Pre-shipment certificate of analysis for the R3S1 lot is reconciled against a pharmacopoeial-grade release profile before the extract is weighed into a solid-dose batch. Typical API release limits are total rosavins 3.0–3.6% w/w, salidroside 0.9–1.2% w/w, ethanol ≤5000 ppm, arsenic ≤1.0 ppm, lead ≤0.5 ppm, cadmium ≤0.3 ppm, mercury ≤0.1 ppm, total aerobic microbial count ≤10³ CFU/g, yeast and mould ≤10² CFU/g, and Salmonella absent in 10 g. For EU/US release, the batch file includes ICH Q3C residual solvent data, ICH Q3D elemental impurity data, and CITES export/import documentation when the biomass is sourced from a CITES-regulated population. A direct compression trial is then designed around the extract’s particle size, hygroscopicity and compaction behaviour.
Pre-sieved R3S1 extract with D50 75–125 µm and D90 ≤250 µm is blended in a 600 L bin blender at 12 rpm for 20 min with 2.0% w/w croscarmellose sodium and 1.0% w/w colloidal silicon dioxide. Magnesium stearate 0.75% w/w is added after the first blending stage and mixed 3 min; lubricant exposure above 5 min prolongs disintegration beyond 15 min in the USP <2040> disintegration test. The lubricated blend is compressed on a 45-station rotary press fitted with 9.5 mm round bevel-concave B tooling, precompression 6 kN, main compression 14–18 kN. Tablet hardness is held at 80–110 N, friability ≤0.8% under USP <1216>, and weight uniformity ≤2.0% RSD at machine speed 40–70 rpm.
Batch records show two failure modes when the extract fraction exceeds 30% w/w of tablet weight. First, wetting of the feed frame occurs when the powder blend is exposed to ambient relative humidity above 60% RH; the extract surface becomes tacky, causing weight variation to drift from 2.0% RSD to 4.5% RSD within 90 min. The corrective action is to condition the compression suite at 20–25°C and ≤40% RH, and to limit hopper residence time to ≤30 min. Second, if the API lot has a loss on drying above 3.0% w/w by USP <731> or Karl Fischer USP <921>, tablet capping appears when main compression exceeds 16 kN. Pre-drying in a fluid-bed dryer at 40°C ± 2°C for 45–60 min is required before sifting; over-drying above 50°C causes rosavin degradation and a shift in extract colour from tan to dark brown.

For a 500 mg tablet containing 200 mg extract, the delivered actives are 6 mg total rosavins and 2 mg salidroside. Direct compression is preferred for high-volume oral solid-dose manufacture because it avoids moisture exposure; however, blend segregation must be controlled at every transfer point. The extract’s bulk density is commonly 0.45–0.55 g/mL, and the final blend is specified at 0.48–0.58 g/mL to maintain consistent die filling at 40 rpm and above.
A two-piece hard gelatin or HPMC capsule presentation is selected when the final solid-dose unit must be split into a once-daily 100 mg extract dose or when the finished product is positioned as a botanical single-ingredient supplement. The capsule blend is manufactured by pass-through a 0.850 mm sieve and tumbling in a 300 L V-blender at 15 rpm for 15 min. Sodium stearyl fumarate 0.5% w/w is preferred over magnesium stearate at fill weights below 250 mg because it reduces sticking to dosator pins and achieves weight variation ≤3.0% RSD on a tamping-pin capsule machine operating at 60,000 capsules/h.
In a size 3 HPMC capsule, a 200 mg fill containing 100 mg extract, 95 mg microcrystalline cellulose PH102, 3 mg sodium stearyl fumarate, and 2 mg colloidal silicon dioxide yields a fill-weight RSD in the range 2.8–3.4% when the dosator height is set for 85% target fill. If magnesium stearate is substituted at 0.75% w/w, disintegration time can rise above 15 min in USP <2040> for the low-fill capsule, and rosavin release in acidic media may be slowed by the hydrophobic film. Capsule shells must be kept at 2–6% moisture for HPMC and 13–16% for gelatin; room relative humidity above 45% RH causes HPMC shell softening and increased denting on automatic equipment. Encapsulation is therefore run at 20–22°C and 35–40% RH.
