Application of Baiyu Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

Direct compression of the Baiyu Powder veterinary-grade API onto a 36-station rotary tablet press begins with a pre-blending sequence that reduces segregation risk in low-dose formulations. The API is first passed through a 500 µm stainless steel sieve and combined with a sieved diluent in a V-blender filled to 60% of gross volume. For tablets containing less than 5% w/w active, a three-stage geometric dilution is used: 1:1 API-to-microcrystalline cellulose, then 1:4, then addition of the remaining diluent. Blend uniformity is sampled from 10 positions and analyzed by HPLC with a target RSD of ≤5.0%, while finished tablet content uniformity is assessed by USP <905>. The press is run at 30–50 rpm with 3–6 kN precompression force and 8–18 kN main compression force; the resulting breaking force is held between 40 N and 80 N as measured by USP <1217>. If the Carr index of the final blend exceeds 35, die fill becomes non-uniform and tablet weight variation exceeds ±5%. Lubrication is limited to 0.5–1.0% w/w magnesium stearate; above this range the hydrophobic lubricant film delays disintegration beyond 15 min in water at 37°C per Ph. Eur. 2.9.1. Capping and lamination occur when the granulation or blend moisture is below 2% or above 4%; water content is measured by a halogen moisture analyzer at 105°C or by Karl Fischer titration per USP <921>. The finished veterinary tablets are packaged in HDPE bottles with a desiccant when the equilibrium moisture content is below 2%, and bulk hopper hold times are kept below 30 min when the active D90 exceeds 200–250 µm to avoid segregation.

When terminal sterilization is replaced by aseptic processing for injectable veterinary products

Injectable solution manufacturing with Baiyu Powder API begins with a compounding pH screen between 4.5 and 6.0. A terminal steam cycle at 121.1°C with an F₀ of not less than 8 minutes is the preferred sterilization approach and is validated under USP <1229>. When the active compound degrades under moist heat, the process is transferred to a Grade A isolator with hydrogen peroxide decontamination, qualified under ISO 13408-1:2008. The bulk solution is passed through a 0.22 µm PVDF or PES filter; the filter must pass bacterial retention according to ASTM F838-20. Pre-filtration bioburden is controlled below 10 CFU/100 mL to limit endotoxin accumulation. The sterile solution is filled into Type I glass vials closed with elastomeric closures tested per USP <381>. Finished product testing includes sterility per USP <71>, bacterial endotoxins per USP <85>, and subvisible particulate matter per USP <788>. The terminal solution is a ready-to-inject product with pH adjusted to the stability optimum and viscosity low enough for withdrawal through a 21-gauge needle.

For injectable suspensions, sterile filtration is not applicable. The API must be either pre-sterilized before aseptic wet milling or sterilized after milling in a dry heat cycle. Gamma irradiation at doses above 25 kGy can increase particle surface free energy and alter suspension flocculation behavior; therefore, the minimum validated dose under ISO 11137-2 is used. The aseptic bead mill is charged with 0.2–0.3 mm yttria-stabilized zirconia beads and operated at tip speeds that reduce D90 to 10–20 µm. The milled suspension is held at a viscosity below 40 mPa·s at 25°C as measured by USP <911> to maintain syringeability through a 21-gauge needle. A production bottleneck observed on aseptic suspension lines is the transfer of sterile API powder into the mixing vessel without breaking sterility; split-butterfly valves and alpha-beta transfer ports are used for this step. In-process hold time after sterile powder charging is kept within the validated bioburden recovery window, typically no more than 4 hours. The finished long-acting suspension is packaged in Type I vials under an inert headspace when the molecule is oxygen-sensitive.

On a 12-segment capsule filling line, the Baiyu Powder API is processed as a dust-controlled pre-blend. The API is first passed through a 300 µm sieve and mixed with microcrystalline cellulose in a 1:10 ratio to break up cohesive agglomerates. The main blend is then prepared in a bin blender with lactose monohydrate and croscarmellose sodium. The finished blend must show a Carr index below 30 and a Hausner ratio below 1.25, calculated from bulk and tapped densities per USP <616> and USP <1174>. Size 1 capsules are filled to a target fill weight of 300–450 mg; weight variation is checked per USP <905> and an acceptance value greater than 15 is out of specification. The dosator nozzle diameter is set between 8 mm and 10 mm and the tamping pin compression depth between 2 mm and 4 mm. Static charge becomes a critical defect source below 30% relative humidity, when the powder adheres to the dosing disc and causes weight drift. Above 60% relative humidity, hygroscopic fillers can soften gelatin shells. Dissolution testing is performed with Apparatus 2 in 900 mL of buffer matching the target species at 37°C per Ph. Eur. 2.9.3 or USP <711>; the usual release criterion is not less than 75% at 45 min. Filling speed is limited to approximately 50,000 capsules per hour if the flow function coefficient falls below 4, because bridging above the dosator chamber produces recurrent weight spikes. The filled capsules are sealed and packaged in PVC/PVDC blisters with a moisture barrier when the formulation is humidity-sensitive.

