Chinese gallnut-derived gallotannin (CAS 1401-55-4) is processed into drug tannate salts by controlled stoichiometric reaction with basic amine drugs in purified water at 45–55 °C in stainless-steel production reactors. The gallotannin solution is metered until residual free base in the mother liquor falls below 0.5% of the initial charge. The molar ratio of gallotannin to basic drug is held between 1.05:1 and 1.8:1; above 2.0:1, the filtered precipitate becomes tacky, extends vacuum tray-drying time beyond 24 h at 60 °C and 5 kPa, and shifts the particle-size distribution to a d90 above 500 µm. After drying and conical milling through a 0.5 mm screen, the tannate salt is blended with microcrystalline cellulose, pregelatinised starch, and colloidal silicon dioxide to a final tannate salt proportion of 30–65% w/w of the tablet core or capsule fill. Compression is performed on a rotary tablet press with precompression force 4–6 kN and main compression force 25–35 kN; tablet tensile strength is maintained above 1.2 MPa to avoid lamination during subsequent film coating. In-process controls include loss on drying at 105 °C not exceeding 3.0% and uniformity of dosage units according to USP <905>. Residual solvent testing follows USP <467>, elemental impurity control follows ICH Q3D, and finished-product dissolution testing is performed according to USP <711> apparatus 2 at 50 rpm for extended-release antihistamine and antitussive tannate tablets. Terminal finished dosage forms are film-coated tablets, scored tablets, and powder-filled hard gelatin capsules for oral administration.

Why does gallotannin content dictate tablet hardness and disintegration in albumin tannate formulations?

Albumin tannate products use gallnut-derived tannic acid as the pharmacologically active astringent component in wet-granulated tablets and granules. The mass ratio of tannic acid to albumin is controlled between 1:0.8 and 1:1.2; each finished tablet typically delivers 250–500 mg albumin tannate, corresponding to 125–250 mg tannic acid equivalents per unit. Granulation is performed in a top-spray fluid-bed granulator with inlet air temperature 55–65 °C, spray rate 10–20 g/min/kg, and atomising air pressure 1.0–1.5 bar. The tannic acid-albumin complex forms at pH 4.0–4.8 and yields granules with a bulk density of 0.42–0.55 g/mL. Granules are dried to a final moisture specification of 2.0–4.0% by loss on drying at 105 °C, lubricated with magnesium stearate at 0.3–0.5% w/w, and compressed at 10–18 kN. If gallotannin content is below 90% on the dried basis, non-complexing phenolic fragments increase, producing fines that cause capping at precompression force as low as 2.0 kN and hardness variability above 15% RSD. Compliance is anchored to the Japanese Pharmacopoeia monograph for albumin tannate, USP <701> disintegration in water at 37 °C, USP <905> uniformity of dosage units, and ICH Q3D for elemental impurity control. Terminal finished dosage forms include uncoated tablets, film-coated tablets, and granules filled into stick packs for oral suspension.

Where oxidation-sensitive APIs are formulated as tablets or hard capsules, gallic acid obtained from Chinese gallnut hydrolysis is incorporated as a free-radical chain-breaking antioxidant. The addition proportion is qualified in forced degradation studies at 40 °C/75% RH across a range of 0.005–0.20% w/w of the core mass. At 0.005% w/w the antioxidant effect is generally insufficient to reduce peroxide value after 6 months, and the degradation rate remains within 10% of an unprotected control. An incorporation level between 0.01% and 0.05% w/w is therefore selected for most oxygen-sensitive tablet cores. Above 0.2% w/w, the phenolic compound causes visible brown discoloration, increases tablet disintegration time by 30–45 s, and may complex with iron-containing excipients to form dark gray specks. The gallic acid is dissolved in the aqueous granulating fluid or sprayed as a 5% w/w solution during high-shear granulation at impeller speed 150–200 rpm and chopper speed 1500–2000 rpm; direct powder blending is avoided because the plate-like crystal habit produces segregation and variable assay results. The finished cores are preferably packaged in cold-form aluminium foil with a moisture vapour transmission rate below 0.5 g/m²/day. Compliance references include USP <467> for residual solvent control, ICH Q3D for elemental impurities, Ph Eur general monograph 2034 for tablets, and USP <905> for content uniformity. Terminal finished dosage forms are film-coated tablets, uncoated tablets, and hard gelatin capsules containing antioxidant-stabilised granules.

