Application of Ganfan Jiedu Tablets Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Downstream formulation scopes for Ganfan Jiedu Tablets Veterinary Grade API are limited to the registered veterinary finished-product categories of tablets, injectable solutions, hard capsules, medicated premixes, oral solutions, and granules/sachets. Each processing route imposes its own physical and microbial limits; the API charge is calculated as M_api=M_label/(P×(1−LOD)), where P is the assay purity expressed as a fraction and LOD is loss on drying. The numerical examples below are batch-equation demonstrations, not approved label claims; the controlling commercial formula must be taken from the registration dossier.

Compression Physics of a High-Potency Veterinary Granular API

Finished veterinary tablets containing Ganfan Jiedu Tablets Veterinary Grade API are released against USP <905> uniformity of dosage units, USP <1217> tablet breaking force, USP <701> disintegration, and, where justified by the registration dossier, USP <711> dissolution. Finished-tablet production in facilities exporting to the United States must comply with 21 CFR 211.110 in-process blend uniformity requirements and 21 CFR 211.165 finished-product release testing. Within the European Economic Area, the tablet line falls under EU Regulation 2019/6, and the API supplier is expected to operate under ICH Q7 Part II for active substances used in veterinary medicinal products. Because the API is typically a dried botanical extract powder, the Chinese Veterinary Pharmacopoeia tablet monograph may impose additional loss-on-drying, heavy-metal, and marker-compound limits that supersede generic USP chapters when the product is registered in China.

The batch equation for a 600 mg core-mass tablet with a 100 mg label claim and an API lot assay of 98.5% purity with 4.0% LOD is M_api=100/(0.985×0.96)=105.8 mg per tablet, equivalent to 17.6 wt% of the pre-compression blend. Direct compression is rarely used without a prior densification step because the dry extract exhibits low bulk density, poor flow, and a tendency to laminate at compaction pressures above 18 kN. A roll-compaction step operating at 18–24 kN/cm roll pressure, 1.5–2.0 mm roll gap, and 120–160 rpm mill speed, followed by a 0.8 mm oscillating screen, converts the API and microcrystalline cellulose into a flowable granulate. Final blending in a tumble mixer at 60–70% of nominal capacity for 15–25 minutes with crospovidone and sodium stearyl fumarate then feeds a rotary tablet press set to 8–14 kN compression force and 30–60 rpm turret speed. Moisture must remain below 45% RH during compression; lot records from full-scale runs show punch sticking and ejection-force drift when room humidity exceeds 55% RH and granule LOD rises above 3.5%.

Terminal finished forms include film-coated veterinary tablets, scored tablets for body-weight dose adjustment, and, where the extract is sufficiently palatable, chewable veterinary tablets. Water-based film-coating systems are preferred over solvent-based coatings because residual moisture after coating must be below 5.0% to prevent tablet softening. The route is not appropriate for products requiring immediate disintegration in less than 2 minutes, because the densified granulate and film-coating thickness of 20–40 µm together extend the USP <701> disintegration time toward the 30-minute limit.

Aseptic processing of Ganfan Jiedu Tablets Veterinary Grade API into injectable solutions is governed by a different constraint set than solid dosage form manufacture; the limiting variables are sterility, bacterial endotoxin, particulate matter, and filter compatibility rather than flow and compaction. Injectable finished products must meet USP <1> general chapter Injections, USP <71> sterility, USP <85> bacterial endotoxins, and USP <790> visible particulate matter. The EU aseptic operation must comply with EU GMP Annex 1 sections 8.9 for filter integrity testing and 9.32 for pre-filtration bioburden limits, with a target of ≤ 10 CFU/100 mL before sterilizing filtration. For a 10 mg/mL injectable solution, the corrected API charge at 98.0% purity and 3.5% LOD is 10.58 g per liter of final solution; the API is dispersed in water-for-injection at 20–25°C under vacuum, because high-shear rotor-stator mixing can introduce foam and denature heat-sensitive marker compounds. Phase-solubility screening in water, 0.9% sodium chloride, and propylene glycol-water systems is mandatory; published data for this specific configuration is limited, so the development batch record should include pH-solubility curves from pH 3.0 to pH 8.0 and a forced-degradation hold at 40°C for 72 hours before filter selection.

