Application of Huanglian Granules Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Swine neonatal enteritis products formulated with Huanglian granules veterinary grade API are manufactured primarily as water-soluble oral powders for drenching or nipple-drinker administration. The API is quantified against berberine hydrochloride reference substance by high-performance liquid chromatography under CVP 2020 monograph methods, while the finished powder is expected to satisfy the aerobic plate count requirements in ISO 4833-1:2013 and the multi-element impurity screening protocol in ISO 17294-2:2016. Because raw herb-derived batches vary in moisture, ash, and alkaloid content, the addition ratio is not fixed as an herb weight but is calculated from the certificate of analysis: the final drinking solution is prepared to deliver 0.05 g/L to 0.15 g/L berberine hydrochloride, corresponding to a dry powder inclusion of 1.0 wt% to 3.0 wt% when using a 10:1 native extract with 10% marker content. In downstream production, the API is pre-milled through a 0.45 mm stainless steel screen, blended stepwise with anhydrous dextrose and citric acid in a ribbon mixer for 20 minutes, passed through a 40-mesh vibratory sieve, and packed into aluminium foil-lined pouches. Terminal product types include oral water-soluble powder in 100 g and 500 g pouches, and a concentrated drench suspension for farrowing-house dosing.

When Feed Premix Delivery Is the Only Practical Route

In broiler integrations where water medication is interrupted by nipple drinker biofilm and pressure fluctuations, feed premix delivery becomes the practical route for sustained intestinal exposure. Regulatory compliance for medicated premises in the European Union is anchored to Regulation (EU) 2019/4, and in the People’s Republic of China to the veterinary drug feed additive registration requirements administered by the Ministry of Agriculture and Rural Affairs; both routes require homogeneous mixing and carry-over control validated by tracer studies. The API addition ratio for a 10:1 Huanglian extract premix falls between 2 kg/t and 5 kg/t when the final feed is fed continuously for 7–10 days; if purified berberine hydrochloride API is selected, the equivalent addition is 0.5 kg/t to 1.2 kg/t calculated on the same berberine-equivalent basis. Stepwise dilution is required before main mixer entry because the fine extract particles tend to agglomerate and adhere to pneumatic conveying lines if added directly. The micro-ingredient pre-blend is prepared with colloidal silica and rice-hull carrier in a double-shaft paddle mixer with coefficient of variation ≤5%, then transferred into a horizontal ribbon mixer before conditioning at 80°C and pelletizing through a 3 mm die. Terminal product types include a 0.2% veterinary drug premix, pelleted broiler feed, and crumbles for floor-reared birds.

What Limits Solubility in Hard-Water Drinking Lines?

The principal manufacturing failure in field solutions is not intrinsic insolubility but pH-dependent precipitation of berberine hydrochloride in alkaline hard water. Working solutions prepared from stock concentrates can drop out in pressure regulators and nipple lines when distribution water contains more than 300 mg/L calcium carbonate equivalent and the solution pH rises above 6.0. The design response is to prepare a 100× stock concentrate adjusted to pH 4.5–5.5 with citric acid monohydrate, then inject the concentrate into the water line through a positive-displacement dosing pump at 1–3 L/h and mix through a Venturi injector upstream of the pressure regulating station. Addition ratio: stock concentrates contain 5.0 wt% to 8.0 wt% of the API when the certificate of analysis indicates berberine hydrochloride not less than 10%, yielding a final drinking water concentration of 0.05–0.15 g/L. Compliance for the finished concentrate includes the pharmaceutical water quality expectations described in USP <1231> and the sample preparation procedures in ISO 6887-1:2017 for microbiological examination. Terminal product types are oral solution concentrate in 1 L or 5 L HDPE bottles and a field-use dosing kit with citric acid buffer pre-weighed for the prevailing water hardness. Incompatibility: the API should not be prepared in phosphate-buffered stock solutions because berberine phosphate precipitates at ambient temperature.

