Immediate-Release Film-Coated Tablet Compression and Aqueous Coating Dynamics

The sildenafil citrate API (molecular formula C22H30N6O4S·C6H8O7, molar mass 666.7 g/mol) is processed into immediate-release film-coated tablets through direct compression or dry granulation, selected on the basis of powder flow behavior. A 100 mg sildenafil strength requires 140.45 mg of the citrate salt, corresponding to a conversion factor of 1.4045 derived from the molar mass ratio. Representative direct compression formulations incorporate microcrystalline cellulose (Avicel PH-102) at 25–35% w/w, anhydrous dibasic calcium phosphate (Fujicalin) at 20–25% w/w, croscarmellose sodium at 3–5% w/w, colloidal silicon dioxide at 0.5–0.8% w/w, and magnesium stearate at 1.0–1.5% w/w. When the API Carr index exceeds 28, dry granulation via roller compactor (Alexanderwerk WP 120, roll force 12–18 kN, gap 2.0–2.5 mm) ensures blend uniformity with relative standard deviation not exceeding 5.0%. Compression on a 45-station rotary press (Fette 2090i, 12 mm round flat-faced bevel-edge tooling) targets main compression force of 12–18 kN, pre-compression force of 2.5–4.5 kN, tablet weight of 350–450 mg, hardness of 80–120 N (Erweka TBH 425), and friability not exceeding 0.50% after 100 revolutions per USP <1216>. On-line ejection force must remain below 1500 N to prevent punch-tip scoring and tablet capping. Aqueous film coating applies 2.0–3.0% w/w weight gain using hypromellose-based dispersion (Opadry II Blue 85G30677) in a 36-inch perforated pan (O'Hara Labcoat M) with inlet air temperature 58–65°C, bed temperature 42–46°C, pan speed 10–14 rpm, spray rate 25–35 g/min, and atomizing air pressure 1.2–1.6 bar. Dissolution testing per the USP monograph for Sildenafil Citrate Tablets employs USP Apparatus 2 (paddle) at 50 rpm in 900 mL of 0.01N HCl for 30 minutes with Q = 80%. HPLC assay uses a C18 column (150 × 4.6 mm, 5 μm) with acetonitrile:0.05 M phosphate buffer pH 3.0 at 30:70 v/v, flow rate 1.0 mL/min, and UV detection at 245 nm.

The pH-dependent solubility profile imposes process constraints. The pyrazolo[4,3-d]pyrimidin-7-one nitrogen (pKa = 6.78) ionizes below pH 5.0, conferring aqueous solubility above 2.0 mg/mL; above pH 5.5, the neutral free base precipitates. Wet granulation of sildenafil citrate therefore requires acidification of the aqueous binder solution to pH 3.5–4.0 with 0.1N HCl before spraying. Failure to acidify induces partial conversion to the poorly soluble free base and reduces dissolution rate below the USP acceptance criterion. Degradation risks comprise acid-catalyzed hydrolysis of the sulfonamide linkage, with measurable sulfonic acid impurity formation when process temperature exceeds 60°C and moisture content exceeds 3.0% w/w. Packaging in aluminum/PVC/PVDC barrier laminate with desiccant maintains water activity below 0.40 and restricts total degradation products to NMT 1.0% over 24 months at 25°C/60% RH per ICH Q1A(R2) long-term storage. Nitrosamine control under ICH M7(R2) requires confirmation that N-nitroso-sildenafil remains below the acceptable intake limit in the finished tablet.