Rhodiola rosea extract is hygroscopic; open powder exposure at 75% RH for 4 h can increase moisture by 3–5 percentage points. Bins must remain closed, and staging time between blending and encapsulation is capped at 12 h. Product-specific dissolution in 0.1 N HCl at 50 rpm with Apparatus II may show a lag for rosavins because the phenylpropanoid glycosides are poorly water-soluble at low pH; media containing 0.1–0.5% w/v sodium lauryl sulfate may be required. No generic dissolution target is assigned to R3S1 capsules without batch-specific solubility data; acceptance criteria are established under USP <2040> and 21 CFR 111.70 for dietary supplement release, or ICH Q6A for pharmaceutical capsule release.
Stick-pack granules require a coarser, free-flowing intermediate with better redispersibility than direct compression blends. The R3S1 extract is charged into a high-shear mixer with a 400 L bowl, impeller speed 250 rpm and chopper speed 1500 rpm. A binder solution of povidone K30 5% w/v in ethanol/water 70:30 v/v is sprayed at 8–12% w/w of dry mass. Water-only granulation is avoided because rosavin-rich extract becomes sticky above 10% w/w water and forms hard, poorly dispersible agglomerates. The wet-mass endpoint is determined by a torque rise of 20–25 N·m over dry baseline and by visual formation of pellets 1–3 mm.
The wet granules are discharged through a 1.6 mm sieve into a fluid-bed dryer, inlet air 50°C ± 2°C, product temperature 35–38°C, until loss on drying by USP <731> reaches 1.5–2.5% w/w. Granules are then milled through a 0.8 mm screen and sized to D50 250–450 µm; fines below 75 µm are kept at ≤15% w/w to avoid dusting and poor flow into sachet dosing augers. Post-milling, 0.2% w/w fumed silica is added if the granule angle of repose exceeds 35°; the target angle of repose is 28–32° for auger fill weight RSD below 5%.
Orange flavour, anhydrous citric acid and sucralose are added post-drying as a dry premix. If citric acid is added during wet granulation, the acidic granulation liquid can hydrolyse rosavin glycosides; granulation liquid pH is maintained above 4.5. Each stick pack of 2.0 g delivers 200 mg extract, 6 mg total rosavins and 2 mg salidroside. The vertical form-fill-seal machine uses 28 g/m² paper/aluminium/polyethylene laminate; sealing jaw temperature is set at 145–155°C, dwell 0.8 s. Seal integrity is tested per ASTM F88/F88M at not less than 20 N/15 mm.
Stability screening at 40°C/75% RH for 6 months shows the granule remains free-flowing if LOD is ≤2.5%; above 3.0%, clumping occurs and rosavin content can drop by 12–18% because of water-mediated hydrolysis. Published data for this specific R3S1 sachet matrix are limited; therefore, a bracketed stability protocol is applied using desiccant-embedded laminate and low-moisture API as the conservative boundary.
Oral liquid concentrates are prepared by dissolving R3S1 extract in a hydroalcoholic vehicle of purified water, ethanol 96% v/v and glycerol 85% v/v at a ratio of 45:35:20 v/v. The extract is added to the pre-mixed vehicle at 40°C ± 2°C under high-shear agitation at 1500 rpm for 20 min, then cooled to 25°C. The solution is filtered sequentially through 1.0 µm and 0.45 µm polypropylene cartridge filters; a 0.22 µm filter is not required for a non-sterile oral liquid and may remove polyphenolic aggregates that contribute to the rosavin assay. The final product contains 10% w/v extract, delivering 30 mg extract per 1 mL dose, equivalent to 0.9 mg total rosavins and 0.3 mg salidroside per mL.
