Application of Sertralin Pharma Grade API for Tablet / Capsule / Granule / Injection, Oral & Injectable

Direct compression of sertraline hydrochloride for immediate-release film-coated tablets places the primary stress on blend flow and particle-size distribution rather than on API chemical stability. The crystalline hydrochloride salt is typically milled to a volume mean diameter that allows a content uniformity acceptance value ≤ 15.0 under USP <905>, but overmilling below a D90 of approximately 40 µm can increase cohesion, die-fill variability, and capping tendency on high-speed rotary presses. In production-scale trials on rotary tablet presses equipped with precompression rollers, main compression force is frequently qualified in the range of 8 to 18 kN for 100 mg strength tablets produced with 9 mm round biconvex tooling; the precise window is press-model dependent and must be established through force-hardness and ejection force profiles. A direct compression formulation commonly includes microcrystalline cellulose PH102, spray-dried lactose monohydrate, sodium starch glycolate as disintegrant, colloidal silicon dioxide as flow aid, and magnesium stearate as lubricant. The order of addition is critical: magnesium stearate is added at 0.5% to 1.0% w/w to the pre-blend and mixed for no more than 3 to 5 minutes at 10 to 15 rpm in a bin blender because excessive shear distributes the hydrophobic lubricant film over the API and disintegrant, retarding dissolution in 900 mL of 0.1 M hydrochloric acid at 37±0.5°C with paddle speed 50 rpm under USP <711>. Tablet hardness is typically controlled between 60 and 120 N and friability is maintained below 1.0% w/w according to USP <1216>. Aqueous film coating with hypromellose, polyethylene glycol and titanium dioxide at a weight gain of 2.0% to 4.0% is applied after tableting; coating pan inlet air temperature and spray rate are balanced to avoid over-wetting, which can activate the disintegrant and cause edge chipping during subsequent packaging. Finished tablets must meet degradation product limits under ICH Q3B; sertraline hydrochloride is sensitive to photolytic and oxidative stress, so the API and blend are protected from light and the coating formulation may include an opacifier to reduce light transmission.

What Mixing Order Prevents Segregation in Sertraline HCl High-Shear Granulation?

In wet granulation, the process risk moves from powder flow to binder distribution and granule density equilibrium. Sertraline hydrochloride loaded at 20% to 40% w/w of the tablet core can be granulated in a high-shear mixer with an aqueous binder prepared from hydroxypropyl cellulose or pregelatinized starch at 3% to 5% w/w solids. The chopper speed and the wet massing time after binder addition are the two parameters most likely to shift the resulting granule size distribution from a target of roughly 100 to 300 µm D50 when a top-spray fluid-bed dryer is used for moisture removal. If the binder is discharged too rapidly, localized overwetting produces hard, coarse granules that survive the subsequent milling step and create content uniformity failures because the sertraline-rich lumps do not break down during compression. If the massing time is too short, the granule fraction below 75 µm remains above 30% w/w, increasing segregation risk in the transfer hopper and press feed frame. Production-scale experience shows that a high-shear mixer with main impeller speed of 150 to 300 rpm and chopper speed of 1,000 to 3,000 rpm is capable of reaching a suitable endpoint, but the exact settings depend on bowl fill level, binder viscosity and API lot particle-size distribution. Drying in a fluid-bed at product temperature not exceeding 45°C is generally specified to avoid thermal degradation; residual moisture is controlled at 1.0% to 3.0% by loss-on-drying at 105°C according to USP <731>. Overdrying below 1.0% produces brittle granules and raises the fines fraction, while moisture above 3.0% can reduce hardness uniformity and increase sticking on the lower punch. After drying and sizing through a 0.8 mm screen, the granules are blended with extragranular disintegrant and lubricant before compression. The extragranular portion of sodium starch glycolate at 2% to 4% w/w is required to achieve immediate-release disintegration below 15 minutes in 0.1 M hydrochloric acid; placing the entire disintegrant intragranularly can slow dissolution because the disintegrant is encapsulated within the granule matrix. Tablets produced by wet granulation normally display higher breaking force at equivalent compression force than direct compression blends, so the main compression force may be reduced to avoid exceeding 120 N hardness and to keep dissolution from falling below the Q value stated in the approved specification. This process is particularly justified when the API lot fails direct compression flow criteria, such as a compressibility index above 25% under USP <616>, or when the drug load exceeds 30% w/w and the direct compression blend cannot maintain an acceptance value below 15.0 for content uniformity.