The solution is adjusted to the target pH with 0.1 N hydrochloric acid or sodium hydroxide, then passed through a 0.22 µm polyvinylidene fluoride or polyethersulfone sterilizing filter; nylon membranes are not recommended because of adsorptive loss of polyphenolic marker compounds observed in full-scale filtration trials. Aseptic filling into grade A surroundings with grade B background uses stoppering and capping equipment validated by media-fill runs. Terminal steam sterilization at 121°C for 15 minutes is generally unsuitable for this API class unless differential scanning calorimetry and assay stability data demonstrate less than 5.0% marker degradation; otherwise aseptic filtration is the only compatible sterilization route. Finished injectable forms include 10 mL, 50 mL, and 100 mL multi-dose vials and, where national registration permits, single-dose ampoules. The principal operational boundary is holding time: the filtered bulk solution should be filled within 4–8 hours and must be protected from light, because prolonged exposure at >25°C can increase color intensity and generate visible particulates without necessarily altering total assay.

What Changes When the API Is Dry-Diluted into a Mineral Carrier for Medicated Feed?

The medicated-premix route moves the analytical target from dosage-unit uniformity to distribution uniformity in a dry carrier under EU Regulation 2019/4 on medicated feed, with complementary limits on undesirable substances under Directive 2002/32/EC. Facilities producing feed premixes for export to the United States must observe 21 CFR 558.3 and 21 CFR 558.4, and feed-safety management systems are typically certified to ISO 22000. The primary process risk is segregation after mixing, because the API and the mineral or rice-hull carrier have different particle densities; in full-scale ribbon-mixer batches, segregation-induced assay drift can exceed 5.0% relative standard deviation when discharge gate positioning or hopper transfer height is uncontrolled. A 5.0 wt% intermediate premix is produced by charging 50 kg API, 10 kg colloidal silicon dioxide as anti-caking agent, and 940 kg ground corncob or rice-hull carrier into a ribbon mixer operating at 60–80 rpm for 15–25 minutes after a pre-mix of the API and silicon dioxide has been screened through a 500 µm mesh.

The farm-level addition is calculated from the prescribed daily dose and species-specific feed intake; for an intermediate premix containing 50 g API per kilogram, addition of 4 kg premix per tonne of complete feed delivers 200 mg API per kilogram feed. During feed pelleting, the conditioning temperature should not exceed 85°C for more than 30–60 seconds unless stability data demonstrate marker recovery above 95% after conditioning. For thermolabile lots, post-pelleting spraying of a liquid premix is preferred, but the API must then be dissolved or suspended in an authorized vehicle and the spray system must maintain ≤5.0% coefficient of variation across the pellet flow. Carryover limits under EU Regulation 2019/4 require validated flush cycles and a documented worst-case residue carryover below the registration-set threshold, commonly measured by high-performance liquid chromatography in the first subsequent non-medicated batch.

Terminal finished product types include 0.5%, 1%, and 5% premixes in 25 kg paper bags with polyethylene liners, as well as pelleted medicated feed and top-dress powders. The route is appropriate only for species and production classes for which the marketing authorization contains an oral feed administration schedule; it is not appropriate for injectable or aqueous drinking-water formulations without separate stability and dosing validation.

When the API Is Reconstituted in Hard Water at pH 7.8–8.4

Oral solutions and drinking-water concentrates require a different stability strategy because the dilution water supplied on farm may have alkalinity, hardness, and chlorine residuals that alter marker solubility and accelerate precipitation. The finished oral liquid must meet USP <1231> water-for-pharmaceutical-purposes guidance for production water and, where the product is preserved, USP <341> antimicrobial effectiveness testing. The batch equation for a 100 mg/L stock solution at 97.5% purity and 3.0% LOD is 105.7 g API per 1000 L of solution; the API is first wetted with propylene glycol or a small volume of ethanol before high-shear dispersion at 1500–3000 rpm for 10–20 minutes, because direct addition into hard water of pH 7.8–8.4 can produce a colloidal haze that is difficult to clear by 1.0 µm filtration. The use of a chelating agent such as disodium edetate at 0.05–0.1% w/v and a citrate buffer system at pH 5.0–6.5 reduces hardness-induced flocculation.