Orally disintegrating tablets containing sildenafil citrate are manufactured by direct compression using co-processed mannitol (Pearlitol 100SD) as the primary diluent at 60–70% w/w. The formulation relies on low-residual-moisture excipients: mannitol with moisture NMT 0.5% w/w (Karl Fischer titration per USP <921>) and crospovidone with peroxide content below 100 ppm. A bifurcated disintegrant system combines crospovidone (Kollidon CL-F) at 5–8% w/w for wicking and swelling with croscarmellose sodium at 2–3% w/w for capillary draw without gelation. Aspartame at 1.5–2.0% w/w and peppermint flavor at 0.5–0.8% w/w mask the API bitterness threshold of approximately 50 ppm in aqueous solution. Compression on a 16-station rotary press (Korsch XL 100, 9 mm flat-faced tooling) applies 6–10 kN force to achieve hardness of 15–30 N, maintaining an in-house disintegration specification below 30 seconds using the USP <701> test method while satisfying friability NMT 2.0% per USP <1216>. The process conflict is explicit: hardness above 30 N prolongs disintegration to 45–60 seconds and fails the manufacturer specification for orodispersible performance. Content uniformity per USP <905> with acceptance value ≤ 15 requires API particle size D90 ≤ 25 μm (laser diffraction, Malvern Mastersizer 3000). Magnesium stearate lubricant at 0.75% w/w must not exceed 1.0% w/w; higher levels retard wetting at disintegration interfaces by 15–20 seconds through hydrophobic film formation. Stability under ICH Q1A(R2) shows no incremental degradation after 12 months at 30°C/65% RH in aluminum/aluminum cold-form blisters with 3Å molecular sieve desiccant. Direct compression avoids the pH-controlled binder requirement of wet granulation, but API concentrations below 5% w/w elevate segregation risk, necessitating geometric dilution with microcrystalline cellulose at 10% w/w as a blending intermediate.

What Process Windows Govern Reconstitutable Oral Suspension Granules?

A reconstitutable powder for oral suspension delivers 10 mg/mL sildenafil following the addition of purified water, with sildenafil citrate equivalent to 600 mg sildenafil per bottle. The dry powder comprises crystalline sorbitol at 800–900 mg/g as bulk sweetening agent, xanthan gum at 15–25 mg/g as pseudoplastic suspending polymer, sodium citrate dihydrate at 10–15 mg/g and anhydrous citric acid at 5–8 mg/g to target reconstituted suspension pH of 3.5–4.5, sodium benzoate at 2–3 mg/g as preservative, disodium EDTA at 1–2 mg/g as transition-metal ion chelator for oxidative degradation suppression, and sucralose at 0.5–1.5 mg/g as high-intensity sweetener. Wet granulation in a fluid-bed processor (Glatt GPCG 3.1) applies 5% w/v povidone K30 solution acidified to pH 4.0 with 0.1N citric acid. Inlet air temperature is controlled at 55–70°C with product bed temperature maintained at 28–35°C and spray rate of 8–12 g/min for a 3 kg batch; the 60°C product ceiling is mandatory because sulfonamide thermal degradation accelerates above this threshold. Post-drying loss on drying is reduced to ≤ 2.0% (halogen moisture analyzer, 105°C) followed by milling through a Co-mil fitted with 1.5 mm Conidur screen at 2000 rpm. Granule size distribution is controlled within D10 ≥ 50 μm, D50 180–300 μm, and D90 ≤ 850 μm (laser diffraction, dry dispersion at 1.5 bar) to ensure accurate fill weight during automated auger filling. Reconstituted suspension viscosity reaches 250–500 cP at 25°C (Brookfield RVDV-II+, spindle RV #2, 50 rpm), and 24-hour sedimentation ratio must exceed 0.90. Preservative effectiveness testing per USP <51> verifies sodium benzoate efficacy against Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Candida albicans, and Aspergillus brasiliensis over 28 days. The critical process window involves xanthan gum hydration: partial hydration during granulation is acceptable, but full hydration yields a gelled mass that blocks fluid-bed filters and reduces granule yield by up to 25%. Post-reconstitution equilibration of 10–15 minutes with intermittent shaking is required to achieve full pseudoplastic character. Acidity during reconstitution serves dual functions—maintaining sildenafil solubility above 10 mg/mL and converting sodium benzoate to undissociated benzoic acid, which necessitates pH below 4.5 for antimicrobial activity. After reconstitution, the suspension retains stability for 60 days at 25°C/60% RH or refrigerated 2–8°C. Assay after reconstitution by HPLC (C18 column, 245 nm detection) must fall within 90.0–110.0% of labeled concentration; viscosity-induced sampling variability drives the widened assay window. Dosing uses an oral syringe calibrated to 0.5 mL increments for pediatric pulmonary arterial hypertension patients weighing ≥ 8 kg.