For hard gelatin capsule filling, the specification set shifts from tablet hardness to powder plug formation consistency under tamping pins and the weight variation limits of USP <905> as applied to capsules. Sertraline hydrochloride in capsule form is often manufactured from a dry blend or from the same wet granulation intermediate used for tablets; the choice depends on whether the API and excipients can maintain a compressibility index below 25% and a flow rate sufficient for automatic capsule filling machines operating at 50,000 to 120,000 capsules per hour. When a dosator nozzle system is used, the blend must form a firm plug under compression and must release the plug without sticking; a tamping-pin system requires lower cohesive strength but higher flow uniformity to keep the individual tamping stations within the target fill weight range. For a 50 mg strength capsule filled into a size 3 hard gelatin shell, the total fill weight is typically in the range of 120 to 180 mg depending on bulk density; the API is pre-blended with lactose monohydrate through geometric dilution and then screened through a 0.5 mm sieve before final blending. Magnesium stearate at 0.5% to 1.0% w/w is added last; the total blending time after lubricant addition is limited to 3 to 5 minutes because lubricant over-mixing can delay disintegration of the capsule plug in the dissolution vessel. In-process controls include fill weight variation at intervals of 15 to 30 minutes, capsule moisture content below 13% w/w to prevent shell brittleness or softening during storage, and visual inspection for cracked shells and powder leakage. The finished capsule must meet a disintegration time of not more than 30 minutes in purified water at 37±0.5°C under USP <701>, and dissolution in 0.1 M hydrochloric acid with the same conditions described for tablets. Capsule formulations containing sertraline hydrochloride should not be stored with unsealed desiccant cartridges that reduce headspace humidity below 20% RH, because excessive drying can make gelatin shells brittle; the same constraint applies to blisters with cold-form foil that approach zero moisture vapor transmission. Published stability data for sertraline hydrochloride capsules stored in clear PVC blisters are limited; therefore, light-opaque and moisture-protective packaging materials are specified unless the coating or shell contains a suitable opacifier.

When Granulated Sertraline HCl Is Filled into Sachets or Reconstituted as an Oral Suspension

Granulated sertraline hydrochloride intended for sachet filling or reconstitution into a suspension introduces a distinct set of constraints: the granule must remain free-flowing after storage, resist segregation during transport, and release the API rapidly when dispersed in water or a pH-adjusted vehicle. Because sertraline hydrochloride is a weak base with a pKa of approximately 9.5, the solubility of the non-ionized free drug in neutral and intestinal pH media is much lower than the solubility of the protonated salt in acidic media; therefore, the reconstitution vehicle is often acidified or buffered below pH 5.0 to maintain the API in solution and to prevent precipitation when the suspension is mixed with gastric fluid. Granule formulations may employ a matrix of sugar spheres, mannitol, or lactose monohydrate built up by fluid-bed spray granulation; a polymer coating such as hydroxypropyl methylcellulose or ethylcellulose can be applied at 3% to 8% weight gain to mask bitterness and to protect the hydrochloride salt from moisture uptake. The coating step is also used to modulate release; however, the established oral dosage forms of sertraline hydrochloride are immediate release, so extended-release coated granules would fall outside the conventional pharmacopoeial dissolution acceptance criteria unless a separate justified specification is developed. For a reconstituted suspension, the granules are packaged in unit-dose sachets with a desiccant and a combined moisture and oxygen scavenger; the sachet film is typically a four-layer laminate containing polyethylene terephthalate, aluminum foil and low-density polyethylene to keep moisture transmission below 0.1 g/m²/day. In-use stability after reconstitution is a critical boundary: a vehicle prepared from purified water may allow microbial growth, so a preserved vehicle containing methylparaben and propylparaben or another pharmacopoeial preservative is required if the product is to be stored beyond 24 hours at room temperature. The oral suspension must pass dose delivery uniformity after shaking, with a delivered dose acceptance range of 90.0% to 110.0% of label claim, and the granules must pass dissolution testing in 0.1 M hydrochloric acid using the same apparatus as the tablet form. The viscosity of the suspending medium is typically adjusted with xanthan gum or microcrystalline cellulose/sodium carboxymethylcellulose to a target of 50 to 200 mPa·s at 25°C; values above 200 mPa·s can slow wetting and drug diffusion from the granules, while values below 50 mPa·s can lead to rapid sedimentation and poor dose uniformity. Sertraline hydrochloride granule formulations are not a direct substitution for the approved oral concentrate solution containing ethanol; the granule route is used where solid-dose convenience is required and where the patient population can tolerate a dispersed formulation. No pharmacopoeial monograph for sertraline hydrochloride granules as such is published, so specifications are derived from the tablet monograph and the general chapters for oral powders and suspensions, including USP <1174> for powder flow and USP <795> for extemporaneous preparations where applicable.