Effervescent Granule Processing: Acid-Source pH Windows and Moisture Control

Gallnut-derived tannic acid is used in effervescent granule and dry suspension systems as a weakly acidic astringent component and antioxidant; the standard effervescent couple consists of sodium bicarbonate, anhydrous citric acid, and tannic acid in a mass ratio of 2:1:0.3 to 2:1:0.8. The tannic acid content is limited to 10–20% w/w of the finished granule because higher concentrations increase hygroscopicity, raise the Hausner ratio above 1.35, and initiate premature carbon dioxide evolution during storage. Granulation is performed in a one-pot vacuum mixer-granulator at 35–45 °C and a residual moisture specification below 0.5% w/w. The dried granules are sieved through a 1.0 mm screen; oversize above 1.0 mm is dry-milled and recycled, but the recycle ratio is kept below 10% to avoid particle attrition. The final granules are packed into aluminium stick packs under nitrogen with silica-gel desiccant sachets. A 5% aqueous solution of the granule is controlled at pH 3.5–4.5, and effervescence time in 200 mL water at 20 °C is specified below 180 s. Compliance references include USP <701> disintegration for dispersible forms, USP <905> uniformity of dosage units, and ICH Q3C for residual solvent limits where ethanol is used in wetting. Terminal finished product types are effervescent granules, dry syrups, and dispersible tablets intended for oral administration.

In parenteral formulation development, oxidation-sensitive active substances have been screened with gallic acid derived from Chinese gallnut as a water-soluble antioxidant and metal-chelating agent. The concentration range explored in published reports falls between 0.005% and 0.05% w/v of the injectable solution. Published data for this specific configuration is limited, and each formulation must be qualified by forced degradation at 40 °C with 21% headspace oxygen over 14–28 days. Above 0.1% w/v, free phenolic groups can bind to proteinaceous APIs or amino acid additives and generate subvisible particles after steam sterilisation. The manufacturing process requires dissolving gallic acid in Water for Injection at 20–30 °C under nitrogen sparging, adjusting the solution pH to 4.5–6.0 with dilute sodium hydroxide, and bubbling nitrogen until dissolved oxygen remains below 0.5 mg/L. The solution is filtered through a 0.22 µm polyethersulfone membrane and filled aseptically into Type I borosilicate glass vials. Terminal sterilisation at 121 °C for 15 min is evaluated only if pilot stability studies confirm no pH drift above 0.3 pH units and no colour increase above 5 units on the Ph Eur colour scale. Compliance references include USP <1> injections, USP <788> subvisible particulate matter, ICH Q3D, and Ph Eur 5.11 parenteral preparations. Terminal product types include aqueous injectable solutions and lyophilised powders for reconstitution as parenteral preparations.

    More Introduction

Chinese Gallnuts Pharma Grade API is prepared from the desiccated gall excrescence produced by Schlechtendalia chinensis on Rhus chinensis petioles. The material is standardised as a hydrolysable tannin fraction for tablet, capsule, granule, injection, oral, and injectable dosage forms. Supplier model codes generally encode the minimum gallotannin content determined by the hide-powder assay adapted from Ph. Eur. 2.8.14: CGT-90, CGT-95, and CGT-98 correspond to no less than 90.0%, 95.0%, and 98.0% gallotannins on the dried substance. The material is an amorphous, pale-cream to light-tan powder. Solid-dose grades are milled to a laser-diffraction D90 below 150 µm for direct blending, while injectable grades are further processed for low endotoxin and low bioburden. The product is a mixture of polygalloylglucose esters with molecular weights ranging from approximately 940 g/mol for pentagalloylglucose to above 1,700 g/mol for decagalloylglucose. This distribution, not free gallic acid content alone, governs protein-binding capacity, astringency, and aqueous hydrolysis behaviour.