How Does Binder Viscosity Shift the Wet Granulation Endpoint for Veterinary Oral Granules?

High-shear wet granulation of the Baiyu Powder API is controlled by torque rather than fixed time. The API and intragranular excipients are loaded into a 65 L high-shear mixer running at an impeller speed of 150–200 rpm and a chopper speed of 1,500–2,500 rpm. Binder solution is prepared as povidone K30 at 5% w/w in purified water and sprayed at 2–4 kg/min. The endpoint is read from the impeller power-draw curve; a stable plateau indicates dense granule formation. A downward trend after the plateau signals overwetting and produces granules larger than 850 µm. The wet mass is discharged through an oscillating granulator fitted with a 1.0 mm screen and transferred to a fluid-bed dryer. Inlet air is set at 60–70°C; drying continues until the final loss on drying is 1–2% w/w by USP <921>. Dried granules are milled and sieved to a D50 of 150–300 µm with a D10 above 75 µm as measured by laser diffraction per USP <429>. The final granule blend is filled into sachets on a form-fill-seal line; fill weight variation is maintained within ±5%. Granule friability is checked on a Pharmatron tester at 100 rotations; values above 1% indicate excessive dust generation in the filling area. Sachet content uniformity is tested by Ph. Eur. 2.9.40. If granule porosity falls below 10%, disintegration in water at 37°C can exceed 15 min. The finished veterinary oral granules are packaged in foil-laminate sachets to protect against moisture and light.

In a medicated feed mill, the Baiyu Powder veterinary-grade API is diluted into a feed premix using a 1,000–2,000 L ribbon or paddle mixer. The carrier is selected from rice hulls, corn cob fraction, or calcium carbonate with a particle size between 20 mesh and 40 mesh. Mixing time is fixed at 10–15 min after a tracer salt study confirms mixer uniformity. For final feed concentrations below 0.5% w/w, at least two sequential dilution steps are used; each step is blended in a separate mixer or after complete discharge to avoid hot spots. The mixer is not filled beyond 70% of gross volume because overfilling creates dead zones. The finished premix is sampled from 10 discharge points and assayed by LC-MS/MS with a method validated under VICH GL49. Mixer uniformity is acceptable when the active assay RSD is ≤5%. Carryover is managed according to FDA 21 CFR 558.4 and FDA 21 CFR 225.1; the mixer is flushed with corn meal or soybean meal until the active residue falls below the carryover limit. Compliance for EU markets follows EU Regulation 2019/4. Cross-contamination risk increases when the active particle size differs from the carrier by more than 300 µm. Segregation is evaluated by sampling the upper and lower zones of the final feed bin; if the assay difference exceeds 10%, the batch is held and re-mixed. The finished premix is packed into 25 kg multiwall paper bags with an inner polyethylene liner, and vibration during bulk transport can still stratify the mixture.

Drinking Water Soluble Powder and Hard Water Complexation in Proportioner Systems

The Baiyu Powder API is formulated as a drinking water soluble powder only after solubility screening in the target proportioner stock solution. A common in-line proportioner setting is 1:128; therefore the stock solution must be 128 times more concentrated than the final drinking water concentration. If the final water concentration is 50 mg/L, the stock solution must hold 6.4 g/L active at 5°C. Source water hardness is measured by ASTM D1126-17; hardness above 200 mg/L CaCO₃ can slow dissolution or precipitate the active if it is a weak acid. Solubility is mapped from pH 4.5 to 8.0 before buffer selection. The dry powder is blended with dextrose or lactose in a ribbon mixer and passed through a 100 µm in-line screen before packaging into foil-lined sachets. At the farm, the reconstituted solution is filtered through a 50–100 µm mesh filter at the proportioner intake; pressure drop across the strainer is monitored for plugging. Residual free chlorine above 2 mg/L can degrade oxidation-sensitive APIs and is measured by the DPD method. Published data for the Baiyu Powder API under all farm water conditions is limited, so a site-specific 24-hour in-use stability study is required before bulk use. Bicarbonate alkalinity above 300 mg/L may require a chelating agent, but the chelator must be compatible with both the API and the target species. The terminal product is a clear or lightly opalescent medicated drinking water prepared at the point of use.