Injectable Sertraline HCl Solubility Boundaries and Sterility Assurance Requirements

Injectable development of sertraline hydrochloride is constrained less by chemical stability than by aqueous solubility at physiological pH and by the absence of a compendial parenteral monograph. Sertraline hydrochloride has pH-dependent solubility; the protonated salt dissolves more readily in acidic aqueous media, but dilution into phosphate-buffered saline or 0.9% sodium chloride injection can raise the local pH and cause precipitation of the free base unless a buffered formulation or a cosolvent system is used. Formulation development for a parenteral product therefore requires a phase-solubility study at 25±0.5°C across pH 3.0 to pH 7.4, with HPLC assay of the supernatant after equilibration and filtration through a 0.45 µm filter. If a target concentration above the aqueous solubility limit is required, a cosolvent such as propylene glycol or polyethylene glycol 400 may be evaluated; however, each cosolvent percentage changes the osmolality, viscosity and possible hemolytic potential, so the final formulation is optimized to an osmolality of 285 to 310 mOsm/kg and a pH between 3.5 and 5.5 where the drug remains ionized. Terminal sterilization by autoclaving at 121°C for 15 minutes may be feasible only if forced degradation studies show no more than a threshold increase in specified degradation products; otherwise, aseptic filtration through a 0.22 µm membrane followed by filling under Grade A conditions is required. The filter compatibility test must demonstrate that the API does not precipitate on the membrane during the full filtration run at 20 to 25°C and that the filter extractables remain within the limits of the filter supplier and the regulatory dossier. Container closure selection for a parenteral sertraline hydrochloride solution uses Type I borosilicate glass vials meeting USP <660>, with chlorobutyl or bromobutyl rubber stoppers meeting USP <381>; the fill volume and headspace are controlled to avoid excessive oxygen exposure, and the vial is flushed with nitrogen if oxidative degradation is identified. Bacterial endotoxin testing is performed using USP <85>; the endotoxin limit is calculated from the maximum labelled dose and route, and a conservative upper bound of 0.5 EU/mg is often adopted for development-grade parenteral sertraline hydrochloride when the maximum dose is 50 mg or less. Particulate matter is limited by USP <788> to not more than 6,000 particles ≥ 10 µm and 600 particles ≥ 25 µm per container for small-volume parenterals. Injectability and precipitation upon dilution must be tested by mixing the product with 0.9% sodium chloride injection and 5% dextrose injection at ratios of 1:1 and 1:10 under static conditions for at least 4 hours; visible precipitation per USP <790> is a rejection criterion. Published clinical and commercial data for injectable sertraline hydrochloride are limited; therefore, all parenteral formulation decisions should be treated as development-stage and supported by ICH Q1A stability protocols, ICH Q8 design-space documentation, and container-closure integrity testing per USP <1207>.

    More Introduction

Sertralin Pharma Grade API is supplied as sertraline hydrochloride, the active pharmaceutical ingredient described chemically as (1S,4S)-4-(3,4-dichlorophenyl)-1,2,3,4-tetrahydro-N-methyl-1-naphthalenamine hydrochloride, with CAS 79559-97-0, molecular formula C17H17Cl2N·HCl, and molecular weight 342.69 g·mol⁻¹. The product is manufactured under ICH Q7 good manufacturing practice for active pharmaceutical ingredients and is suitable for formulation into tablets, capsules, granules, and injectable dosage forms when the relevant dosage form is supported by the marketing authorization. It is not a technical-grade or research-grade sertraline base; the hydrochloride salt is supplied as a crystalline powder with controlled particle size, residual solvent profile, and elemental impurity limits. Model differentiation within the product range is not a chemical distinction but a quality-control distinction: oral solid-dose grades are specified by particle size range for direct compression or granulation, while injectable grade is controlled for reduced bioburden and bacterial endotoxin burden. The active molecule and HCl salt stoichiometry remain identical across the model classes.