Raw gallnut lots vary in gallotannin content depending on growing altitude, harvest time, and aphid population density. Manufacturer blending and normalisation are necessary to bring final API lots within specified monograph limits. Suppliers monitor gallic acid-to-tannin ratio, glucose release after hydrolysis, and polyphenolic molecular weight distribution. The extraction process usually uses hot water or aqueous alcohol followed by filtration, decolourisation, concentration, and spray-drying or vacuum-drying. Spray-dried powder tends to have a bulk density of 0.35–0.50 g/mL, lower dusting, and faster aqueous dispersion; vacuum-dried material is denser and less hygroscopic but may require additional milling. Batch-to-batch variability in residual glucose and galloylation degree is controlled by normalisation and not by terminal blending alone.

Release Limits and Chromatographic Identity for the Hydrolysable Tannin Fraction

The release profile is controlled by orthogonal methods because the hide-powder titration captures total gallotannins but does not resolve the galloylation degree. A representative specification is provided for development use; exact limits are defined by the purchaser–supplier quality agreement and the applicable pharmacopoeial monograph. For oral and injectable development, the following tests are applied.

    Parameter Representative limit Method
    Model designation CGT-90 / CGT-95 / CGT-98 Supplier lot documentation
    Gallotannins, dried substance ≥90.0% / ≥95.0% / ≥98.0% Ph. Eur. 2.8.14 hide-powder assay, adapted

Loss on drying ≤5.0% USP <731> / Ph. Eur. 2.2.32
Sulfated ash ≤1.0% Ph. Eur. 2.4.14
Elemental impurities ≤10 ppm total heavy metals ICH Q3D / USP <233>
Residual solvents Class 3 only Ph. Eur. 5.4
Total aerobic microbial count <1,000 CFU/g Ph. Eur. 2.6.12
Bacterial endotoxins, injectable grade <0.25 EU/mg Ph. Eur. 2.6.14

Free gallic acid is the critical purity marker because it indicates premature hydrolysis during extraction, storage, or drying. HPLC is performed on an octadecylsilyl column with a water–acetonitrile mobile phase containing 0.1% trifluoroacetic acid and ultraviolet detection at 270 nm. The free gallic acid limit prevents lot-to-lot variation in aqueous solubility, colour, and protein binding from reaching the finished dosage form. For injectable development, the endotoxin specification replaces the non-sterile oral limit, and particulate matter is assessed on the finished injection rather than on the API.

Oral dosage forms of Galla chinensis-derived extract are used in astringent, antidiarrheal, and antimicrobial preparations; the Chinese Pharmacopoeia 2020 edition lists Galla chinensis as a traditional medicine input. The API is incorporated into tablets by wet granulation, into capsules by filling milled granules, and into granules for oral suspension by fluid-bed spraying onto sucrose or mannitol carriers. Each route requires a different particle-size and moisture specification: oral granules may permit a wider particle-size band, whereas injectable grade is lyophilised and must be free of visible foreign matter.

What Limits Aqueous Stability in Injectable Formulations?

Hydrolysable tannins undergo pH-dependent ester hydrolysis. At pH above 6.0 and at autoclave temperatures above 121 °C, polygalloylglucose esters degalloylate to free gallic acid and lower-molecular-weight oligomers. Parenteral processing therefore avoids terminal steam sterilisation of aqueous solutions when the molecular-weight distribution is critical. A lyophilised powder is prepared by dissolution in Water for Injection at pH 4.0–5.0, sterile filtration through a low-protein-binding membrane, filling, and freeze-drying. The solution is nitrogen-blanketed because dissolved oxygen accelerates oxidative darkening and free-radical-mediated degradation.