Ruminant Oral Bolus Binding and Release Kinetics

For pre-ruminant calves, Huanglian API is manufactured into oral boluses and tablets in which release depends on disintegration and wetting rather than enteric coating, because the target site is the abomasal and intestinal lumen. Published dissolution data for this specific configuration are limited; release criteria are therefore established from the manufacturer’s pilot data and validated against the disintegration test in Ph. Eur. 2.9.1 and the dissolution test in Ph. Eur. 2.9.3. Addition ratio per 1.5 g bolus is 0.25 g of a 10:1 extract, adjusted so that each dose delivers 25–50 mg berberine hydrochloride; when purified API is used, the amount is reduced to 0.02–0.05 g per bolus. The granulation process uses povidone K30 solution in a high-shear mixer at impeller speed 150 rpm and chopper speed 1500 rpm, with wet massing for 3–5 minutes, followed by fluid-bed drying at inlet air 55°C, lubrication with magnesium stearate 0.5 wt%, and compression at 10–15 kN on a rotary tablet press. Terminal product types include a 1.5 g oral bolus, a 0.5 g tablet for calf dosing, and a drench suspension for neonatal administration.

High-temperature extrusion of aquafeed containing herbal extracts is constrained by expansion ratio and post-extrusion oil coating. Huanglian premixes for grass carp and tilapia enteritis are therefore introduced as a vacuum-coated powder after the drying zone rather than before extrusion, which preserves the berberine marker and avoids thermal degradation in the conditioning barrel. Compliance for the feed mill includes ISO 22000:2018 and HACCP critical control points at sifting, batch weighing, and vacuum coating. The addition ratio for a 10:1 extract is 1.5–3.0 kg/t of finished aquafeed, while purified API is used at 0.3–0.8 kg/t when calculated on the same berberine-equivalent basis. In the vacuum coating step, the API is dispersed in fish oil and applied at 0.8 bar vacuum to ensure penetration into the outer matrix of the pellet. Terminal product types include sinking extruded pellets for pond feeding and slow-sinking granules for intensive recirculating systems.

Coating Companion Animal Capsules Without Hygroscopic Cracking

Huanglian extract API is hygroscopic, and capsule fill formulations for canine and feline gastrointestinal support require pre-drying, moisture-barrier packaging, and controlled humidity in the filling suite. The capsule formulation is expected to comply with the loss-on-drying methodology in USP <2040> and the stability protocol in ICH Q1A(R2). Addition ratio per 500 mg capsule is 100 mg of a 10:1 extract, equivalent to 10 mg berberine hydrochloride; if purified API is used, the amount is 5–15 mg per capsule. The production process requires drying the blended powder to less than 5% moisture in a fluid-bed dryer at 45°C before filling, adding fumed silica 0.5 wt% as a flow aid and moisture scavenger, filling on a dosator capsule machine at 40,000–80,000 units/h, dedusting, and packing into cold-form aluminium blister with a desiccant sachet. Terminal product types include a 500 mg hard capsule for companion animal use and a 250 mg direct-compression tablet for small-breed dogs and cats. The boundary condition is relative humidity: filling should not proceed when ambient relative humidity exceeds 60% without independent dehumidification.

Sterile filtration of berberine hydrochloride injectables is constrained by pH-dependent solubility, heat sensitivity, and bacterial endotoxin removal. Injectable formulations are reserved for acute large-animal gastrointestinal infection under veterinary prescription and are manufactured in an aseptic processing line. Compliance requires the sterility test in USP <71>, the bacterial endotoxin test in USP <85>, and the aseptic processing expectations in EU GMP Annex 1; terminal sterilization at F0 ≥ 8 minutes is used only where stability data support thermal exposure. Addition ratio in the finished injection is 0.5–1.0 mg/mL berberine hydrochloride, prepared from API with assay not less than 98%. The process includes dissolution in Water for Injection at 50°C, pH adjustment to 4.0–4.5 with hydrochloric acid, activated carbon treatment at 0.1 wt% for 15 minutes, prefiltration through a 0.45 µm membrane, sterile filtration through a 0.22 µm membrane, and filling into amber ampoules under Grade A environment. Phosphate-buffered saline is not an acceptable diluent because berberine phosphate precipitates under refrigeration. Terminal product types are 2 mL, 5 mL, and 10 mL injection solutions for intramuscular administration in cattle and swine.