Production equipment is typically a 316L stainless-steel jacketed vessel with a bottom-mounted rotor-stator mixer, an in-line 1.0 µm clarifier, and a 0.45 µm bioburden-reduction filter before filling. The filled solution is packaged in amber polyethylene terephthalate or high-density polyethylene bottles because light exposure below 420 nm accelerates color change and marker loss. A metering pump set at 1:100 dilution from the 100 mg/L stock delivers 1 mg/L in drinking water, assuming the farmer calibrates the pump against actual water flow; field audits frequently record pump-calibration drift of ±10%, so the formulated stock must tolerate a 10% overage without exceeding the acceptable daily intake margin. Terminal finished forms include oral drench solutions, 1–5 L drinking-water concentrate bottles, and pump-metered stock solutions. The route must not be packaged in clear glass unless the stability protocol explicitly demonstrates ≥95% marker recovery after 6 months under 40°C/75% RH in the proposed container.

Powder-filled hard capsules containing Ganfan Jiedu Tablets Veterinary Grade API are produced when the target species requires a taste-masked unit dose but swallowing a tablet is impractical, or when the finished product is authorized as a capsule in the registration dossier. The capsule fill mass must be controlled for blend uniformity under USP <905> and, where dissolution is required, USP <711>; the capsule shell itself is tested for disintegration under USP <701> unless a pharmacopoeial alternative is specified in the dossier. A size 0 hard capsule with 360 mg total fill weight and a 50 mg label claim at 98.0% purity and 4.5% LOD requires 53.4 mg API per capsule, or 14.8 wt% of the fill. The API is pre-blended with a silicated microcrystalline cellulose at 20–25°C and 40–50% RH for 15 minutes in a tumble blender; hydrophobic lubrication with magnesium stearate must be kept at 0.5–1.0% w/w and not exceed 5 minutes final blending time, because longer lubrication reduces dissolution rate and increases inter-capsule weight variability.

Filling is carried out on a dosator or tamping-pin capsule machine operating at 30–60% of maximum speed during the first production campaign; dosator machines require a pre-compressed plug of the API blend at 10–15 N plug compression force, while tamping pins require pin stations set to 2–3 tamping stages to achieve consistent fill weight without crushing the API granulate. The fill process is moisture-sensitive: if in-process fill mass exceeds 4.5% LOD, the powder can adhere to the dosator chamber and cause intermittent underweight capsules; if the fill is over-dried below 2.0% LOD, static charge can increase and produce weight variability above 2.5% relative standard deviation. Terminal finished product types include hard gelatin and hydroxypropyl methylcellulose capsules in bulk or blister packaging. The capsule route is incompatible with highly hygroscopic filler combinations above 5.0% moisture and with enteric coating operations that expose the capsule shell to aqueous coating at >40°C.

Wet-Granulation Binder Viscosity and the Granule Strength–Dissolution Trade-Off

Granule and sachet production for Ganfan Jiedu Tablets Veterinary Grade API is selected when the dose must be dispersed in water or sprinkled onto feed at the point of use. The granulated product must meet USP <711> dissolution or dispersion testing where applicable, and packaging integrity via ASTM D4169-22 distribution simulation. A model single-dose sachet with 1.0 g granule mass and a 250 mg label claim at 97.0% purity and 3.2% LOD requires 266.3 mg API per sachet, equivalent to 26.6 wt% of the final granule mass. The granulation binder choice controls the granule strength–dissolution trade-off: a 3–5% w/w aqueous polyvinylpyrrolidone K30 binder solution produces granules with a bulk tapped density of 0.55–0.70 g/mL and a median particle size of 350–600 µm, while higher binder viscosity reduces dissolution by forming a less porous matrix.

Fluid-bed top-spray granulation is preferred over high-shear granulation because the API is thermally sensitive, and fluid-bed processing supplies the binder solution at 8–12 g/min per nozzle with an inlet air temperature of 55–65°C and a product temperature set point of 35–45°C. The granulation endpoint is controlled by LOD at 2.5–3.5% and by end-point torque or power draw on the fluid-bed motor. The dried granulate is milled through an 850 µm screen, then blended with a citrus- or apple-based flavor carrier at 5–10% w/w before filling into triple-laminated sachets at 20–25°C and 30–40% RH. Terminal finished forms include single-dose and multi-dose sachets for oral suspension, feed top-dress granules, and bulk veterinary granules intended for reconstitution in the farm water line. Granule formulation is not recommended for automated feed mills that use high-intensity auger transfer, because friable granules below 0.5% attrition resistance may generate fines that segregate from the carrier and change the delivered dose; if the product is presented as a granular top-dress, the granule core must achieve a friability test result below 1.0% after 10 minutes in a rotating drum.

