Application of (R)-N-Boc-3-pyrrolidineacetic acid Pharma Grade API for Tablet / Capsule / Granule / Injection, Oral & Injectable
In direct compression processing of (R)-N-Boc-3-pyrrolidineacetic acid as a crystalline free acid with a molecular mass of 229.27 g/mol, particle size distribution parameters exert the dominant influence on blend uniformity and tablet content uniformity. Milling through a Quadro Comil 197 equipped with a 0.032-inch round screen at 2000 rpm reduces D90 to ≤ 150 μm, verified by laser diffraction per USP 429. A rotary tablet press configured with a 16-station turret (Korsch XL 100, maximum compression force 60 kN) operating at 20–40 rpm with a compression force of 8–20 kN and a dwell time of 20–100 ms produces tablets with hardness 5–8 kp (Schleuniger Model 6D). The formulation window across three oral solid dosage formats is summarized in the comparative matrix below.Table 1 — Comparative formulation composition ranges for oral solid dosage processing of (R)-N-Boc-3-pyrrolidineacetic acid.| Component | Function | Tablet (% w/w) | Capsule (% w/w) | Granule (% w/w) ||---|---|---|---|---|| (R)-N-Boc-3-pyrrolidineacetic acid | Active | 5–25 | 10–30 | 5–20 || Microcrystalline cellulose (Avicel PH-102) | Diluent | 60–75 | 50–70 | 40–60 || Lactose monohydrate | Diluent | 10–20 | 10–25 | 15–25 || Croscarmellose sodium | Disintegrant | 2–5 | 2–5 | 2–5 || Povidone K30 | Binder | — | — | 3–5 || Magnesium stearate | Lubricant | 0.5–1.0 | 0.5–1.0 | 0.5–1.0 || Colloidal silicon dioxide | Glidant | — | 0.5–1.0 | 0.5–1.0 || Purified water (removed during drying) | Granulating fluid | — | — | q.s. |Friability remains ≤ 1.0% per USP 1216 for tablets with a diameter ≥ 8 mm and a thickness of 3–4 mm. Dissolution testing per USP 711 using Apparatus 2 (paddle) at 50 rpm in 900 mL of phosphate buffer pH 6.8 maintained at 37.0 ± 0.5°C yields Q ≥ 80% at 30 min for immediate-release specifications. Content uniformity per USP 905 requires an acceptance value ≤ 15.0, reliably achievable when the D90 of the active component is controlled at ≤ 150 μm. The critical boundary condition resides at relative humidity above 60%: the tert-butoxycarbonyl protecting group undergoes stepwise acid-catalyzed cleavage, liberating 2-methylpropene and carbon dioxide. Measurable deprotection occurs at a water content of 0.5% w/w in the presence of acidic microenvironments from hydrolyzed excipients. Processing suites are therefore maintained at 30–45% RH with dew point control to prevent batch rejection attributable to Boc loss. Residual solvent limits per ICH Q3C apply: ethyl acetate is a Class 3 solvent with a limit of 5000 ppm, and any residual dichloromethane carried from upstream isolation steps is controlled at NMT 600 ppm (Class 2). Water content by Karl Fischer titration per USP 921 is specified at NMT 0.5% w/w at batch release. Loss on drying per USP 731 at 105°C for 2 h must not exceed 0.5%.
What Process Parameters Govern Hard Capsule Filling Uniformity at 200–500 mg Target Fill Weights?
