Application of Bailong Cangzhu Powder Veterinary Grade API for Tablets / Injections / Capsules / Powders / Granules / Premix / Solutions

In rotary tablet presses handling Bailong Cangzhu Powder, the first process conflict occurs at the pre-blend stage because residual moisture from the polysaccharide-rich rhizome fraction can exceed 5.0 wt% after 8 h of ambient exposure above 60% RH. Incoming powder is characterized for loss on drying by USP <731>, particle size distribution by USP <786>, bulk and tapped density by USP <616>, heavy metals by USP <233>/<232>, and microbial limits by USP <61>/<62>. Pre-drying in a fluid-bed dryer with inlet air at 50–60 °C to a final loss-on-drying value below 4.0% is required before direct compression or dry granulation. Batches that enter compression above 4.5% moisture typically develop picking and sticking on flat-faced punches at press speeds above 40 rpm. Air-classifier milling at 6000–8000 rpm with a classifier frequency of 50–70 Hz reduces the D90 to 150–180 µm, but the mechanical action increases electrostatic charging of dried polysaccharide surfaces and may raise the fraction below 45 µm to more than 30% if the classifier is not balanced.

Wet granulation is performed in a top-driven high-shear mixer with an impeller speed of 300–500 rpm and a chopper speed of 1500–3000 rpm. Aqueous starch paste at 2–5 wt% or povidone K30 at 3–5 wt% in a 1:1 water/ethanol mixture is sprayed at 5–10 g/min per kg of dry blend. Endpoint is determined by amperage draw, granule size distribution, and visual growth rather than a fixed time; overwetting beyond 12% granule moisture produces dense granules with a bimodal size distribution and a lower dissolution rate. The wet mass is passed through a 1.5–2.0 mm screen and dried in a tray dryer or fluid bed at 45–55 °C to a final moisture of 2.5–4.0%. If a solvent-free process is required, dry granulation by roller compaction at 15–25 kN/cm roll force is feasible, but re-milling often yields fines above 25% and reduces compressibility unless a pre-compression step is added.

Compression formulations for veterinary tablets typically contain microcrystalline cellulose at 30–50 wt%, crospovidone at 2–5 wt%, and magnesium stearate at 0.5–1.0%. Tablet hardness is maintained between 60 and 90 N measured by USP <1217>; friability must remain below 1.0% after 100 rotations in a USP <1216> friabilator. Disintegration follows USP <701> for uncoated veterinary tablets, with a target of < 15 min in water at 37 ± 2 °C. Dissolution acceptance is based on USP <711> Apparatus II at 50 rpm in 900 mL of 0.1 N HCl or pH 6.8 phosphate buffer, with Q set at 80% released in 60 min. However, published data for this specific API configuration is limited, so the dissolution specification should be prospectively validated on three production-scale lots.

Tablet coating with hydroxypropyl methylcellulose at 5–10 cP viscosity is performed in a perforated pan coater at an inlet air temperature of 60–70 °C, exhaust temperature of 38–42 °C, and pan speed of 8–12 rpm. Atomization air pressure of 1.5–2.5 bar and spray rate of 20–40 g/min per kg of tablets prevent overwetting of the water-sensitive core. Incompatibility is observed with strong alkaline excipients and with croscarmellose sodium above 5% because the resulting pH shift accelerates degradation of ester-linked volatile fractions. Final dosage forms are uncoated or film-coated veterinary tablets packaged in PVC/aluminum blisters with silica gel desiccant sachets.

What Limits Sterile Filtration Throughput for a Polysaccharide-Containing Veterinary API?

Bailong Cangzhu Powder cannot be injected as a direct suspension because lignified cell fragments and high-molecular-weight polysaccharides present a particulate load that fails USP <788> limits unless a validated clarification train is used. Aqueous extraction of the soluble fraction is performed with Water for Injection at 70–80 °C for 2–4 h, followed by centrifugation at 4000–6000 rpm and depth filtration through a cellulose-polypropylene composite rated at 0.45 µm. The resulting solution is passed through a 0.22 µm polyethersulfone membrane; however, polysaccharide concentrations above 1.0 mg/mL reduce throughput to < 10 L/m²/h at 1.0 bar differential pressure. Filter train validation uses 47 mm disc filters and ASTM F838-20 bubble point testing before scale-up to 10-inch cartridges. If flux decline exceeds 30% after 50 L/m², a second depth prefilter or an additional centrifugation step is introduced.