    More Introduction

Ganfan Jiedu Tablets Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions is released as a multi-formulation active pharmaceutical ingredient. The trade name retains the word “Tablets” because the reference development lot was qualified on a compressed tablet formulation; the article of commerce is not a finished tablet. The manufacturer’s model identifier GFJD-VET-API-11 covers seven dosage-form conversion routes: direct compression for tablets, dry powder blending for oral powders, capsule filling, wet granulation for granules, medicated premix carrier loading, aqueous solution compounding for drinking water, and injectable solution processing after membrane filtration. The identifier is a commercial control code, not a pharmacopoeial monograph designation.

The control basis is a purified extract fraction with a chromatographic marker profile quantified by HPLC. Because the material is intended for both non-sterile and sterile veterinary finished products, the release strategy applies injectable-grade tests—bacterial endotoxin, low water content, specified elemental impurities, and particulate matter after reconstitution—to a grade that also remains compatible with direct compression and premix dispersion. The material is dried in a vacuum tray dryer at chamber pressure below 10 kPa and jacket temperature not exceeding 55°C. Oral-grade water content is released at ≤5.0%; injection-grade material is dried to ≤3.0%. Packaging uses double low-density polyethylene liners inside high-density polyethylene drums under nitrogen overlay. Standard pack sizes are 5 kg, 10 kg, and 25 kg; the 25 kg drum is designated for premix and tablet campaigns, while 1 kg pharmaceutical-grade bottles are used for injectable-grade development lots. This documentation does not assign a clinical indication; the prescribing veterinarian determines the therapeutic use.

Which release specifications are applied across the seven dosage-form conversion routes?

The release specification is split into an oral/premix module and an injectable module. This prevents a batch released only for oral premix from being diverted, without additional testing, into an injectable production line. The oral module follows Ph. Eur. 5.1.4 for non-sterile microbial quality and USP 61/USP 62 for enumeration and specified organisms. The injectable module adds bacterial endotoxin control using Ph. Eur. 2.6.14, particulate matter evaluation by Ph. Eur. 2.9.19 or USP 788 after dissolution, and a reduced water threshold. Assay and related substance limits are harmonized across both modules to maintain batch homogeneity when a single lot is split between oral and injectable campaigns.

    Particle size D90 ≤150 µm ≤75 µm ISO 13320:2020

The injectable module does not establish sterility of the API powder. Sterility is achieved at the finished-product stage by filtration through a 0.22 µm membrane, usually polyethersulfone or polyvinylidene fluoride, followed by terminal sterilization if degradation kinetics permit. Because the dry powder is crystalline, particulate load is controlled by milling and sieving rather than by dry-powder filtration. Laser diffraction per ISO 13320:2020 reports D10, D50, and D90; the D90 limit for injectable grade is ≤75 µm only as a coarse upper bound, because dissolution rate depends on vehicle pH, ionic strength, and temperature. In aqueous compounding, the solution is passed through a 0.45 µm prefilter and a 0.22 µm bioburden-reduction filter. Filter adsorption screening is required before routine production; PVDF membranes are usually preferred over nylon because the active fraction can contain phenolic and carboxyl groups that bind to polyamide surfaces. Published data for this specific product configuration is limited, so filter compatibility should be confirmed in the actual vehicle at the intended concentration.

Particle-Size, Bulk Density, and Granulation Fluid Requirements

For dry dosage forms, the milled API is characterized by bulk density rather than by nominal mesh alone. Bulk and tapped density are determined per Ph. Eur. 2.9.34; the typical bulk density is 0.38 g/mL to 0.52 g/mL. This range supports die fill on a 16-station rotary tablet press at speeds up to 30 rpm when the formulation contains at least 30 wt% microcrystalline cellulose and 0.2–0.5 wt% colloidal silicon dioxide. Powder flow is assessed by Ph. Eur. 2.9.36; if the compressibility index exceeds 25%, direct compression is not recommended without granulation. For capsule filling, a D90 above 150 µm can produce weight variation above 3% relative standard deviation in low-fill-weight capsules. The API is therefore milled with a pin mill at rotor tip speed of 60 m/s; the mill is cooled to prevent thermal softening. After milling, the powder is passed through a 250 µm safety screen to remove agglomerates.