Hard gelatin capsule filling of the compound on a Bosch GKF 1500 intermittent-motion machine requires a pre-blending step in a bin blender rotating at 25 rpm for 10–15 min to ensure homogeneous distribution of the active component throughout the fill matrix. Weight variation per USP 905 must not exceed ±3% RSD for 10 of 30 individual capsules weighing less than 300 mg. A blend with a Carr compressibility index below 20% and a Hausner ratio below 1.25, determined per USP 616 Method I, is achieved by incorporating colloidal silicon dioxide at 0.5–1.0% w/w. Filling of size 0 or size 1 hard gelatin capsules proceeds at 1500–2000 capsules per hour per segment, with tamping pin penetration depths of 2–4 mm across 3–5 tamping stations. Capsule shell moisture content of 13–16% w/w for gelatin shells and 3–7% w/w for HPMC shells is maintained to prevent brittleness, softening, or premature dissolution. Dissolution from filled capsules is evaluated per USP 711 using Apparatus 1 (basket) at 100 rpm in 900 mL of 0.1 N hydrochloric acid for 60 min or until Q ≥ 80%. Storage stability data for capsules in aluminum/aluminum blister packs at 25°C/60% RH and 40°C/75% RH over 6 months demonstrates that assay per USP 621 remains within 95.0–105.0% of label claim when the capsule formulation excludes acidic fillers such as citric acid or tartaric acid. Acidic excipients accelerate Boc cleavage during accelerated stability storage, generating free pyrrolidine degradation products that are quantified by HPLC at 210 nm.High-shear wet granulation of the compound with a 5% w/v povidone K30 aqueous binder solution in a GEA Aeromatic-Fielder PMA 10 high-shear granulator at an impeller speed of 300–500 rpm and a chopper speed of 1500–2500 rpm for 3–5 min yields granules with a mean particle size of 250–400 μm. The terminal carboxylic acid group participates in intermolecular hydrogen bonding with the pyrrolidone carbonyl oxygen of PVP K30, retarding binder migration toward the granule surface during subsequent fluid bed drying. Drying is conducted at inlet air temperatures of 50–60°C with product temperatures maintained at 35–42°C to preserve the Boc group. Loss on drying per USP 731 is monitored until NMT 2.5% w/w is reached, typically within 20–30 min of drying time. Dried granules are milled through an 18-mesh (1.0 mm) screen using a Quadro Comil 197 at 2000 rpm with a round impeller. The resulting granule fraction exhibits a bulk density of 0.45–0.55 g/cm³, a tapped density of 0.55–0.65 g/cm³, and a compressibility index below 20% per USP 616. A documented incompatibility exists with granulating fluids buffered at pH below 3.0: quantitative Boc deprotection is detectable by HPLC within 60 min at 25°C, producing the free pyrrolidine species. Granules intended for sachet reconstitution are filled into aluminum foil laminate pouches with silica gel desiccant, with headspace oxygen controlled at NMT 5% v/v and sealing performed under nitrogen. The Boc protecting group is thereby preserved for a 24-month shelf life at 2–8°C. Batch-to-batch variance in granule size distribution has been observed to correlate inversely with impeller tip speed consistency; lines operating with worn impeller blades exhibit a 15–20% shift in D50 toward larger particle sizes, necessitating blade inspection at 500-batch intervals.Sterile Injectable Processing and the Lyophilization Envelope
Aseptic processing of (R)-N-Boc-3-pyrrolidineacetic acid into a sterile lyophilized injectable dosage form begins with dissolution in Water for Injection at a concentration of 10–50 mg/mL, followed by pH adjustment using 0.1 N sodium hydroxide or 0.1 N hydrochloric acid to a target range of 6.5–7.5. The solution is passed through a 0.22 μm PVDF membrane filter (Millipore Durapore) with a filtration pressure differential not exceeding 0.7 bar to prevent filter integrity failure. Sterility testing per USP 71 using direct inoculation on fluid thioglycollate medium and soybean-casein digest medium requires 14 days of incubation at 30–35°C and 20–25°C, respectively. The specification matrix for release of the injectable-grade material is compiled in the compliance checklist below.Table 2 — Injectable-grade release specification matrix for (R)-N-Boc-3-pyrrolidineacetic acid.