    Certification & Compliance
    More Introduction

The product designated Baiyu Powder is a veterinary-grade active pharmaceutical ingredient powder supplied for formulation into tablets, injections, capsules, powders, granules, premix, and solutions. The material is released as a crystalline or spray-dried powder depending on the downstream dosage form, with a representative assay range of 98.0–102.0% on the dried basis by high-performance liquid chromatography against the relevant veterinary monograph. The specification distinguishes between injectable and oral grades primarily by endotoxin load, bioburden, particle size, and residual solvent profile. For injectable preparation, the powder is milled to a target d90 of ≤150 µm, with a d50 controlled between 25–75 µm by laser diffraction according to ISO 13320:2020. For direct-compression tablets and capsules, the d50 is adjusted to 75–150 µm with a Hausner ratio 1.25–1.45 to maintain flow. Loss on drying is controlled to ≤0.5% by Ph. Eur. 2.2.32. Tap density, angle of repose, and Carr index are reported on each certificate of analysis because these parameters influence die-fill uniformity and content uniformity in low-dose solid dose forms.

    Regulatory Baseline and Product Identification

Baiyu Powder is positioned as a multi-dosage-form veterinary API with batch release aligned to Ph. Eur. or USP general chapters where a monograph exists, and to ICH Q3C for residual solvents, ICH Q3D for elemental impurities, and ICH Q6A for specification setting. Identification is confirmed by infrared absorption spectrophotometry and retention-time matching against a reference standard; the infrared spectrum is recorded by the attenuated total reflectance technique under Ph. Eur. 2.2.24. The current specification carries the model designation Baiyu Powder as the trade name and does not assign a separate pharmacopoeial name beyond the active moiety described in the destination-registration file. Batch records require the supplier to state whether the powder is micronized, spray-dried, or crystalline-milled because this affects reconstitution time in solution formulations and blending time in low-dose premixes.

For tablet and capsule manufacturing, the Baiyu Powder grade is dry-blended with direct-compression excipients after passing through a 500 µm sieve to eliminate agglomerates. Content uniformity risk is greatest in formulations with active mass ≤10 mg per unit; in such cases, a geometric pre-blend with lactose monohydrate or microcrystalline cellulose is performed for 10–15 minutes in a bin blender at 60–70% vessel fill. Blend uniformity acceptance follows FDA 21 CFR 211.110 with relative standard deviation ≤5.0% for the first three validation batches. Tablet compression on a rotary press at 40–80 rpm uses compression force 8–18 kN to achieve target hardness 60–120 N depending on the formulation. Capsule filling uses dosator or tamping-pin machines; the powder should be pre-conditioned to 20–25 °C and 40–50% RH because higher humidity raises the moisture content above 0.5% and reduces flow. When the blend contains hygroscopic excipients, drying of the API is required at 60 °C for 2–4 hours under vacuum if the loss on drying exceeds 0.5%. These parameters are not predictive for all formulations; process validation batches must establish final limits for each dosage form.

What Limits the Use of Baiyu Powder in Prefilled Veterinary Syringe Solutions?

Injectable solution preparation requires that the Baiyu Powder injectable grade meet bacterial endotoxin limits of <0.50 EU/mg by Ph. Eur. 2.6.14, and for large-volume parenterals, the formulated solution should comply with Ph. Eur. 0520 or USP <1>/<85> according to the target market. The powder is dissolved under aseptic conditions in Water for Injection at 15–25 °C with low-shear mixing; if the active moiety has pH-dependent solubility, adjustment with dilute hydrochloric acid or sodium hydroxide is performed within the pH range specified in the product-specific formulation file. Terminal sterilisation by autoclaving at 121 °C for 15 min is used only when the active moiety is thermally stable; otherwise, sterilising-grade filtration through a 0.22 µm membrane is required. Particulate contamination is controlled by the light obscuration method under Ph. Eur. 2.9.19 to ≥10 µm: ≤6000 particles/container and ≥25 µm: ≤600 particles/container for small-volume injectables. The injectable grade of Baiyu Powder should not be blended with amine-based preservatives unless compatibility data confirm no adduct formation; published data for this specific configuration is limited, so forced degradation studies are required before registration.