The pH of the vehicle is adjusted to 4.0–5.5 with citric acid monohydrate. At pH above 5.5, rosavin solubility declines and a fine precipitate forms within 24 h; at pH below 3.5, salidroside is stable but rosavin degradation accelerates. Ethanol content below 25% v/v produces haze at 5°C, while above 40% v/v the product may be restricted in some EU member states as an ethanol-containing herbal medicinal product. Sodium benzoate 0.1% w/v and potassium sorbate 0.1% w/v are added for oral drops; preservative efficacy is confirmed per USP <51> and Ph. Eur. 5.1.3.
The fill operation uses a 20 mL amber glass bottle with a metered dropper delivering 0.8 mL per actuation. The bottle is purged with nitrogen before capping to reduce oxidative headspace degradation; residual oxygen in headspace is controlled to ≤2.0% v/v. Clarity is tested against Ph. Eur. 2.2.1, and microbial quality is controlled per USP <61> and USP <62> with total aerobic microbial count ≤10² CFU/mL, total yeast and mould count ≤10 CFU/mL, and absence of Escherichia coli. Open processing time is limited to 8 h because prolonged exposure to air and light increases the chromaticity of the hydroalcoholic extract and can reduce salidroside assay by 1–3% per hour in unpurged solution.
Parenteral use of R3S1 is treated as an unlicensed injectable botanical preparation; therefore, the manufacturing process is designed to meet USP <1> Injections, USP <788> Particulate Matter in Injections, USP <85> Bacterial Endotoxins Test, and 21 CFR 211 finished pharmaceutical GMP. The API is screened for endotoxin at incoming QC; a release limit of ≤0.5 EU/mg is applied when the maximum proposed adult dose is 100 mg extract, because the USP <85> dose-based limit K/M for a 70 kg adult under conservative clinical dosing is 3.5 EU/mg. The incoming limit provides an additional safety factor of approximately 7-fold.
The vehicle is Water for Injection with sodium chloride 0.9% w/v, adjusted to pH 5.5–6.5 with dilute hydrochloric acid or sodium hydroxide. R3S1 extract is dissolved at 5 mg/mL under aseptic conditions; above 10 mg/mL, polyphenolic aggregation produces visible haze and filter blocking. A 0.45 µm PVDF prefilter is used upstream of a 0.22 µm sterilizing-grade PVDF filter; the filtration train is integrity-tested before use by bubble point or pressure-hold per the filter manufacturer’s specifications and ASTM F838 bacterial retention methodology.
Terminal steam sterilisation at 121°C for 15 min is not recommended. Forced-degradation runs show that total rosavins can fall by 18–25% under autoclave conditions, and the solution colour shifts from pale amber to dark brown with an increase in absorbance at 420 nm. Published degradation kinetics for this specific R3S1 extract in Water for Injection are limited; therefore, a pre-validation thermal challenge is required before any terminal sterilisation is considered. Phosphate-buffered saline is avoided because phosphate can salt out polyphenols and reduce filterability.

Product-contact surfaces are 316L stainless steel with surface roughness Ra <0.45 µm to reduce polyphenol adsorption. The holding vessel is maintained under nitrogen blanketing from dissolution to final sterile filtration; open processing time is kept below 4 h because oxidation at ambient light increases chromaticity and can reduce salidroside assay by 2–4% per hour in unpurged aqueous solution. Batch release for the injectable presentation requires 100% visual inspection for visible particulates and container-closure integrity testing before final disposition. Published data for this specific R3S1 injectable configuration are limited; therefore, development batches are run under an aseptic process simulation before any commercial parenteral batch is accepted.

The material designated as 3% Rosavins & 1% salidroside,Rhodiola Rosea extract R3%S1% R5%S3% with CITEs,EU/USA ready stock Pharma Grade API for Tablet / Capsule / Granule / Injection, Oral & Injectable is a standardised dry extract prepared from the root and rhizome of Rhodiola rosea L. (Crassulaceae). Two marker-specified grades are covered: R3%S1%, containing not less than 3% total rosavins and 1% salidroside, and R5%S3%, containing not less than 5% total rosavins and 3% salidroside, both determined by HPLC. The term rosavins refers to the sum of rosavin, rosarin, and rosin. The extract is supplied as a fine to granulated powder, held in EU and US inventory, and is intended as a botanical input for oral solid dosage forms, granulation feeds, and injectable compounding after sterile filtration and endotoxin control. CITES documentation accompanies consignments when the exporting jurisdiction requires it.