Endotoxin control is a critical path item because root powders frequently carry Gram-negative bacterial residues. The formulated solution must meet USP <85> criteria based on the maximum dose per kg body weight; for a veterinary injectable administered at 1 mL/kg, an internal limit of 5 EU/mL or lower is common depending on target species and maximum dose. Depyrogenation of glass vials at 250 °C for 60 min per USP <1228.1> is required. The solution is adjusted to pH 5.5–7.0 with dilute hydrochloric acid or sodium hydroxide, and sodium chloride is added to 0.9% for isotonicity. Residual ethanol from any extraction step must remain below 5000 ppm as a Class 3 solvent under USP <467>; if methanol is present, the limit is 3000 ppm per VICH GL18 guidance.

Solubility of hydrophobic sesquiterpene constituents is improved with propylene glycol at 10–30% or polysorbate 80 at 0.1–0.5%, but polysorbate above 0.5% may destabilize membrane filters by reducing surface tension below the wettability threshold. Terminal sterilization at 121 °C for 15 min is accepted only after forced degradation studies show assay loss below 5%; otherwise aseptic filtration through a 0.22 µm membrane is used. The sealed vials are inspected for particulate matter against USP <788> limits of ≤6000 particles per container at ≥10 µm and ≤600 particles per container at ≥25 µm. The final product is a clear to slightly opalescent solution in 10 mL or 50 mL amber borosilicate vials.

Capsule filling lines with dosator or tamping-pin units face a different failure mode: flow variability caused by the high aspect ratio of milled root particles and the electrostatic charging of dried polysaccharide surfaces. A pre-blend of Bailong Cangzhu Powder with colloidal silicon dioxide at 0.5–1.5 wt% and talc at 1–3 wt% is passed through a 0.5 mm screen to dissipate charge and break loose agglomerates. Bulk density and tapped density are measured by USP <616>; a Hausner ratio above 1.35 indicates the need for roller compaction or wet granulation before encapsulation. Powder flow through a 10 mm funnel orifice below 10 g/s is associated with weight variation outside ±5% at high machine speeds. Capsule fill weight acceptance is governed by USP <905>, with an acceptance value ≤15 and weight variation ≤5% for powder-filled capsules above 300 mg fill weight.

On a tamping-pin capsule machine operating at 30,000–60,000 capsules/h, the powder bed height should be maintained between 20 and 30 mm to prevent density segregation. The tamping force is set to produce a plug density of 0.7–0.8 g/mL; higher forces increase dust generation and can crush brittle plant cell fragments, releasing hygroscopic cores that later cause gelatin shell brittleness. Capsule shell selection matters: gelatin capsules lose flexibility when equilibrium moisture falls below 10%, while HPMC capsules withstand 2–5% moisture without cracking. If the powder blend contains volatile oil fractions above 0.5%, HPMC capsules or gelatin capsules with a moisture-barrier packaging film are preferred. Electrostatic charge above 50 nC/g measured by a Faraday cup is controlled by humidifying the filling room to 50–60% RH or adding purified water at 0.25% to the blend.

Finished veterinary capsules are typically size 0 or size 1 for swine and poultry oral administration; for larger ruminants, a size 000 capsule may be used but is poorly accepted under field conditions. The filled capsules are polished with sodium chloride in a capsule polishing machine and metal-checked with a tunnel metal detector at Fe > 1.0 mm and non-Fe > 1.5 mm sensitivity limits. Storage in sealed HDPE bottles with desiccant canisters is required when the API is dried below 4% moisture; above 60% RH during bottling, moisture regain exceeds 1.0% within 2 h.