Premix and granule manufacture uses the Baiyu Powder oral grade with larger d50 75–150 µm to reduce dusting. Mixing with feed carriers at active concentrations below 1% requires a stepwise dilution process because the potency gradient becomes a critical control point. A low-shear ribbon blender with L/D 1.5–2.0 is operated at 15–25 rpm for 10–20 minutes; mixer validation shows that extending mixing beyond 30 minutes can induce electrostatic segregation. Granulation by high-shear wet granulation uses an aqueous binder at 3–5% by weight and final moisture 1.5–3.0% before drying. The resulting granules are sized through 850 µm and 250 µm screens to control the particle-size distribution. For solution and soluble-powder formulations, the API is dissolved with stirring at 20–30 °C, and clarity is evaluated according to Ph. Eur. 2.2.1 or 2.2.2. Solutions should be protected from light when photostability data indicate degradation.

When Preblending with Acidic Excipients Reduces Assay Recovery

Compatibility screening is required for Baiyu Powder when the formulation includes acidulants such as citric acid or malic acid because localized pH reduction can promote degradation in the solid state. In a binary blend containing 5% citric acid and 5% API, accelerated stability at 40 °C/75% RH for 4 weeks may show assay loss if the active moiety is acid-labile; the threshold for acceptable loss is ≤2.0% relative to initial assay. If this threshold is exceeded, the formulation is reformulated with alkaline buffers or the acidulant is coated. The powder should not be co-milled with oxidizing agents; peroxide residues above 10 ppm in crospovidone or other excipients have been associated with degradation in structurally sensitive actives. Where published data for the specific active moiety in Baiyu Powder is limited, a binary compatibility matrix under ICH Q1A(R2) stress conditions is generated with drug-to-excipient ratios of 1:1 and 1:10 at 40 °C/75% RH and 25 °C/60% RH for 14 days. The specification includes a related-substances limit of single ≤0.5% and total ≤2.0% by HPLC, with each unknown impurity reported if area percentage exceeds 0.10%.

Particle-Size-Dependent Content Uniformity in Low-Dose Tablet Cores

Potency uniformity for tablet cores containing 10 mg or less of Baiyu Powder is influenced by the D90 and the number of particles per unit dose. Particle-size reduction from d50 120 µm to d50 45 µm increases particle count per 10 mg dose by roughly an order of magnitude if density remains constant. The milled grade with d90 ≤150 µm and d50 45–75 µm is therefore specified for low-dose tablets. Sieve analysis according to Ph. Eur. 2.9.12 or laser diffraction under ISO 13320:2020 is performed on each batch. The acceptable content uniformity is AV ≤15 by Ph. Eur. 2.9.40 for single-dose tablets unless the target market applies USP <905> acceptance values. Blend segregation is assessed by sampling 10 locations after 5, 10, and 15 minutes of mixing; a relative standard deviation above 5% at the 15-minute time point indicates the need for a finer particle size or a surfactant wetting agent. Direct compression is preferred over dry granulation when the API is cohesive, because dry granulation increases fines and may reduce bulk density below 0.35 g/mL. If the bulk density is below 0.30 g/mL, capsule filling weight variation exceeds ±5% on high-speed machines.

Solution reconstitution and injectable blending of Baiyu Powder require control of dissolved oxygen. The powder is dispensed under nitrogen blanketing in the manufacturing vessel if the active moiety contains oxidation-susceptible functional groups; the dissolved oxygen level is maintained below 0.5 mg/L in the final solution by sparging with pharmaceutical-grade nitrogen for 15–20 minutes at a flow rate that does not cause foaming. Buffering capacity is established through preformulation screening with phosphate, citrate, or acetate buffers at 10–50 mM ionic strength. For solution products intended for oral drench or drinking-water administration, hardness of the dilution water should be assayed because calcium and magnesium ions can reduce solubility or form complexes if the active moiety is a weak acid salt. The oral solution is prepared at 1–10 mg/mL active concentration, and pH is adjusted to 4.0–7.0 unless species-specific acceptance criteria require otherwise. The powder should be stored in tight containers at 15–25 °C and protected from humidity above 60% RH; if the container is opened at relative humidity above 60%, drying before use may be required because the powder can adsorb moisture and shift loss-on-drying beyond 0.5%.

Is the Same Lot Acceptable for Granule, Premix, and Injectable Production?