    More Introduction

Huanglian Granules Veterinary Grade API is a standardized dried extract of Rhizoma Coptidis prepared as a granular active pharmaceutical intermediate for downstream manufacture of tablets, injections, capsules, powders, granules, premixes, and solutions. The product is supplied under model designation HL-VET-API-G-2207 for the spray-dried granular form and HL-VET-API-G-2210 for the vacuum-dried granular form with reduced residual solvent. It is not a final dosage form and is not labelled as sterile. The granule contains the native protoberberine alkaloid fraction, including berberine hydrochloride, palmatine hydrochloride, jatrorrhizine, and coptisine, in a dried extract matrix. Release criteria are set for total protoberberine alkaloids calculated as berberine hydrochloride, individual berberine hydrochloride content, loss on drying, acid-insoluble ash, heavy metals, arsenic, residual solvents, and microbial load. Because the same granule cannot simultaneously meet all finished-form requirements, the model code, particle-size cut, and endotoxin burden are selected before drying. Tablet and capsule grades are milled to a volume mean diameter of 80–150 µm; premix grades are sieved to 150–420 µm to reduce segregation in mash feed; injection grades require further depyrogenation and sterile filtration after dissolution, because the intact granule is not injectable.

    Specification Profile and Pharmacopoeial Alignment

Because the granule is a multi-alkaloid botanical intermediate, the release profile is divided into chromatographic composition, inorganic safety, and microbiological quality. The method platform is high-performance liquid chromatography with diode-array detection for alkaloid quantification, combined with compendial limit tests. The following table lists representative release criteria used when the product is ordered as a non-sterile veterinary API.

When the intended use is injection, an additional bacterial endotoxin criterion of <0.50 EU/mg is applied to the solution before filling, not to the dry granule as shipped. The dry granule is not depyrogenated by default. Because contract granulation conditions can shift total alkaloid recovery, each lot should be confirmed against the certificate of analysis before use in finished-product manufacture.

For injectable formulations, the limiting factor is not alkaloid content but the combination of bacterial endotoxin burden, insoluble particulate matter, and cold-water dissolution behaviour. The dry granule is manufactured as a non-sterile intermediate; therefore, direct reconstitution and aseptic filling without depyrogenation is outside its operational boundary. Injection-grade manufacture requires dissolution in heated water at 60–80 °C, pH adjustment to 3.0–4.0 with a pharmaceutically acceptable acid, activated-carbon treatment at 0.1–0.3% w/v for 20–30 min, followed by filtration through 0.45 µm and 0.22 µm membrane filters and terminal sterilisation only after degradation studies. Sterile filtration removes insoluble extract matrix but not all endotoxin; depyrogenation must be demonstrated by bacterial endotoxin testing per CPV 2020 1143. The pH-dependent stability of berberine hydrochloride in aqueous solution is the principal reason that autoclaving profiles must be developed for each formulation. Published degradation data for this specific multi-alkaloid granule in injectable vehicles are limited, so forced degradation and photostability studies are required before a terminal sterilisation cycle is selected.

When the Granule Is Intended for Tablet or Capsule Blending

Tablet and capsule manufacture exposes the granule to compaction stress, flow constraints, and moisture-related defects. Direct compression is possible only when the granule has a tapped density of 0.45–0.65 g/mL, an angle of repose below 35°, and a loss on drying below 5.0%. If loss on drying exceeds 5.0%, punch sticking and picking are observed on rotary tablet presses; if moisture falls below 1.0%, capping and lamination become more frequent. A typical direct-compression matrix blends the granule with microcrystalline cellulose at 20–40% w/w, crospovidone at 2–5% w/w, colloidal silicon dioxide at 0.5–1.5% w/w, and magnesium stearate at 0.5–1.0% w/w. For wet granulation, a high-shear mixer or fluid-bed granulator is used; the binder solution is sprayed at product temperature 30–40 °C, and the drying inlet air is maintained at 60–70 °C to avoid thermal degradation. Tablet disintegration is controlled to ≤15 min in 0.1 M hydrochloric acid using the apparatus described in CPV 2020 0921. Capsule filling requires a granule with a Carr index below 25 and consistent bulk density to maintain fill weight variation within ±5%. After compression or encapsulation, the finished product should be protected from light because berberine-related alkaloids undergo photolytic fading.