For intravenous administration in the management of pulmonary arterial hypertension, sildenafil citrate is formulated as a sterile, clear, colorless ready-to-use solution containing 0.8 mg/mL sildenafil (as citrate) with glucose monohydrate at 50 mg/mL as tonicity modifier in Water for Injection. pH adjustment to 4.5–5.5 using 1N sodium hydroxide or 1N hydrochloric acid per USP <791> maintains the ionized pyrazolo[4,3-d]pyrimidin-7-one species below the pKa of 6.78, preventing free base precipitation. Manufacturing proceeds by dissolving sildenafil citrate in nitrogen-sparged Water for Injection at 25–30°C, followed by pH adjustment, glucose addition, and sterile filtration through a 0.22 μm PVDF membrane (Durapore GVWP). Terminal autoclaving at 121°C for 15 minutes is not applicable because forced degradation at 80°C for 8 hours generates sulfonamide hydrolysis impurity exceeding 0.5%; aseptic processing is therefore mandatory. Nitrogen overlay in the vial headspace reduces residual oxygen to ≤ 2%, suppressing oxidative degradation of the piperazine ring. Filling into 10 mL USP Type I borosilicate glass vials with 20 mm chlorobutyl rubber stoppers and flip-off aluminum seals delivers 12.5 mL per vial containing 10 mg sildenafil. In-process controls comprise pH (4.5–5.5), osmolality (280–310 mOsm/kg, vapor pressure osmometer), fill volume (12.4–12.7 mL gravimetric), and visual clarity per USP <790> under 2000–3750 lux illumination. Release testing requires sterility by membrane filtration per USP <71>, bacterial endotoxins per USP <85> with limit NMT 1.0 EU/mL (equivalent to NMT 12.5 EU/dose for the vial), and subvisible particulate matter per USP <788> with limits of NMT 6000 particles ≥ 10 μm and NMT 600 particles ≥ 25 μm per container using light obscuration. The infusion is administered after dilution in 0.9% sodium chloride or 5% dextrose to 50 mL, or via direct slow intravenous injection over 3 minutes. Diluted solutions in PVC or polyolefin infusion bags remain stable for 24 hours at 25°C protected from light. Precipitation occurs upon admixture with alkaline injectables having pH above 8.0, particularly sodium bicarbonate 8.4%; administration sets must be flushed with 0.9% sodium chloride before and after infusion to avoid in-line crystallization in catheter dead volumes. Off-label dosing for persistent pulmonary hypertension of the newborn employs a loading dose of 0.4 mg/kg over 3 hours followed by continuous infusion at 1.6 mg/kg/day; hemodynamic monitoring is required because systemic vasodilation may produce clinically significant hypotension in the first 30 minutes of infusion.

When Aqueous Film Coating Overhead Is Removed, Hard Capsule Shell Encapsulation Parameters Apply

Hard gelatin capsule filling serves as a lower-cost unit operation for development batches and compounded preparations. Sildenafil citrate is blended with lactose monohydrate (dried, LOD ≤ 1.0%) at 60–75% w/w, microcrystalline cellulose at 15–25% w/w, croscarmellose sodium at 2–4% w/w, colloidal silicon dioxide at 0.5% w/w, and magnesium stearate at 1.0% w/w, then filled into size 1 (25 mg), size 0 (50 mg), or size 00 (100 mg) hard gelatin shells using a semi-automatic capsule machine (CapPlus CP240, 4800 capsules/hour). The shell moisture content of 13–16% w/w per USP <921> protects the API from atmospheric hydration, but fill powder LOD must remain below 2.0% w/w to prevent shell embrittlement in amber HDPE bottles with 33 mm child-resistant closures. Dissolution mirrors the tablet monograph: USP Apparatus 2 at 50 rpm in 900 mL of 0.01N HCl, Q = 80% at 30 minutes, with wire sinkers to prevent flotation. Content uniformity per USP <905> for the 25 mg strength (fill weight 180–200 mg) requires API particle size D90 ≤ 50 μm. Relative humidity above 60% in the filling suite causes gelatin softening and shell deformation; environmental control at 40–45% RH and 20–25°C is therefore mandatory per FDA 21 CFR 211.42 general facility requirements for capsule manufacturing operations. Beyond the defined blending, filling, and dissolution parameters, no additional formulation complexity is required for this established dosage form.