Feed-Safety Sampling Math Is Different for Herbal Premix Uniformity

Premix production with Bailong Cangzhu Powder is governed by EU Regulation 183/2005 and, where applicable, FDA 21 CFR 225 current good manufacturing practice for medicated feeds. Direct addition of the powder to a final feed mixture at 0.1% inclusion fails to achieve uniformity because the API has a higher bulk density than carrier grains and migrates downward during pneumatic conveying. The standard corrective procedure is sequential dilution: a 1:10 pre-blend with ground rice hull or corn cob carrier is mixed for 10 min in a ribbon mixer, followed by a second 1:10 dilution and a final 1:10 dilution to reach the target concentration. Each stage uses a different mixer configuration; the final stage is completed in a double-ribbon mixer with a working volume of 100–200 kg and a fill level of 55–70% to provide adequate shear without dead zones.

Dilution stageAPI massCarrier massTarget concentrationMixer typeMixing timeCV acceptanceStandard
    Stage 110 kg90 kg10%ribbon mixer10 min≤5%ISO 6497
    Stage 210 kg of stage 190 kg1%ribbon mixer10–15 min≤5%ISO 6497
    Stage 310 kg of stage 290 kg0.1%double-ribbon mixer15 min≤7%ISO 6497

Sampling must follow ISO 6497 using at least 10 incremental samples from a 100 kg batch, with a coefficient of variation below 5% for the active marker. Carryover is a recognized problem because polysaccharide-rich powder adheres to mixer shafts, screw conveyor flights, and dust collection bags; a flush sequence with 10 kg of ground rice hulls after each campaign reduces cross-contamination to < 0.1% of the previous batch. The final premix is filled into 20 kg paper bags with inner polyethylene liners and stored below 30 °C at < 60% RH; under these conditions, moisture gain is less than 0.5% over 90 days. Avoid combination with choline chloride or strongly acidic mineral premixes unless the free moisture content is below 4%, because local pH depression and water activity increase accelerate degradation of the volatile oil fraction.

Fluid-bed granulation of Bailong Cangzhu Powder requires a narrower spray-rate window than typical cellulose-based granules because soluble polysaccharides can form surface crusts when spray rate exceeds the bed evaporation capacity. A top-spray granulator with a 1.5–2.5 mm nozzle tip and atomization air pressure of 2.0–3.0 bar operates with inlet air at 60–70 °C, product temperature at 35–45 °C, and spray rate of 10–25 g/min per kg of starting powder. The binder is povidone K30 at 3–5 wt% or hydroxypropyl cellulose at 2–4 wt% in purified water. The endpoint is determined by the product temperature increase and the disappearance of fine particles below 150 µm; over-granulation leads to a D50 exceeding 850 µm and a bulk density below 0.35 g/mL. In-line NIR moisture analyzers calibrated with R² > 0.98 against USP <731> detect granule moisture excursions within 0.5%, reducing batch rejection caused by surface crusting.

After granulation, the product is dried in the same fluid bed at 50–60 °C until loss on drying by USP <731> is < 5.0%. The final granules are sieved through 850 µm and 250 µm screens; acceptable fraction is between 250 and 850 µm, with fines below 150 µm not exceeding 15%. Flowability is assessed by USP <1174>; a flow rate through a 10 mm funnel orifice below 10 g/s indicates residual surface moisture or electrostatic bridging, requiring an additional drying pass or the addition of colloidal silicon dioxide at 0.25–0.5%. Granules intended for drinking-water application are tested for dispersibility by adding 20 g to 1 L of water at 25 °C; complete dispersion through a 250 µm mesh must occur within 3 min with less than 5% residue.

Microbial quality of the granulated product is tested according to USP <61> and USP <62>, with total aerobic microbial count below 10³ CFU/g and yeast and mold below 10² CFU/g. The final package is an aluminum-foil laminate sachet to prevent moisture ingress and volatile oil loss. Granule size distribution, moisture, and microbial limits are the primary release attributes; dissolution testing is not used for this dosage form because the product is administered as a suspension or top-dress rather than a compacted solid.

