Application of Halimide Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Ruminant oral drench, oral powder, and medicated premix use. Halimide Veterinary Grade API enters ruminant endoparasite control programmes through oral drench, oral powder, and medicated premix lines where the active is dispersed onto a carrier before stepwise dilution into finished feed. On a production-scale ribbon blender with working volume 1200 L and fill ratio 0.62, the API is first pre-blended with colloidal silicon dioxide at 1:9 w/w for 10 min, then combined with ground corn cob carrier, microcrystalline cellulose, and lactose monohydrate. The addition rate in the oral premix is kept within 0.5–2.0% w/w, with finished feed carryover typically 20–100 g/tonne depending on target species and local maximum residue limit approvals. Manufacturing follows EU Regulation 2019/4 for medicated feed and, for shipments to the United States, 21 CFR Part 225; residual solvent control follows VICH GL18; final moisture is held below 10.0% w/w using ISO 6496:1999; microbial enumeration follows ISO 4833-1:2013. The downstream process is a three-stage geometric dilution: stage one blends the API–silica masterbatch with 5 kg carrier for 15 min; stage two extends to 50 kg in a paddle mixer at 12 rpm for 20 min; stage three transfers to a double-cone blender to complete a 1000 kg batch. Content uniformity acceptance is a coefficient of variation <5.0% per VICH GL9. The blend is sifted through a 500 µm sieve and packed into PE-lined paper sacks with headspace nitrogen purging. Finished product types include 0.5% w/w oral powder, 2.0% w/w medicated premix, and 1.5% w/v oral drench solution. A known process boundary is moisture pickup above 12.0% w/w in the carrier, which causes particle agglomeration and assay loss after 60 days at 30°C; producers therefore pre-dry corn cob to <8.0% w/w before blending.

What limits terminal sterilisation in Halimide Veterinary Grade API injectable solutions?

In cattle and sheep injectable programmes, Halimide Veterinary Grade API is formulated as a 5.0% w/v solution in a non-aqueous water-miscible system of propylene glycol 60% v/v, benzyl alcohol 1.0% v/v, and water for injection q.s. The API is dissolved at 55–60°C, cooled to 25°C, and adjusted to pH 8.0–8.5 with 1 M sodium hydroxide before making to volume. The addition ratio in the injectable concentrate is 5.0% w/v active, corresponding to 50 mg/mL, and the product is filled into 50 mL single-dose and 250 mL multidose vials under Grade C conditions with terminal sterilisation or aseptic filtration. Sterility is verified under USP <71>, bacterial endotoxins under USP <85>, particulate matter under USP <788>, and visible particulates under USP <790>; the injection route in the United States is governed by 21 CFR Part 522 approval requirements. The principal process conflict is terminal sterilisation: an autoclave cycle at 121°C for 15 min can produce a pH drift greater than 0.3 pH units and raise subvisible particles ≥10 µm when the fill volume exceeds 100 mL and stopper headspace oxygen is not purged. Aseptic filtration through a 0.22 µm PVDF membrane is therefore used for heat-sensitive batches, with a maximum hold time of 4 h between dissolution and filter integrity testing. Incompatibility is observed with chlorobutyl stoppers that have not been pre-washed; stoppers are autoclaved separately and rinsed with water for injection to reduce extractables. Finished product types are injectable solution for subcutaneous administration in cattle and sheep, supplied in multidose amber glass vials. Published data for this specific configuration is limited, so every sterilisation cycle is bracketed by pilot-scale qualification with product assay and pH drift tracked at 0, 6, and 12 months under 25°C/60% RH and 40°C/75% RH.

Swine grower-finisher feed-grade granules utilise Halimide Veterinary Grade API at 1.0% w/w in a Type B medicated premix that is extended 1:50 into complete feed before pelleting. The granule line consists of a vertical high-shear granulator with an impeller speed of 150–250 rpm and a chopper speed of 1500–3000 rpm, a fluid bed dryer with inlet air at 65–75°C, and a conical mill fitted with a 1.2 mm rasping screen. The binder is povidone K30 dissolved in purified water to 5.0% w/v, added at 12.0% w/w of the dry charge; granulation endpoint is judged by an initial impeller torque increase of 15–20% over the dry mix baseline. The granulate is dried to 2.0–3.0% w/w loss on drying and milled to a particle size between 180 µm and 850 µm. The process deliberately avoids pre-pelleting addition because exposure to feed conditioning at 80°C for 40 s reduces assay by 2–4% in uncoated granules; post-pelleting dry dosing or post-conditioning liquid addition is preferred. Compliance for this segment references 21 CFR Part 225 and 21 CFR Part 226 for medicated feed manufacturing, ISO 6497:2002 for sampling, ISO 4833-1:2013 for enterobacteriaceae and total viable count, and VICH GL18 for residual solvents. Finished product types include 1.0% w/w feed granules, 0.5% w/w top-dress powders, and 5.0% w/w intermediate premix for further dilution by feed mills. A specific limitation is incompatible combination with alkaline mineral mixes containing more than 3.0% w/w calcium oxide or sodium bicarbonate, which raises local pH above 8.5 and accelerates hydrolysis in stored complete feed.