Sterile filtration must be qualified for adsorptive losses. Nylon and certain polyvinylidene fluoride membranes can remove polyphenolic fractions; polyethersulfone or polytetrafluoroethylene filters are evaluated according to PDA Technical Report 26. Injectable-grade material such as CGT-98-I is controlled for total aerobic microbial count below 100 CFU/g, bacterial endotoxin below 0.25 EU/mg, and subvisible particulate matter compatibility per USP <788>. The API is not autoclaved; depyrogenation is applied to containers and components. In formulation, avoid combination with ferric iron, alkaloid drugs, and protein-based excipients unless compatibility studies are performed because gallotannins form flocculent precipitates or blue-black ferric complexes. Once reconstituted, the aqueous solution should be used within 8 h and stored below 25 °C to limit gallic acid formation.

When High-Shear Wet Granulation Reaches the Overwetting Threshold

Wet granulation is used to improve tablet content uniformity, but the process window narrows because gallotannin fractions bind water rapidly. In a 25 L high-shear granulator with impeller speed of 200–300 rpm and chopper speed of 1,500 rpm, water addition beyond approximately 8–10% w/w of dry granulate can produce an overwetted mass that sticks to the bowl wall and yields non-uniform granule growth. Typical practice is to use a binder solution of 3–5% povidone or hydroxypropyl methylcellulose and to add the binder over 2–3 min under impeller load monitoring. The wet mass is dried at 45–50 °C in a fluid-bed dryer to a final loss on drying below 3.0%. Drying above 60 °C is avoided because thermal degalloylation can raise free gallic acid and darken the granulate. Milled granules are blended with 0.5–1.0% magnesium stearate and 0.5–1.5% colloidal silicon dioxide before compression.

For capsule filling, granules are filled into hydroxypropyl methylcellulose shells rather than high-moisture gelatin shells when storage humidity exceeds 60% RH; gallotannins can cross-link gelatin under accelerated conditions and delay dissolution. Dissolution of finished tablets should be evaluated per USP <711> using apparatus II at 50 rpm in 0.1 N hydrochloric acid or water, but the method must be product-specific. Hot-melt extrusion above 100 °C is generally avoided because the gallotannin ester bonds are thermally labile; published data for hot-melt extrusion of Chinese gallnut API is limited, and preformulation stability data are required before selecting a lipid carrier or solid dispersion platform.

Comparing Chinese Gallnut API With Turkish Gallnut and Synthetic Gallic Acid

Chinese gallnut API differs from Turkish gallnut extract and synthetic gallic acid in compositional profile and processing risk. Turkish gallnut extract contains a higher ellagitannin fraction; acid hydrolysis releases ellagic acid, which is poorly soluble and can precipitate in aqueous formulations. Synthetic gallic acid is a single molecular entity that does not reproduce the multi-site protein binding of polygalloylglucose esters.

Parameter Chinese Gallnut API Turkish Gallnut Extract Synthetic Gallic Acid
Source Insect gall on Rhus chinensis Oak gall formed on Quercus species Chemical synthesis or hydrolysis
Major constituents Gallotannins, polygalloylglucose esters Gallotannins plus ellagitannins Monomeric trihydroxybenzoic acid
Hydrolysis products Glucose and gallic acid Glucose, gallic acid, and ellagic acid Not applicable; monomer
Protein-binding behaviour Multi-site, pH-dependent, high capacity Multi-site but affected by ellagitannin content Single-site hydrogen bonding
Key specification risk Free gallic acid, residual moisture, endotoxin Ellagic acid precipitation, source variability Purity and residual synthesis reagents

These compositional differences are relevant in manufacturing when a formulation requires high gallotannin content without the solubility risk of ellagic acid. For injectable applications, the CGT-98-I grade should be stored below 25 °C, protected from light, and used within 8 h after reconstitution; oral granules should be packaged in double polyethylene-lined fiber drums with desiccant because the product darkens and clumps at relative humidity above 60%. Avoid contact with uncoated carbon steel and copper equipment, and use 316L stainless steel or fluoropolymer-lined contact surfaces during manufacturing.