When Bailong Cangzhu Powder Is Dispersed into Oral Solutions, pH and Preservative Partitioning Become the Primary Constraint

Dispersion of Bailong Cangzhu Powder into oral solutions creates two simultaneous stability pressures: rapid sedimentation of insoluble cellulosic fractions and pH-dependent hydrolysis of polysaccharide side chains. Xanthan gum at 0.2–0.5% or a combination of microcrystalline cellulose and sodium carboxymethylcellulose at 0.5–1.5% produces a yield stress of 1.0–3.0 Pa that retards sedimentation. The suspension is prepared by first hydrating the suspending agent in purified water for 30–60 min under high-shear mixing at 3000–5000 rpm, then adding the API through a 0.5 mm sieve to eliminate agglomerates. The API is micro-milled to D90 50–100 µm before dispersion; if D90 exceeds 250 µm, sedimentation rate increases above 0.5 mm/min and resuspension becomes difficult after 24 h storage. The target pH is 4.5–6.5; at pH below 3.5, acid hydrolysis of polysaccharide side chains reduces viscosity, while at pH above 8.0, ester-linked volatile fractions degrade rapidly under accelerated storage conditions.

Preservative selection is not trivial because botanical sediment can adsorb sodium benzoate and potassium sorbate, lowering the aqueous concentration below the effective minimum inhibitory concentration. A use level of 0.1–0.2% potassium sorbate plus 0.05–0.1% sodium benzoate is typical, but free preservative content must be verified by HPLC under USP <51> antimicrobial effectiveness testing at 7, 14, and 28 days. The final solution is packaged in amber polyethylene terephthalate bottles with induction-sealed caps because oxygen permeation above 0.5 cm³/m²·24 h·atm accelerates oxidation of the volatile fraction. During long storage, the sediment volume ratio should remain below 0.2; sediment that becomes difficult to redisperse after 30 s of manual shaking indicates a need to reformulate with a higher yield stress suspending agent.

The dispersed oral solution is filled at a temperature below 30 °C to prevent thermal gelation of the suspending polymer; fill volume tolerance is ±2% for a 1 L bottle. Homogeneity before filling is checked by drawing samples from the top, middle, and bottom of the holding tank; assay variance across sampling points must be ≤3% RSD before the line starts. The finished product is a ready-to-use oral drench or drinking-water concentrate for calves, swine, and poultry; published data for this specific API concentration in long-term veterinary field use is limited, so the formulation should be bracketed by an accelerated stability study at 40 °C/75% RH for 6 months.

    Certification & Compliance
    More Introduction

Bailong Cangzhu Powder is released as a veterinary-grade active pharmaceutical ingredient for direct incorporation into tablets, injections, capsules, powders, granules, premixes, and solutions. The manufacturer assigns model designations that distinguish the standard non-sterile powder, the controlled-sieve fine-powder grade, and the low-endotoxin spray-dried grade; each model code is printed on the batch certificate of analysis and is linked to a defined particle-size specification. The material is not a formulated dosage form and requires downstream processing before administration. Published data for this specific configuration is limited; therefore, batch-release acceptance criteria should be based on the current Chinese Veterinary Pharmacopoeia monograph where available or on a buyer-approved specification sheet derived from the receiving market’s veterinary drug registration file.

What Pharmacopoeial Limits Govern Oral and Premix Applications?

For tablet, capsule, powder, granule, and premix use, the material is controlled against the general monograph for herbal powders in the current Chinese Veterinary Pharmacopoeia and, where imported, against USP <561> for articles of botanical origin. Identity is confirmed by high-performance liquid chromatography fingerprinting against a reference Cangzhu extract; the chromatogram must show the retention time and peak area ratio of the atractylodin marker within the monograph-specified window. Loss on drying for the standard oral grade is commonly set at ≤5.0% by USP <921> Method Ia, although the limit may be tightened to ≤3.0% for capsule formulations susceptible to cross-linking. Total ash is controlled at ≤10.0%, acid-insoluble ash at ≤2.0%, and heavy metals by ICP-MS with lead ≤5 ppm, cadmium ≤1 ppm, arsenic ≤2 ppm, and mercury ≤0.1 ppm for multi-species products. These are typical acceptance values for veterinary botanical APIs; they must be verified against the registered specification because regional pharmacopoeias adopt different heavy metal ceilings.