In sublingual applications, sildenafil citrate is explored for pharmacokinetic advantages bypassing first-pass hepatic metabolism; published data on commercial-scale sublingual manufacturing remains limited and cannot be generalized without formulation-specific bioavailability studies. Compounded sublingual tablets are prepared by direct compression with lactose monohydrate at 45–55% w/w, microcrystalline cellulose (Avicel PH-101) at 20–30% w/w, crospovidone at 5–8% w/w, sodium starch glycolate at 2–3% w/w within a triple-disintegrant system, and peppermint flavor at 0.5–1.0% w/w. A 25 mg sublingual tablet compresses at 5–8 kN on a 10-station rotary press with 8 mm round concave tooling to hardness 12–25 N, targeting disintegration of 15–45 seconds in 900 mL water at 37°C using the USP <701> test method. Clinical pharmacology references report sublingual Cmax at 15–30 minutes versus 60–90 minutes for the conventional oral tablet, with relative bioavailability of 70–90% versus oral administration. The operational boundary involves local tissue irritation: the acidic formulation pH required to maintain sildenafil solubility below pH 5.0 may induce sublingual mucosal irritation over repeated dosing. Compounding pharmacies must verify content uniformity under USP <795> and document in-use stability not exceeding 90 days in tight, light-resistant containers at controlled room temperature per USP <795> beyond-use dating tables. Published data for specific commercial sublingual sildenafil configurations is limited.

    More Introduction

Slidenafil Citrate Pharma Grade API is released in two model designations: SLC-PG-101 for tablet, capsule, and granule manufacture, and SLC-IG-102 for injectable and sterile-liquid preparation. The article is the citrate salt of 5-[2-ethoxy-5-(4-methylpiperazin-1-ylsulfonyl)phenyl]-1-methyl-3-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-one, CAS 171599-83-0, molecular formula C22H30N6O4S·C6H8O7, relative molecular mass 666.70 g/mol. The API is manufactured under ICH Q7 cGMP and controlled against USP Sildenafil Citrate and Ph. Eur. Sildenafil Citrate monographs. It is an active pharmaceutical ingredient for further processing, not a finished dosage form. The oral solid dosage grade is intended for direct compression, wet or dry granulation, and capsule filling; the injectable grade is intended for terminally sterilized or aseptic parenteral solution preparation.

Which Compendial Release Limits and Methods Apply to the Oral Solid Dosage Grade?

Release of SLC-PG-101 is based on a combination of compendial methods and ICH Q3A, Q3C, and Q3D requirements. Pharmacopoeial identification requires infrared absorption and HPLC retention time against certified reference standard. Assay by HPLC on the dried basis is controlled to 98.0–102.0%; loss on drying is controlled to ≤0.5% by USP <731>. Related substances limits are set according to ICH Q3A thresholds unless a stricter monograph limit applies: unspecified impurities ≤0.10%, total impurities ≤1.0%. Particle size is controlled because it directly modifies dissolution and content uniformity in low-dose tablet and capsule manufacture.

Direct compression and capsule filling performance require more restrictive physical control than chemical release alone. A batch with D90 ≤100 µm and Hausner ratio between 1.15 and 1.35 generally discharges uniformly from a bin blender at 10 rpm over 15–20 min. If the Hausner ratio exceeds 1.40, wet granulation or slugging is recommended because inter-tablet content uniformity risk increases. In capsule filling on a dosator-type machine, bulk density below 0.30 g/mL can produce cap-to-body weight variation above 3%. Low-dose capsules are therefore filled with a pre-blend containing lactose monohydrate or microcrystalline cellulose. Dry granulation by roller compaction at roll pressure 5–10 kN/cm and screen size 1.0 mm typically yields granules with Carr index 15–20. Wet granulation with povidone K30 at 2–3% w/w is used when higher tablet hardness is required; granulate drying at inlet air temperature 50–60 °C and milling through a 0.8 mm screen reduces over-granulation.