| Test | Method | Acceptance Limit ||---|---|---|| Appearance | Visual inspection | White to off-white lyophilized cake || Chemical purity | HPLC, 210 nm | ≥ 98.0% area || Chiral purity | Chiral HPLC (Chiralpak AD-H) | ≥ 98.0% ee || Water content | USP 921 (Karl Fischer) | NMT 1.0% w/w || Bacterial endotoxin | USP 85 | NMT 0.25 EU/mg || Sterility | USP 71 | Meets requirements || Particulate matter | USP 788 Method 1 | NMT 6000 particles ≥ 10 μm per container || pH of reconstituted solution | USP 791 | 6.5–7.5 || Residual solvents | ICH Q3C | Per monograph || Elemental impurities | ICH Q3D | Per daily exposure limits |Lyophilization requires freezing at -40°C at a ramp rate of 0.5°C/min, primary drying at -20°C under a chamber pressure of 0.2 mbar for 24–36 h, and secondary drying at 25°C under 0.05 mbar for 6–8 h. The Boc protecting group remains intact throughout the lyophilization cycle. Terminal steam sterilization at 121°C for 15 min is not a viable alternative: published data for this specific configuration is limited, but measurable thermal deprotection of the Boc group is expected under these conditions based on the established acid-lability of the tert-butoxycarbonyl moiety. Aseptic processing is therefore the only permitted route for sterile manufacture. Particulate matter per USP 788 Method 1 (Light Obscuration) sets limits of NMT 6000 particles per container at ≥ 10 μm and NMT 600 particles per container at ≥ 25 μm for container volumes ≤ 100 mL. Residual moisture in lyophilized cakes is controlled at NMT 1.0% w/w by Karl Fischer titration per USP 921.When the Carboxylic Acid Terminus Engages in Amide Bond Formation
Activation of the carboxylic acid terminus with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC·HCl) at 1.1 equivalents in the presence of 1.1 equivalents of 1-hydroxybenzotriazole hydrate (HOBt) in anhydrous dichloromethane at 0–5°C for 2–4 h, followed by addition of the amine nucleophile, achieves isolated yields of 72–88% with enantiomeric excess retained at ≥ 98% ee as determined by chiral HPLC on a Chiralpak AD-H column with n-hexane/2-propanol (90:10 v/v) at 0.8 mL/min. Alternative activation with HATU (1.2 equivalents) and N,N-diisopropylethylamine (3.0 equivalents) in dimethylformamide at 20–25°C for 12–16 h is preferred when sterically hindered amine nucleophiles are employed. Removal of the Boc group subsequent to coupling proceeds quantitatively with a mixture of trifluoroacetic acid and dichloromethane (1:1 v/v) at 0–5°C for 30–60 min, generating the free secondary amine as its TFA salt. An alternative deprotection protocol uses 4 M hydrogen chloride in 1,4-dioxane at 0°C for 1–2 h to yield the hydrochloride salt directly. Racemization at the C3 stereocenter is not observed at temperatures below 40°C under these conditions. Exposure to ≥ 2.0 equivalents of strong base (e.g., sodium hydride) in aprotic solvents at temperatures above 60°C results in measurable epimerization of the α-carbon, reducing enantiomeric excess to 85–90% within 4 h. The deprotection reaction is monitored by thin-layer chromatography using silica gel 60 F254 plates with a mobile phase of dichloromethane/methanol (9:1 v/v). Coupling reactions are conducted under inert atmosphere (nitrogen or argon) to prevent oxidation of the liberated amine during work-up. Process-scale batches exhibit a 5–10% yield variation attributable to residual moisture in the solvent feed stream; solvent drying over activated 3Å molecular sieves for 24 h prior to use reduces this variability.Preserving the (R)-Configured Pyrrolidine Scaffold Across Multi-Step API Syntheses