If the powder is intended for sterile compounding, the low-endotoxin grade is required and the oral microbial limits do not apply. Before terminal sterilization, the material is controlled for bioburden and, after reconstitution, for bacterial endotoxins. Microbial quality of the non-sterile grades is evaluated by USP <61> and USP <62>; total aerobic microbial count is typically 10³ CFU/g or less, with combined yeast and mould count 10² CFU/g or less and absence of Salmonella and Escherichia coli in 10 g samples.

When the Powder Is Used in Aqueous Injectable Formulations

The low-endotoxin spray-dried model is the only acceptable starting material for parenteral solutions, emulsions, or suspensions intended for injection. Bacterial endotoxin limits are species-specific and route-specific; a limit of 5 EU/kg body weight/hour for large-animal intravenous products is common but must be calculated from the registered dose. Solutions are compounded under ISO 7 controlled conditions and filtered through a 0.45 µm prefilter followed by a 0.22 µm sterilizing membrane. Sterility is confirmed by membrane filtration according to USP <71>. The powder contains water-soluble polysaccharides that produce a slight haze at neutral pH; turbidity should be measured against a ≤10 NTU release limit for injectable solutions unless otherwise justified. A pH shift below 3.0 or addition of polyvalent metal ions can precipitate polyphenolic constituents; untreated water with high calcium hardness should be avoided. If a suspension injection is developed, particle-size reduction to D90 ≤ 10 µm is required to reduce syringe needle occlusion and local irritation. The terminal sterilization cycle should be chosen after thermal mapping; saturated steam at 121°C for 15 min may degrade heat-labile sesquiterpenoids, so an alternative dry heat or radiation sterilization cycle should be justified with assay recovery data.

On a high-speed rotary press, direct compression of the standard 80-mesh grade is acceptable only when the formulation contains at least 20–30% microcrystalline cellulose or dicalcium phosphate diluent; otherwise die fill variation exceeds weight uniformity limits because the powder bulk density is low and flow is cohesive. The fine-powder grade is processed by high-shear wet granulation, not direct compression, because its high surface area causes sticking to punches at compression forces above 25 kN. Granule moisture content should be maintained between 1.5% and 2.5% at the end of drying; over-drying below 1.0% generates fines and increases capping, while moisture above 3.0% promotes picking during extended runs. Tablet hardness is monitored as a process indicator; a typical target range of 60–100 N for veterinary oral tablets is used, but disintegration time under the current Chinese Veterinary Pharmacopoeia conditions must remain within the registered limit.

Particle-Size Boundaries for Direct Compression and Automatic Capsule Filling

The standard grade is milled so that 100% passes through an 80-mesh sieve and ≥90% passes through a 100-mesh sieve. A particle-size distribution with excessive fines below 45 µm increases surface area and moisture uptake; in a rotary tablet press with 35 stations, this appears as sticking to upper punches after approximately 20 min of run time when the active concentration exceeds 40% by weight. To manage this, the fine-powder grade is specified with a D10 of 15–25 µm, D50 of 45–70 µm, and D90 of 100–150 µm by laser diffraction. These boundaries are process targets, not compendial requirements. They are derived from production-scale behaviour with 25 mm diameter, 9 mm flat-faced bevel-edge tooling and compression forces of 15–30 kN. Published data for this specific configuration is limited; new tooling shapes or press speeds may shift the acceptable particle-size window.