Broiler drinking water solutions and premix compatibility

In broiler integrators where water medication is used, Halimide Veterinary Grade API is prepared as a water-soluble powder at 20.0% w/w active content and administered in drinking water at 0.02–0.05% w/v final medicated water concentration. The powder is manufactured in a low-shear paddle mixer of 500 kg capacity using a citric acid–sodium bicarbonate effervescent couple at 35.0% w/w and lactose monohydrate at 44.5% w/w; the effervescent system ensures dispersion within 5 min in water at 15–20°C without high-shear mixing. Dissolution and dispersion are tested using Ph. Eur. 2.9.3, and finished water solubility is checked by measuring residue after passing through a 0.45 µm filter. Residual solvent limits are controlled under VICH GL18; the marketing authorisation file follows EU Regulation 2019/6 for veterinary medicinal products. The finished product types are 20.0% w/w water-soluble powder and 0.05% w/v drinking water solution prepared on farm. A critical process boundary is acidification of drinking water with organic acids below pH 5.5, which causes the active to precipitate as free acid and reduces recovered concentration by up to 18% after 2 h; water chlorination above 2 ppm free chlorine is also avoided because oxidative degradation produces two unidentified non-active peaks in HPLC analysis. Stability studies under VICH GL11 show that the soluble powder in aluminium-foil sachets remains within specification for 24 months at 25°C/60% RH but drops to 6 months at 40°C/75% RH.

When Halimide Veterinary Grade API is compressed into tablets or filled into capsules for companion animal dosing

Companion animal dosage forms based on Halimide Veterinary Grade API require wet granulation because the API batch density is typically 0.35 g/mL and flow through a 2 mm funnel is insufficient for direct compression. A representative 50 mg tablet consists of Halimide 25.0% w/w, lactose monohydrate 60.0% w/w, microcrystalline cellulose 10.0% w/w, croscarmellose sodium 4.0% w/w, and magnesium stearate 1.0% w/w. Granulation uses povidone K30 as a 5.0% w/v binder solution in isopropanol–water 1:1, added at 10–12% w/w; drying in a fluid bed at 55–65°C continues until loss on drying is 1.5–2.5% w/w. Compression on a rotary press runs at 35–50 rpm with a compression force of 14–18 kN, target hardness 80–120 N, friability <1.0% per USP <1216>, and disintegration <15 min per USP <2040>. Capsule filling for a 50 mg capsule in size 2 gelatin shells uses the same granulate, with sodium lauryl sulfate 0.5% w/w added intragranularly to improve wetting; fill weight is 200 mg and uniformity of dosage units is tested under USP <905>. Dissolution acceptance is Q=75% in 45 min using USP <711> apparatus 2 at 50 rpm in 900 mL of pH 6.8 phosphate buffer. Finished product types are 50 mg tablets and 50 mg immediate-release capsules for companion animal species where local marketing authorisation provides an indication, packaged in PVC/PE/PVDC blister strips with aluminium foil. In high-humidity facilities where ambient relative humidity exceeds 60%, sticking and picking during compression cannot be controlled by magnesium stearate alone; the granulation suite is therefore maintained at 45±5% RH and 20±2°C to prevent water uptake above 2.0% w/w in the final blend.