Sertraline hydrochloride belongs to the selective serotonin reuptake inhibitor class and has a tetrahydronaphthalene ring system, which separates it analytically from fluoxetine hydrochloride and paroxetine hydrochloride at the level of retention behaviour, impurity profile, and stereochemical control. The product is controlled for the (1S,4S) enantiomer and for the trans diastereomer, which is the principal isomeric impurity arising from synthetic routes that pass through cis/trans tetralone intermediates. Because the pharmacopoeial assay and related substances methods are specific to sertraline, they cannot be exchanged with monographs for other SSRI salts without revalidation under ICH Q2(R1).

What Limits Residual Impurities and Polymorphic Drift in Sertraline Hydrochloride?

Residual impurity control is derived from three linked frameworks: USP general chapter 467 for residual solvents, ICH Q3C for class-specific solvent limits, and USP 232/233 for elemental impurities under ICH Q3D. Residual solvents are quantitated by headspace gas chromatography with flame ionization detection; injectable grade is assigned a lower residual solvent target than oral grade when solvent removal during drying is less efficient. Water content is measured by Karl Fischer coulometry under USP 921. Unless the API is dispensed in a humidity-controlled isolator, exposure to ambient air above 60% relative humidity during weighing can raise surface moisture and shift compression behaviour. The acceptance criterion for water is ≤0.5%, based on the anhydrous assay calculation.

Polymorphic identity is confirmed by X-ray powder diffractometry under USP 941; differential scanning calorimetry under ISO 11357-1 shows a melting endotherm near 245 °C for the crystalline hydrochloride. Conversion to the free base or partial amorphization during high-energy milling changes dissolution rate and triboelectric charging of the powder. Pin mills and jet mills used for particle size reduction are therefore operated with cooling jackets and inert nitrogen blanketing when the required D90 is below 50 µm. Mill speed and feed rate are monitored because the energy input interacts with any amorphous surface layer generated during size reduction.

Assay is determined by high-performance liquid chromatography using a C18 or phenyl stationary phase with UV detection at the wavelength specified in the current monograph; the acceptance range is 98.0–102.0% on the anhydrous basis. Impurity methodology resolves trans-sertraline, desmethylsertraline, and the chlorophenyl positional isomers. Total impurity content is controlled by the monograph rather than by a single blanket limit; process validation batches establish a reference chromatographic fingerprint against which subsequent commercial batches are compared. This fingerprint approach is required under ICH Q7 change management because a shift in the late-stage reductive amination catalyst can alter the relative area percent of the trans isomer.

For oral solid dosage forms, the critical processing attributes are particle size distribution, bulk density, and surface moisture. Direct compression blends containing sertraline hydrochloride are typically pressed on rotary tablet machines with 8–12 mm round concave tooling, followed by film coating in perforated pan coaters. The API is pre-screened through a 40-mesh screen (425 µm) to break loose agglomerates and then blended with microcrystalline cellulose, modified starch, and magnesium stearate. Content uniformity testing under USP 905 is performed on 10 dosage units, and dissolution testing under USP 711 is performed in the monograph-defined medium. In-process controls are maintained under 21 CFR 211.110, and final release testing is conducted under 21 CFR 211.165.

Direct Compression, Wet Granulation, and Dry Granulation Handling Boundaries

Direct compression is feasible when the D90 remains below 150 µm and the compressibility index is maintained below 25%. Lots with high fines below 10 µm can cause sticking to punch faces and tablet picking on high-speed rotary equipment. Over-lubrication with magnesium stearate above 0.75% w/w or extended blending beyond 2 minutes at high shear can reduce tablet breaking force and raise friability. Wet granulation is used when direct compression cannot meet weight variation or when the active load requires a more robust granule structure. The granulation liquid is purified water, an aqueous starch paste, or a hydroxypropyl cellulose solution; water level is adjusted to produce granules with a loss on drying between 3.0% and 4.5% before tray drying at 50–60 °C. Endpoint control is by torque rheometry on a high-shear granulator; a torque plateau indicates granule coalescence, while over-wetting leads to oversized granules above 2 mm that retard drying and cause tablet surface mottling after coating. Fluid-bed drying is preferred for heat-sensitive batches because it removes surface moisture more quickly and reduces binder migration to the granule surface.