Wet granulation is the preferred route for tablet formulations exceeding 40% active loading. In a high-shear granulator with main blade 150–250 rpm and chopper 1500–2000 rpm, binder mass is added over 3–5 min to an endpoint of 1.5–2.5% residual moisture after drying. The granulation process reduces segregation in the feed frame, but it also exposes atractylodin to shear and moisture; assay loss after drying should be monitored against the initial input assay. If assay loss exceeds 5.0% during granule manufacture, the formulation should be reformulated with a lower binder water content or a protective excipient such as maltodextrin.

For Premix and Granule Lines, Blend Uniformity and Moisture Are the Primary Constraints

Premix manufacture uses a two-stage dilution into a ground corn or dextrose carrier. A 10:90 active-to-carrier preblend is mixed in a ribbon blender at 60% nominal fill for 12 min; the preblend is then diluted with the remaining carrier and mixed for an additional 8 min. Blend uniformity acceptance is set at relative standard deviation ≤5.0% for the marker compound across 10 sampling points. For granule production, wet mass is prepared with a binder solution at 2–5% povidone and dried in a fluid-bed dryer with inlet air temperature not exceeding 60°C to limit volatile atractylodin loss. Solution preparation from the standard powder is limited to oral drench or drinking-water products; if solution clarity is a critical quality attribute, the material must be hot-water extracted, cooled, and filtered through 1.0 µm depth media before final packaging.

After batch-release testing, the standard grade is assigned a tapped bulk density between 0.35 g/mL and 0.55 g/mL by USP <616>. For powder sachets, the particle-size distribution is adjusted to 100% pass 80 mesh and ≥90% pass 100 mesh to ensure palatability and uniform reconstitution. These are measured by analytical sieving according to USP <786> and reported on the certificate of analysis. Capsule filling on an automatic dosator machine is more sensitive to bulk density variation than tablet compression; therefore the standard grade is preferred for capsules unless the fine-powder grade is justified by dissolution testing.

Differentiating the Whole-Powder Veterinary API from Solvent Extracts and Feed-Grade Powders

Bailong Cangzhu Powder occupies an intermediate position between feed-grade botanical powder and purified molecular APIs. In feed-grade material, heavy metal and mycotoxin results are often batch-variable and not reported against a pharmacopoeial article monograph; in this veterinary API, ICP-MS and LC-MS/MS data are required before release. In purified atractylodes extract, polysaccharides and fibre are removed, which simplifies injectable formulation but eliminates the native polymer fraction that contributes to suspension stability and oral residence. The whole-powder API therefore creates both opportunity and constraint: it preserves the multi-component composition of the source plant, but it also requires more rigorous control of particle size, moisture, and bioburden across each dosage form. Commodity Cangzhu powders marketed as feed additives are not interchangeable with this API. The veterinary API is manufactured under current veterinary drug GMP principles, with lot-level identity, purity, and microbial limits reported against a registry standard. In contrast, feed-grade powders often carry only proximate analysis and mycotoxin screening. The difference is material when the powder is compounded into injectable or tablet products because pharmacopoeial article controls provide the baseline for finished-product stability and dissolution method development.

Dosage form Recommended grade Primary batch-control parameter Typical processing boundary
Tablet Fine-powder grade Granule moisture 1.5–2.5%
Capsule Standard grade Tapped density 0.35–0.55 g/mL
Injection Low-endotoxin spray-dried grade Endotoxin ≤5 EU/kg/h or registered limit
Premix Standard grade Blend uniformity RSD ≤5.0%
Solution Standard or low-endotoxin grade Clarity ≤10 NTU if injectable
Granule Fine-powder grade Inlet air temperature ≤60°C

At receipt, storage conditions for the standard grade are specified at room temperature, 15–25°C, with desiccated packaging when relative humidity exceeds 60%. The low-endotoxin grade is double-bagged in low-particulate polyethylene and sealed under nitrogen; once opened, it should be used within 24 h or re-qualified for moisture and bioburden. The powder is incompatible with strong oxidising agents and with concentrated mineral acids. Any blend containing reactive acidic excipients should be evaluated for hydrolytic degradation of atractylodin by accelerated stability testing at 40°C/75% RH for 6 months; a negative degradation result at this condition is not a substitute for real-time species-specific residue studies.