When Injectable-Grade Material Is Selected for Sterile Manufacturing

SLC-IG-102 imposes additional release tests beyond the oral solid dosage grade. Bacterial endotoxins are tested by Ph. Eur. 2.6.14 or USP <85>. A common API release criterion is ≤0.50 EU/mg when the finished product is a 10 mg parenteral dose, but the final limit is derived from the finished product monograph. Aqueous solubility of Slidenafil Citrate is approximately 3.5 mg/mL at 25 °C; a 1% w/v solution typically has a pH of 3.0–4.5. Raising pH above 5.5 with sodium hydroxide, phosphate buffer, or sodium bicarbonate may precipitate the free base. For aseptic processing, a 0.22 µm PVDF or polyethersulfone filter is typically validated; nylon and mixed-cellulose esters may show adsorptive loss of this weakly basic drug substance, and filter validation data for this specific API-filter configuration should be generated. The injectable grade is low-bioburden only, not sterile; terminal sterilization or aseptic filtration of the final solution remains a finished-product operation. Visual clarity of the API solution at 1% w/v is controlled as an incoming-materials test, while subvisible particulate matter is controlled on the finished product by USP <788>.

Dry powder stability is maintained at 15–25 °C and relative humidity below 60% in double polyethylene bags inside an HDPE drum. In aqueous formulations, hydrolysis can occur above pH 7; oral suspensions and reconstituted injectable solutions should therefore be buffered to the acidic range and protected from light. Avoid blending with strong alkaline lubricants or effervescent bases because carbon dioxide release and pH rise can alter dissolution rate and particle agglomeration. Published data for long-term storage in single-use plastic containers are limited; extractables and leachables studies should follow USP <1664> for parenteral packaging. Containers opened outside controlled temperature and humidity should not remain exposed for more than 8 h without re-testing loss on drying and particle size.

Residual Solvent and Elemental Impurity Control Strategy

Residual solvent limits follow ICH Q3C. Ethanol, isopropanol, and acetone are Class 3 solvents controlled at ≤5000 ppm. Methanol, if used as a crystallization solvent, is controlled at ≤3000 ppm; methylene chloride, if present as a Class 2 solvent, is limited to 600 ppm. Elemental impurities are evaluated by ICP-MS according to USP <233> and ICH Q3D. For oral products, the permitted daily exposure values are 5 µg/day for cadmium, 5 µg/day for lead, 15 µg/day for arsenic, and 30 µg/day for mercury. For injectable products, the limits are 2 µg/day for cadmium, 5 µg/day for lead, 15 µg/day for arsenic, and 3 µg/day for mercury. When the API is used in parenteral formulations, the lower parenteral permitted daily exposures control release of Class 1 and Class 2A elements. The manufacturer provides an ICH Q3D Table A.2.2 risk assessment for each batch.

The citrate salt differs from the free base and from in situ mixtures of base and citric acid in solubility, processing consistency, and compendial control. In situ salt formation requires stoichiometric conversion verification; a preformed citrate salt removes that variability. The free base is practically insoluble in water, typically below 0.1 mg/mL, while the citrate salt is reported at approximately 3.5 mg/mL at 25 °C. This improves wettability and reduces the need for surfactants in oral suspension manufacture, and permits solution-based injectable manufacturing without organic co-solvents.

Particle size distribution is controlled not only as a release parameter but as a process performance variable. For immediate-release tablets, dissolution rate depends on wetting, particle surface area, and intra-granular distribution. A micronized API lot with D90 ≤15 µm in a directly compressible blend generally meets a dissolution criterion of not less than 80% dissolved in 30 min in 0.1 M HCl at 50 rpm using USP apparatus 2, while an unmicronized lot with D90 above 150 µm may show higher inter-tablet variability and slower release because of poor wetting. Over-micronized cohesive powder can agglomerate; colloidal silicon dioxide at 0.5–1.0% w/w and a two-stage blending sequence reduce this risk. The two-stage sequence blends API plus diluent for 10 min, then adds magnesium stearate at 0.25–0.50% w/w for an additional 3 min. Magnesium stearate above 1.0% w/w may create hydrophobic films and delay dissolution.

Incoming handling requires sealed double polyethylene bags inside HDPE drums at 15–25 °C. Re-test after 24 months if stored in the original unopened container. Do not use after exposure to relative humidity above 60% for more than 8 h without re-testing loss on drying, particle size, and microbial limits. The API is incompatible with strong oxidizing agents and strong alkaline materials; any formulation combining these agents should be assessed by forced-degradation studies under ICH Q1B photostability and ICH Q1A stress conditions.