Granule and powder line design under high-humidity tropical conditions

Production of Halimide Veterinary Grade API granules and powders in tropical sites with ambient humidity above 75% is constrained by the blend’s hygroscopic behaviour and requires dehumidified suites with supply air at 22±2°C and 45±5% RH. A dry powder for reconstitution into oral solution is prepared at 10.0% w/w Halimide, mannitol 70.0% w/w, sodium citrate 10.0% w/w, xanthan gum 2.0% w/w, and sucralose 0.1% w/w; the powder is blended in a low-shear paddle mixer at 10 rpm for 20 min, then vacuum-dried at 40–45°C under -0.08 MPa for 4 h before sifting through a 355 µm sieve. The environment is classified as ISO 14644-1 Class 8 for non-sterile powder operations, and moisture is tested under ISO 6496:1999 with acceptance below 5.0% w/w. The addition ratio for this oral powder is 10.0% w/w, and the reconstituted solution is typically diluted 1:200 into drinking water for swine or poultry according to local prescription. The finished product types include 10.0% w/w granules for reconstitution and 5.0% w/w oral powders in single-dose sachets. A production bottleneck observed in these conditions is caking of the spray-dried mannitol when residual moisture exceeds 6.0% w/w, which increases tapped density from 0.45 g/mL to 0.62 g/mL and blocks the sieve; sacks are therefore opened only in the dispensed booth and bulk material is never stored overnight. Any deviation above 50% RH during packaging is corrected by increasing nitrogen purge in the aluminium-lined sachet to 0.4 L/min.

    More Introduction

Halimide Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions is supplied as a crystalline active pharmaceutical ingredient intended for further manufacture into veterinary dosage forms. Three article codes are available: HLM-VET-P, an oral powder; HLM-VET-G, a granulation-optimized low-dust powder; and HLM-VET-S, a sterile low-endotoxin injection grade. The API is released against a pharmacopoeial-style specification with assay by HPLC in accordance with USP <621> and Ph. Eur. 2.2.29; assay acceptance is 98.0–102.0% on the dried basis. Residual solvents are controlled under VICH GL18, residual water by Karl Fischer USP <921>, and elemental impurities by ICP-MS using the risk-based limits of ICH Q3D. Because this is an active substance and not a finished dosage form, the powder requires identity confirmation, supplier qualification, and formulation-specific stability testing before veterinary use.

Identification is by infrared absorption spectrophotometry against a reference standard and by X-ray powder diffraction; the crystalline pattern has principal reflections at 7.2°, 14.8°, 19.5°, 24.1°, and 27.6° 2θ when scanned at 2° per min with Cu-Kα radiation. The melting event is observed at 214°C by differential scanning calorimetry at 10°C/min under nitrogen; decomposition onset is 240°C. These data distinguish the veterinary grade from amorphous spray-dried material and from other salt forms.

What release specifications define Type S against oral Type P?

The critical distinction between HLM-VET-S and the oral grades is microbial quality and particle-size reduction. HLM-VET-S is intended for injection or sterile solution manufacture and is controlled for bacterial endotoxin and sterility; oral grades are not. The following release parameters are included on certificates of analysis.

The endotoxin limit of <0.50 EU/mg is set so that the final injection does not exceed the 5 EU/kg threshold described in USP <85>. At a dose of 10 mg/kg in a 500 kg animal, the calculated endotoxin contribution is 2,500 EU, which equals the threshold; therefore, high-dose formulations may require a lot-specific limit of 0.10 EU/mg or lower. The reduced water limit for HLM-VET-S is necessary because residual moisture accelerates hydrolytic degradation during terminal sterilization.

Particle-size control is relevant because HLM-VET-P is intended for dry blending before wet granulation; laser diffraction D90 values above 250 µm have been associated with content uniformity failures in low-dose tablet batches where active weight per tablet is below 10 mg. For HLM-VET-G, the specified D90 of ≤150 µm is paired with a specific surface area of 0.8–1.5 m²/g by a 5-point nitrogen adsorption BET method following ISO 9277. This surface area range balances granulating liquid uptake without over-wetting.

Thermal and moisture sensitivity in granulated oral forms

Wet granulation with HLM-VET-P or HLM-VET-G is limited by a hydrate transition that can occur above 55°C in aqueous granulating fluid. In a high-shear mixer with impeller tip speed of 5–10 m/s, product temperature is maintained below 50°C to avoid polymorph conversion; near-infrared moisture probes monitor the transition. Granulation end point is controlled by power consumption rather than fixed time because batch viscosity rises sharply when water content exceeds 12–14% w/w. Fluid-bed drying with inlet air at 60°C and final product temperature of 40–45°C produces loss-on-drying of 1.5–2.5% w/w; XRPD verifies preservation of the crystalline form. The API is incompatible with strong oxidizing agents and should not be hammer-milled at temperatures above 40°C, because localized frictional heating can generate more than 2.0% amorphous content and reduce subsequent dosage-form stability.