Dry granulation by roller compaction is limited by the reduced reworkability of the material after compaction. Slug tensile strength below 1.5 MPa produces high fines that segregate in the tablet feed frame, whereas compaction above 4.0 MPa creates hard ribbons that require aggressive milling and can alter the dissolution profile. Roller compaction is therefore specified by roll force, roll speed, and screen aperture bracketed during design of experiments; a single fixed parameter set is not transferable across batches. Granule bulk density is measured using USP 616 and recorded as a routine in-process check, while near-infrared spectroscopy can be used for real-time blend uniformity monitoring.

When Injectable-Grade Sertraline Hydrochloride Is Required

Injectable-grade material is the same chemical salt but is controlled for bacterial endotoxin, bioburden, and sub-visible particulate matter. Endotoxin is assessed by kinetic chromogenic limulus amoebocyte lysate or recombinant Factor C under USP 85; the acceptance limit is calculated as 1.75 EU/mg when the maximum adult daily dose is 200 mg and the patient body weight is 70 kg. This calculation follows the K/M formula with K=5 EU/kg for parenteral administration. Sterility is not a routine bulk API release test because the API is not terminally sterilized in its bulk state; instead, the finished injectable dosage form is sterilized by terminal moist heat or aseptic filtration under validated conditions. The API is packed in low-endotoxin polyethylene liners with desiccant, and handling occurs in an ISO 7 cleanroom with HEPA-filtered airflow.

Sub-visible particulate matter for the finished injection is tested by light obscuration under USP 788; the finished product must meet no more than 6000 particles ≥10 µm and 600 particles ≥25 µm per container for a small-volume injection. The API itself is not released by USP 788 because it is not supplied as a finished injection, but the supplier can provide a particle size distribution with D99 controlled by laser diffraction under ISO 13320:2020 to demonstrate that material aggregates are below the filtration threshold. Published data for this specific configuration is limited: sertraline hydrochloride has no widely established commercial injectable monograph, so parenteral development must be justified with additional stability and compatibility studies in the chosen solvent system. The oral grade should not be substituted for the injectable grade solely on assay equivalence because endotoxin and particulate burden are not reduced by terminal filtration to the same degree in every formulation.

A separate processing condition arises when the API is used in oral solution or oral concentrate rather than a solid dosage form. Sertraline hydrochloride is dissolved in a vehicle containing ethanol, glycerol, and purified water; because aqueous solubility is limited, mixing is performed in stainless steel vessels with ethanol or propylene glycol as co-solvents. The solution is filtered through a 0.45 µm membrane and protected from light. Mixing speed and temperature are controlled because the hydrochloride salt can release HCl in aqueous media and accelerate hydrolysis of ester-containing flavouring agents. This route is less sensitive to particle size than tablet manufacture but more sensitive to residual water and elemental impurity content.

The difference between sertraline hydrochloride API and other SSRI APIs in formulation is not primarily clinical efficacy but solid-state and impurity behaviour. Fluoxetine hydrochloride and paroxetine hydrochloride have their own residual solvent, moisture, and particle size challenges; sertraline’s tetrahydronaphthalene ring requires a stereochemical purity method that separates the cis and trans isomers, while fluoxetine and paroxetine require enantiomeric purity methods based on their own chiral centres. These analytical differences mean that a method transfer from one SSRI product to sertraline hydrochloride without revalidation of the chiral stationary phase and extraction procedure is not valid under ICH Q2(R1) and ICH Q7.

For tablet and capsule formulations, the API is matched to the granulation route by its particle size and bulk density, not by total purity alone. Two batches can both meet assay 98.0–102.0% yet behave differently on a rotary tablet press if one batch has a D90 of 140 µm and another has a D90 of 250 µm. The higher-D90 lot may require a pre-milling step through a cone mill fitted with a 1 mm screen; this operation is performed under a nitrogen blanket when the API remains in the mill for more than 30 minutes. Process analytical technology using near-infrared spectroscopy can control blend uniformity in real time, reducing the risk that segregation in the feed frame produces low-potency tablets at the start of the batch.