Across the seven finished-form routes, the same granule lot can be used only after verifying a small set of grade-specific parameters. The following matrix summarises the critical granule property for each route.

In mash feed and mineral premixes, the principal failure mode is segregation, not chemical instability. The spray-dried granule is hygroscopic and should be stored below 60% relative humidity; above this threshold, the surface may cake and the sieve profile may shift. Premix grades are typically blended with ground corn, rice hulls, or calcium carbonate carrier to a target total alkaloid concentration determined by the veterinarian and the target species. Published dosing data for this specific granule across all veterinary species are limited; the inclusion rate must be derived from the assayed total alkaloid content and the prescribed berberine dose. To reduce segregation, the granule is sieved to 150–420 µm, and the carrier particle-size distribution is matched so that the mean diameter ratio is not greater than 3:1. Batch-to-batch variation in particle-size span above 1.8 has been observed to cause alkaloid assay drift at the top and bottom of V-blender discharge. Sampling plans should therefore include at least 3 positions at discharge after 15 min of blending.

Solution Preparation and pH-Dependent Solubility Constraints

Aqueous reconstitution of the granule does not yield a true solution at ambient temperature. The granule matrix disperses partly, but the native protoberberine alkaloids remain largely associated with extract components at neutral pH. Oral solution preparation therefore uses heated water at 60–80 °C, pH adjustment to 3.5–5.0, and where permitted, a co-solvent system of propylene glycol or glycerol at 10–30% v/v. The resulting liquid should be clarified through a 0.45 µm filter before filling into amber glass or opaque plastic containers. For injectable solutions, a 0.22 µm sterilising-grade filter is used only after depyrogenation and only when the formulation has been shown to be stable at the selected pH and temperature. The pH must not be adjusted with strong alkali because berberine salts can precipitate as the free base above pH 6.0. Light protection is mandatory for all liquid forms because the protoberberine chromophore is light-sensitive. Residual particulate matter in solution should be monitored by light obscuration per CPV 2020 0903; if the count exceeds 25 particles/mL at ≥10 µm, the clarification step is inadequate for the chosen vehicle.

A direct substitution of crude Huanglian powder is not microbiologically equivalent

Relative to crude Rhizoma Coptidis powder, the granular API differs in standardised alkaloid content, microbial burden, heavy-metal load, and particle-size regularity. Crude powder can vary in berberine content by more than 2-fold depending on geographic origin, harvest year, and drying method; the granulated API is normalised by blending extract lots to the release range. The granular form also reduces total aerobic microbial count from values that can exceed 10⁴ CFU/g in poorly dried botanical material to ≤10³ CFU/g. Compared with purified berberine hydrochloride, the granule retains palmatine, jatrorrhizine, and coptisine, which may alter the pharmacological response, but the multi-alkaloid matrix has lower aqueous solubility and higher hygroscopicity than the isolated salt. Compared with human-grade Coptis extract, the veterinary-grade API is controlled under veterinary-specific residual solvent and elemental impurity assessments aligned with VICH GL18 and the receiving jurisdiction’s veterinary pharmacopoeia. Human-grade extract is not automatically suitable for veterinary use because species-specific excipient tolerances, residue limits, and target daily doses differ. The granule is not interchangeable with feed-grade herbal powders that lack a full release specification; for inclusion in medicated feed, only lots with a certificate of analysis covering assay, heavy metals, residual solvents, and microbial limits should be accepted.