Direct compression with HLM-VET-P is not recommended when active loading is below 10 mg per tablet unless a premix step is used; published data for this specific configuration is limited, and feasibility batches are required. High-shear blending of a 10–20% active premix with lactose monohydrate and microcrystalline cellulose in a 100 L bin blender at 10 rpm for 5 min achieves blend uniformity with relative standard deviation <3.0% for 500 kg batches. Tablets compressed on a 16-station rotary press at 10–18 kN punch force show tensile strength of 1.2–1.8 MPa and friability <0.5% by USP <1216>. Capsule filling with HLM-VET-G on a dosator-type machine requires bulk density of 0.45–0.60 g/mL; magnesium stearate should not exceed 1.0% w/w, or disintegration exceeds 15 min in 0.1 N HCl at 37°C. Dissolution testing with USP Apparatus 2 at 50 rpm in 0.1 N HCl shows ≥80% release in 20 min when croscarmellose sodium is present at 2.5% and magnesium stearate is 0.5%.

When a low-dust granular API is preferred for medicated premix production

For feed-mill premix production, HLM-VET-G is selected over HLM-VET-P because the controlled particle-size distribution reduces dust generation and cross-contamination. In a horizontal ribbon mixer with 500 kg capacity and 50 rpm mixing speed, a 10% active premix on calcium carbonate carrier reaches blend coefficient of variation <3.0% after 15 min. The granular API can be further diluted with ground corn or soybean meal to 1.0–20.0% active content. Electrostatic adhesion can occur below 30% RH; therefore blending areas are controlled at 45–60% RH. The powder does not require pre-drying at these conditions. Premix stored in paper bags with low-density polyethylene liners at 25°C/60% RH retains assay ≥99% for 12 months.

Injectable preparations use HLM-VET-S dissolved in Water for Injection at 20–40 mg/mL. The pH is adjusted to 4.0–5.5 with hydrochloric acid or sodium hydroxide before terminal sterilization at 121°C for 15 min, equivalent to F0 ≥15 min. At pH >6.5, hydrolytic degradation increases; pH 7.4 produces 2.1% degradant after one cycle. Sterile filtration through a 0.22 µm PVDF membrane is acceptable for heat-sensitive formulations provided filter adsorption is ≤1.0% of the assayed active. The resulting solution is protected from light and stored below 25°C; repeated autoclaving is not recommended.

Comparative hygroscopicity and pH-solubility boundaries in salt-form selections

Halimide Veterinary Grade API is the free base form. Compared with hydrochloride or sulfate salt alternatives used in other veterinary actives, the free base has lower moisture uptake: dynamic vapor sorption at 25°C/80% RH gives 0.8–1.2% mass gain, whereas many hydrochloride salts exhibit 4.0–5.5% under the same conditions. This reduces sticking during tablet compression and improves handling in high-humidity feed mills. Aqueous solubility is pH-dependent: 18 mg/mL at pH 4.0, 5 mg/mL at pH 5.5, and 0.4 mg/mL at pH 6.8. Consequently, oral solution formulations require a buffer system below pH 5.0; formulations above pH 6.0 risk precipitation. The powder contains less than <0.05% residual ethanol and no Class 1 solvents by GC-HS. Binary mixture compatibility at 40°C/75% RH for 4 weeks shows no greater than 0.2% degradant formation with lactose monohydrate, microcrystalline cellulose, croscarmellose sodium, or magnesium stearate. Citric acid mixtures show 0.8% degradant; if effervescent granules are formulated, anhydrous citric acid should be limited to ≤5.0% and granulation dried to ≤0.5% water. The API does not contain lactose, gluten, porcine-derived materials, or BSE/TSE materials. Unlike non-veterinary Halimide industrial material, the veterinary grade is manufactured under EU GMP Part II with dedicated equipment and is not intended for human medicine.

Oral solution manufacture uses HLM-VET-P or HLM-VET-G in purified water with sodium benzoate 0.1% and citric acid 0.05% as antimicrobial and buffer. The solution in amber HDPE bottles retains ≥98% assay at 25°C/60% RH for 30 days, and ≥98% assay at 2–8°C for 90 days; pH drift is <0.2 units. Because the free base has a pKa of 7.1, hydrochloric acid or sodium hydroxide is added slowly with mixing to avoid localized pH excursions that cause precipitation.

For tablets and capsules, the oral grades are double polyethylene-bagged inside a fiber drum; the inner bag is purged with nitrogen. Recommended storage is 15–25°C in a dry place, with a re-test period of 24 months when stored in the original sealed container. Opened containers should be re-tested for residual water before use if relative humidity exceeds 60%, because moisture uptake can exceed 0.2% within 8 h in an unprotected open hold. HLM-VET-S is packaged in sterile polyethylene terephthalate bottles with tamper-evident seals and should be opened only in an ISO 5 environment.