The same lot of Baiyu Powder is not automatically acceptable for all seven dosage forms. Injectable use requires endotoxin testing <0.50 EU/mg, sub-visible particle control, and the lower d90 ≤150 µm. Oral powder and premix use can accept a higher endotoxin limit ≤2.5 EU/mg and larger d50 75–150 µm. If a single lot is intended for cross-formulation use, it must meet the tightest specification across all intended routes: endotoxins <0.50 EU/mg, d90 ≤150 µm, total microbial load ≤102 CFU/g, and loss on drying ≤0.5%. However, milled injectable-grade powder may produce greater dust and poorer flow in direct-compression tablet formulations compared with the oral-grade d50 range. Conversely, the oral-grade larger particles may produce slow dissolution and low assay in injectable compounding. Therefore, the specification file assigns grade-specific particle-size ranges rather than a single universal distribution. For oral granules and premixes, the powder may be blended with mineral oil or propylene glycol as a dust suppressant at 0.5–2.0% by weight, but such treated lots are unsuitable for injection.

Batch-to-batch variability is monitored through process analytical technology where available. Near-infrared spectroscopy is used for blend uniformity in tablet production with a spectral range 1,600–1,800 nm; the D2 distance limit is set at 3 for conforming blends. The supplier’s release data for Baiyu Powder includes the polymorphic form by X-ray powder diffraction because changes in crystalline form alter solubility and bioavailability. If amorphous content exceeds 5%, dissolution may increase but physical stability may decline under humid storage. The powder should be protected from direct light; photostability testing under ICH Q1B conditions with near-UV radiation ≥200 Wh/m² and visible light ≥1.2 million lux·h determines if amber packaging is required. When used in capsules, the moisture content of the empty capsule shell is controlled to 13–16%; if the filled powder has loss on drying >2.0%, the shell may become brittle or soften. The encapsulation process is therefore run at 20–25 °C and 40–45% RH. After filling, the capsules are sealed or banded if the formulation is hygroscopic.

When Feed Premix Homogeneity Fails at the Farm Blending Stage

Premix formulations containing Baiyu Powder are intended for dilution into complete feed at inclusion rates from 0.1% to 1.0% depending on species pharmacokinetics and regulatory approval. Homogeneity tests follow ISO 6497:2002 for animal feeding stuffs. A coefficient of variation ≤10% is required at the farm blending stage, and ≤5% at the manufacturing stage. If the API is added directly to ground corn without stepwise pre-blending, segregation may produce a coefficient of variation above 15%. The powder should be mixed into a mineral or lactose carrier at 5–10% drug-to-carrier ratio before final feed dilution. Species-specific differences in gastrointestinal pH and transit time affect the choice of particle size: for monogastric animals, finer particles with d50 45–75 µm are used to accelerate dissolution in the stomach; for ruminants, larger granules or protected particles may be required to survive rumen degradation if the active moiety is not rumen-stable. The premix grade should not contain reducing sugars in the carrier if the active moiety contains an amine group; Maillard-type adducts may form during storage at temperatures above 30 °C. When this incompatibility is suspected, the carrier is changed to calcium carbonate or microcrystalline cellulose.

Attribute Baiyu Powder veterinary grade Technical-grade powder
Endotoxin control <0.50 EU/mg injectable; ≤2.5 EU/mg oral not routinely controlled; may exceed 10 EU/mg
    Particle-size distribution d90 ≤150 µm injectable; d50 75–150 µm oral irregular, d90 commonly >300 µm
Residual solvent profile ICH Q3C Class 3 total ≤0.5% may contain Class 2 solvents above pharmacopoeial limits
Microbial limits TAMC ≤103 CFU/g; E. coli absent not validated for oral or parenteral use
Documentation Batch certificate with full release data, stability summary, residual solvent and elemental impurity profiles limited certificate of analysis
Application range Tablets, injections, capsules, powders, granules, premix, solutions typically limited to non-pharmaceutical technical use

Injectable compounding of Baiyu Powder in borosilicate glass vials or cyclic olefin copolymer syringes requires compatibility screening with the primary packaging. Leachable compounds from rubber stoppers, typically alkyl phenols and siloxanes, are monitored by HPLC-UV/MS; the acceptance threshold is ≤0.05 mg/day for any identified leachable unless lower limits apply in the registration dossier. Terminal sterilization and aseptic filling lines should maintain Grade A environment according to EU GMP Annex 1; the fill line velocity is validated to achieve a contamination rate <1 CFU per 10,000 units. For lyophilized vials, the frozen solution is lyophilized with primary drying at -20 to -10 °C shelf temperature and chamber pressure 0.05–0.15 mbar, followed by secondary drying up to 25–35 °C. The final moisture limit for lyophilized cake is ≤2.0%. Reconstitution time for injectable Baiyu Powder is specified at <60 seconds with manual swirling after addition of Water for Injection. If reconstitution exceeds this, particle-size reduction or a lyoprotectant is required.