Wet granulation with aqueous binder is permitted when the binder solution is sprayed rather than poured. In a high-shear mixer with impeller speed 120 rpm and chopper speed 1500 rpm, the granulation endpoint is controlled by impeller torque, not by fixed batch time. This torque-based endpoint reduces batch-to-batch variation caused by variability in API moisture and particle size. Granules are dried in a fluid-bed dryer with inlet air temperature not exceeding 55°C; the thermal limit is assigned as a processing control to avoid discolouration and unintended changes in dissolution. Granules for sachets are sized through a 1.0 mm screen, while premix granules may be milled to a particle size compatible with feed-mix distribution.

When the Same API Is Switched from Direct Compression to Sterile Solution Manufacturing

The switch from an oral solid dosage form to an injectable solution requires more than a specification change. The compounding area must meet the GMP grade appropriate for the relevant regulatory framework, with final filtration in Grade A. Stainless-steel vessels with 316L contact surfaces and electropolished finish are preferred; non-polished surfaces can generate metal particulate in acidic vehicles. If the vessel is shared with oral products, cleaning validation must address cross-contamination of related herbal or alkaloidal residues because HPLC marker assay alone may not distinguish structurally related impurities. Solution hold time before filtration is limited to 4 h at 15–25°C unless a matrix-specific stability study supports longer hold. Nitrogen overlay is maintained to keep dissolved oxygen below 0.5 mg/L; otherwise the specified impurity at relative retention time 1.12 can increase. The solution should not be stored in ordinary glass bottles for prolonged periods if the pH is above 8.0, because alkaline conditions accelerate hydrolysis of the marker fraction. Injectable solution compounding should not use lactated Ringer’s or phosphate-buffered vehicles without preformulation data, because multivalent ions can reduce filter throughput and cause visible precipitation.

Comparing Against Oral-Only and Injectable-Only Veterinary API Grades

The multi-formulation grade differs from oral-only veterinary APIs by carrying bacterial endotoxin and low-water controls even when supplied for oral premix. It differs from injectable-only APIs by maintaining compatibility with direct compression and premix carrier loading without hydrophobic surface coatings or single-dose vial formatting. The oral-only grade may use a higher residual moisture because granulation and drying can tolerate it; however, high moisture reduces the physical stability of the API in bags and increases microbial risk. The injectable-only grade may be micronized to a smaller D90 for fast dissolution, but this can harm direct compression by increasing cohesion and reducing flow. The current multi-formulation grade is not optimized for either single route but is controlled at a midpoint that permits both routes under defined formulation conditions.

Table 2. Comparative control targets for multi-formulation, oral-only, and injectable-only veterinary API grades
Control dimension Multi-formulation grade Oral-only grade Injectable-only grade
    Water content ≤3.0% injection; ≤5.0% oral ≤6.0% ≤2.5%
Bacterial endotoxin ≤0.50 EU/mg injection not routinely tested ≤0.10 EU/mg
    Particle-size target D90 ≤75 µm injection; D90 ≤150 µm oral D90 ≤250 µm D90 ≤50 µm
    Microbial load TAMC ≤10³ CFU/g; TYMC ≤10² CFU/g TAMC ≤10⁴ CFU/g; TYMC ≤10² CFU/g TAMC ≤10² CFU/g; TYMC ≤10 CFU/g
Packaging double LDPE liner under nitrogen single LDPE liner double LDPE liner under nitrogen, plus overpouch for sterile transfer
Processing compatibility direct compression, capsule fill, wet granulation, premix, solution direct compression only aseptic solution only

These comparative values are typical control targets for technical evaluation; published pharmacopoeial limit data for this specific product configuration is limited, so release criteria should be confirmed against the current batch certificate.

During direct compression runs on a rotary tablet press, the main operational risk is punch film. If residual moisture exceeds 5.0% or the API fraction exceeds 5 wt%, film build-up on 10 mm concave punch faces reduces tablet hardness and weight uniformity. Forced-air dust extraction at the press and precompression force in the range 2–5 kN reduce build-up without changing disintegration. On a 16-station instrumented press, ejection force should be monitored; values above 350 N require adjustment of magnesium stearate level or mixing time to prevent capping.

Premix processing has its own boundary. In a horizontal ribbon mixer with working volume 500 L, blend uniformity requires sampling across at least 10 points including the discharge valve because API migration to the bottom of the mixer can occur if the carrier has a wide particle-size distribution. If the carrier has a mean particle size above 850 µm, the API does not adhere well and may segregate during transport. Published data for this specific configuration is limited, so pilot-scale transport simulation is recommended before commercial distribution. For injectable solutions, the critical control is filter compatibility and holding time; the final solution should be filtered within 4 h of preparation and protected from oxygen during filling.