During downstream syntheses in which the (R)-configuration at C3 of the pyrrolidine ring must survive multiple bond-forming operations, the Boc protecting group functions as a stereodirecting element that shields the ring nitrogen from undesired coordination and suppresses elimination side reactions. The compound is isolated as a white to off-white crystalline solid with a melting onset of 80–87°C determined by differential scanning calorimetry at a heating rate of 10°C/min under nitrogen. Specific optical rotation is reported on each batch Certificate of Analysis and typically falls within the range of -18° to -22° (c 1.0, methanol, 20°C). Chemical purity is specified at ≥ 98.0% by HPLC area normalization at 210 nm. Enantiomeric purity is specified at ≥ 98.0% ee, with the (S)-enantiomer controlled as the principal chiral impurity at NMT 1.0%. Process-related impurities including N-Boc-3-pyrrolidinecarboxylic acid and unreacted tert-butyl dicarbonate are monitored by LC-MS and controlled at NMT 0.5% individually. The compound is stored at 2–8°C in tightly sealed, nitrogen-flushed containers, with a retest period of 24 months when maintained under these conditions. In multi-step synthesis campaigns, the Boc group remains intact through Grignard additions conducted at -78°C, sodium borohydride reductions at 0–25°C, and palladium-catalyzed cross-coupling reactions at 60–80°C. The protecting group is removed only at the terminal deprotection step to release the pharmacologically active secondary amine. Published data for specific API configurations incorporating this intermediate is limited to sponsor Drug Master Files; the material is supplied under a Type II DMF with supporting CEP documentation from EDQM where applicable. The compound is compatible with standard reactor materials including glass-lined steel and 316L stainless steel; prolonged contact with copper or copper alloys in the presence of trace amines should be avoided because copper-catalyzed decomposition of the Boc group has been documented under reflux conditions.

More Introduction
(R)-N-Boc-3-pyrrolidineacetic acid, Pharma Grade API, CAS 204688-61-9, molecular formula C11H19NO4, molecular weight 229.27 g/mol, is supplied as a white to off-white crystalline powder. The molecule comprises a pyrrolidine ring carrying a pendant acetic acid group at C3 and a tert-butoxycarbonyl protecting group on the nitrogen; the (R) stereocentre is retained through downstream coupling and deprotection sequences only when temperature, pH, and residual moisture are controlled. Manufacture is performed under ICH Q7 and 21 CFR 210/211 conditions, with batch release covering infrared identification, high-performance liquid chromatographic purity, chiral purity, water content, residue on ignition, residual solvents, and assay. No harmonized USP or Ph. Eur. monograph exists for this protected amino acid; supplier specifications and validated in-house procedures therefore serve as the release basis. The pharma grade is intended for tablet, capsule, granule, and injectable dosage forms, but the route of administration determines the applicable microbial, pyrogen, and particulate limits. Published data for this specific configuration in aqueous injectable formulations is limited; forced-degradation work under ICH Q1A is required before selecting pH and thermal sterilisation conditions.
What Distinguishes the (R)-Boc-Protected Pyrrolidine Acetic Acid from the (S) Enantiomer and the Racemic Form?
The (R)-configured product differs from the (S)-enantiomer and the racemic form primarily by chiral identity. The (S)-enantiomer, CAS 204688-60-8, has identical molecular formula and mass but opposite optical rotation; in diastereomeric salt formation or chiral recognition, the two isomers can produce divergent crystallisation outcomes and biological activity when the stereocentre is preserved in the final active molecule. The racemic N-Boc-3-pyrrolidineacetic acid is a 50:50 mixture of (R)- and (S)-isomers; it is useful for chiral method development and racemic crystallisation screening but does not satisfy single-isomer release criteria. Compared with unprotected (R)-3-pyrrolidineacetic acid, the Boc-protected product reduces the nucleophilic reactivity of the pyrrolidine nitrogen and shifts solubility toward dichloromethane, methanol, and dimethyl sulfoxide rather than neutral aqueous buffers. Acid-catalyzed removal of the Boc group liberates the free acid; this conversion is exploited in downstream API preparation but must be minimised during drug product manufacture and storage.

