanol <300 ppm, and ethanol <500 ppm. Elemental impurities meet USP <233>: lead <10 ppm, cadmium <1 ppm, and arsenic <2 ppm. Peroxide value of extracted lipids is <5 meq/kg.

Direct compression performance of LVG-API-07-T is specified on a 10-station instrumented rotary tablet press operating at 45 rpm. A standard blend containing 40% w/w microcrystalline cellulose, 30% w/w mannitol, 4% w/w crospovidone, and 0.8% w/w magnesium stearate yields tablets with hardness 60 N to 90 N, ejection force below 650 N, and friability <0.8% by USP <1216>. Disintegration time in 0.1 M hydrochloric acid at 37 °C is not more than 20 min per USP <701>. Dry powder filling into size 1 hydroxypropylmethylcellulose capsules is targeted at 180 mg to 220 mg fill weight with weight variation <±5% per USP <905>. For oral powders, bulk density is 0.38 g/cm³ to 0.45 g/cm³ and tapped density is 0.52 g/cm³ to 0.60 g/cm³, giving a Carr index between 22 and 27. The powder passes through a 180 µm screen at ≥95% sieve yield.
LVG-API-07-I is reconstituted with water for injection to a nominal active concentration of 20 mg/mL. The resulting dispersion has pH 6.5 to 7.4, osmolality 280 mOsm/kg to 330 mOsm/kg, and viscosity 1.1 mPa·s to 1.4 mPa·s at 25 °C. Terminal filtration is performed through a double 0.22 µm PVDF membrane stack at constant pressure 0.8 bar to 1.2 bar; flux below 18 L/m²/h at 20 °C is rejected as membrane fouling. Post-reconstitution particle size must remain below 130 nm after 5 freeze-thaw cycles between 2 °C and 8 °C. Because phospholipid dispersions are heat-labile, terminal moist-heat sterilization is not assigned to this grade; aseptic processing under ISO 13408-1 applies. Injection-site irritation is scored according to VICH GL43; a score above 2 at 72 h after administration triggers reformulation. The maximum volume per injection site is limited to 10 mL for intramuscular use and 20 mL for subcutaneous use in cattle; for swine the intramuscular limit is 5 mL due to local tolerance constraints.
High-shear granulation of LVG-API-07-T at 200 L to 300 L batch scale is run with impeller speed 120 rpm to 180 rpm and chopper speed 1500 rpm to 2000 rpm. Purified water is added at 12% w/w to 15% w/w over 60 s to 120 s, followed by wet massing for 90 s to 180 s. The dried granule in a fluid-bed dryer retains moisture <2.5% w/w and yields a sieve fraction 250 µm to 850 µm at ≥85%. For medicated feed premix, the product is diluted with 25 kg of calcium carbonate or ground corn carrier in a double-cone blender at 12 rpm for 10 min; blend uniformity is assessed by sampling 10 points and requires a relative standard deviation <5%. The 1% aqueous slurry pH is 6.8 to 7.2.
Dry blending of liposome powder into low-density premix carriers can segregate if particle size difference exceeds 1:4. In those conditions, the granulated form is preferred over direct powder addition. Granulation with polyvinylpyrrolidone 5% w/w binder solution at fluid-bed inlet temperature 60 °C to 70 °C reduces segregation tendency and maintains liposome bilayer integrity provided the product temperature does not exceed 40 °C. The main gel-to-liquid-crystalline transition of the hydrogenated soy phosphatidylcholine bilayer is observed at 52 °C to 55 °C by differential scanning calorimetry per ISO 11357-1. Therefore, drying air above 65 °C without strict granule moisture control is avoided. Production-scale fluid-bed drying in a 120 kg unit at inlet air 60 °C and exhaust 32 °C maintains granule temperature below 38 °C, preserving encapsulation efficiency above 85% in post-dried granules.
LVG-API-07-S is supplied as an aqueous dispersion containing the equivalent of 20 mg/mL active compound, with pH 6.8 to 7.2, zeta potential -28 mV ± 5 mV, and viscosity 1.2 mPa·s ± 0.2 mPa·s at 25 °C. The solution grade passes a 5 µm clarifying filter and remains physically stable for 24 months at 2 °C to 8 °C in the sealed original container. For multi-dose oral application, benzyl alcohol 1.5% v/v is added as preservative per Ph. Eur. 5.1.3; the solution should not be mixed with polyvalent cation-containing drinking water because calcium and magnesium ions above 50 ppm induce surface charge neutralization and aggregation. Diluted field solutions at 1:100 in water should be used within 24 h when no preservative is present.
Pre-drying is required when ambient relative humidity exceeds 60%; the lyophilized cake adsorbs atmospheric water at 1.8% w/w per hour at 75% RH, which raises reconstitution particle size. The product is incompatible with anionic surfactants below the zeta potential zero point; addition of sodium lauryl sulfate above 0.05% w/v collapses the zeta potential to -5 mV and increases particle size beyond 200 nm. Avoid combination with ethanol above 10% v/v during reconstitution because bilayer permeabilization releases payload. For tablet blending, magnesium stearate should remain below 1.2% w/w because extended mixing with higher concentrations reduces tablet hardness by more than 15% due to hydrophobic lubrication of granules.
Compared with a non-encapsulated free-drug API powder, the liposome grade changes the release envelope in standard dissolution testing. Use of USP <711> Apparatus 4 flow-through cell at 16 mL/min in pH 6.8 phosphate buffer at 37 °C gives cumulative release of <45% at 2 h and 78% to 85% at 6 h, compared with >90% within 30 min for the same crystalline free drug at identical particle size. This difference is attributed to the intact lipid bilayer, not to a pH-dependent polymer coat. In tablet formulations, the liposome vehicle also masks bitter payload taste without requiring ethylcellulose or methacrylate coatings. For parenteral administration, the product avoids organic solvent residues associated with solvent-deposited free-drug powders; residual solvent limits are lower than USP <467> options for Class 2 solvents.
Differences from pro-liposome or simple phospholipid blends are equally defined: this product has particle size controlled at release, encapsulation efficiency measured after reconstitution, and zeta potential from -25 mV to -35 mV. A simple phospholipid mixture does not form a unified particle size distribution and may have variable encapsulation capacity. Compared with polymeric nanoparticle APIs, the liposome grade has a phospholipid bilayer that degrades enzymatically in vivo and does not require chronic injection-site residency studies for non-biodegradable polymer fragments. Published data for this specific veterinary model across all species is limited; species-specific bioavailability must be confirmed by VICH GL52 bioequivalence protocols.