In tablet and capsule manufacturing, the crystalline powder is milled in a conical mill under nitrogen to a particle size D90 ≤ 150 µm, measured by laser diffraction according to ISO 13320. Dry blending with microcrystalline cellulose, crospovidone, and magnesium stearate is performed in a bin blender at batch-specific rotation speeds; lubricant addition time is limited to 3–5 min because over-lubrication reduces tablet tensile strength and slows dissolution. Tablets are compressed on a rotary press equipped with 10 mm standard concave tooling; pre-compression force is maintained between 5 kN and 20 kN, and ejection force is monitored to remain below 800 N. Tablet hardness is measured according to USP 1217, friability according to USP 1216, disintegration according to USP 701, and dissolution according to USP 711. Wet granulation is performed in a high-shear mixer with impeller tip speed 5–10 m/s and chopper speed 1000–3000 rpm using an ethanol-water granulation fluid; granule end point is controlled by power-consumption rather than fixed time. The wet granules are dried in a fluid-bed dryer with inlet air temperature not exceeding 40°C and product temperature below 30°C because elevated temperature and residual moisture promote acid-catalysed Boc removal. Loss-on-drying before dry granulation is maintained at ≤ 0.5%; final granules for capsule or sachet use are controlled at water activity ≤ 0.6 at 25°C.
Capsule filling is performed on dosator or tamping-pin equipment, with fill weight calculated from batch bulk density and tapped density. Granule formulations for sachet packaging use a sieve-cut distribution of D10 ≥ 30 µm, D50 80–120 µm, and D90 ≤ 250 µm; this distribution reduces segregation during multi-dose container filling. Capsules are tested for disintegration per USP 701 and dissolution per USP 711 using a validated finished-product method.
Injectable processing of the protected API requires a vehicle compatible with the Boc group. The formulated solution is filtered through a 0.22 µm PVDF sterilising-grade membrane validated for bacterial retention according to ASTM F838-20; the filtered solution is lyophilized where aqueous stability is insufficient. Primary drying at shelf temperatures between −40°C and −20°C and secondary drying below 25°C are typical for similar Boc-protected amino acid derivatives; however, published data for this specific configuration is limited. Terminal steam sterilisation is not applied unless forced-degradation studies demonstrate ≤ 0.10% total impurities after the thermal cycle. Acidic vehicles below pH 2.0 are incompatible with storage because the tert-butoxycarbonyl group undergoes acid-catalysed deprotection. Nitrogen overlay is used to maintain residual oxygen in the final vial headspace below 5.0%. Container closure integrity is evaluated according to USP 1207, sterility according to USP 71, bacterial endotoxins according to USP 85, and particulate matter according to USP 788.
Release Specifications, Stability-Indicating Methods, and Pharmacopeial Alignment
The release programme applies general compendial chapters because no specific monograph exists for this protected amino acid. High-performance liquid chromatographic methods are validated for specificity, precision, linearity, accuracy, and robustness under ICH Q2(R1). Forced-degradation studies include acid, base, peroxide, heat, and light stress; the Boc group is the primary degradation site under acid and heat stress. Stability samples are evaluated at 25°C/60% RH and 40°C/75% RH according to ICH Q1A. Long-term storage is maintained at 2–8°C.

Bulk packaging is performed in ISO Class 8 cleanrooms compliant with ISO 14644-1. The product is double-bagged in low-density polyethylene, placed in aluminium-laminated fibre drums, and purged with nitrogen until residual oxygen is ≤ 5.0%. Desiccant is added when water content must remain ≤ 0.5% during storage and transport. Storage is maintained at 2–8°C; freeze-thaw cycles are avoided because condensation may initiate crystalline bridging and localised water uptake. Incompatibilities include strong mineral acids, acid-functionalised excipients, and prolonged contact with unbuffered aqueous solutions. Batch-to-batch particle size variance is controlled by milling through a 0.5 mm screen; larger screens increase D90 above 250 µm and reduce blend uniformity.