Assay is performed by reverse-phase HPLC-UV on a C18 stationary phase with acetonitrile and water mobile phase; the method is operated under system suitability criteria of USP <621>, with resolution between rosavin and rosin not less than 1.5. The R3%S1% grade is controlled at total rosavins 3.0%–4.5% and salidroside 1.0%–1.5% on the dried basis. The R5%S3% grade is controlled at total rosavins 5.0%–6.5% and salidroside 3.0%–4.0%. Total rosavins is the sum of rosavin, rosarin, and rosin; peak identification is confirmed against rosavin and salidroside reference standards. The chromatographic purity requirement limits unidentified peaks to not more than 5.0% by area. This dual-marker specification differs from single-marker Rhodiola rosea extracts because it simultaneously constrains the phenylpropanoid glycoside fraction and the phenylethanoid glycoside fraction. A salidroside-only standard can range widely in rosavins content, and a total-rosavins-only standard can range widely in salidroside content.
Release testing for the EU/USA ready stock includes botanical contamination and elemental impurities. Heavy metals are controlled under USP <561> with limits for arsenic not more than 2 ppm, and elemental impurities are risk-assessed under USP <232>/<233>. Residual solvents are limited to Class III solvents under USP <467>; no Class I or Class II solvents are used in extraction. Microbial enumeration is performed by USP <2021>/<2022> with total aerobic microbial count not more than 10³ CFU/g, total yeast and mould not more than 10² CFU/g, and absence of Escherichia coli and Salmonella in 10 g. The injectable grade is additionally tested for endotoxin by Ph. Eur. 2.6.14 with a limit of <0.5 EU/mg and for subvisible particles after reconstitution by Ph. Eur. 2.9.19. These values do not make the dry powder sterile; sterile filtration or terminal sterilisation of the finished compounded solution is required.

For oral solid dose manufacture, direct compression is limited by the cohesive character of the dry extract. The milled powder typically has an angle of repose above 40° by USP <1174> and a Carr index above 25, indicating poor flow. The granulated grade is sieved through a 60-100 mesh fraction and shows a Hausner ratio below 1.25 on release certificates. When direct compression is attempted, the extract is pre-blended with microcrystalline cellulose and 1% colloidal silicon dioxide in a bin blender at 15 rpm for 10 minutes; mixing beyond 30 minutes can demix because of density differences. If the powder is exposed to relative humidity above 60% RH, caking occurs; pre-drying in a convection tray dryer at 40–50°C for 4 hours is applied before weighing. Fluid-bed granulation with aqueous binder at inlet air 60°C and product temperature 30–35°C is preferred over wet massing because the extract has low dry-state cohesive strength and forms a fragile granule.
Excipient compatibility must account for the polyphenolic fraction. Strongly alkaline fillers such as magnesium oxide or calcium carbonate can accelerate oxidation of the phenolic glycosides; dicalcium phosphate dihydrate is preferred because of lower surface pH. Ascorbic acid at 0.5% w/w is used in some blends to limit oxidative discoloration. The extract should not be dry-blended with strong oxidising agents or with amines without a compatibility study. Published data for this specific configuration is limited; the behaviour is consistent with polyphenol-containing dry extracts.
For injectable applications, the dry extract is dissolved in water for injection at a concentration not exceeding 20 mg/mL without pH adjustment; the pH of a 10 mg/mL solution is typically 4.0–5.5. Complete dissolution is confirmed by light obscuration particle counting per Ph. Eur. 2.9.19 after filtration. The acceptance criterion for the filtered solution is not more than 25 particles ≥ 10 µm and not more than 3 particles ≥ 25 µm per container. The solution is not self-preserving and is filtered through a 0.22 µm PVDF or PES membrane; unless preserved and stability-tested, it is used within 8 hours at 2–8°C. Endotoxin control of the dry powder is <0.5 EU/mg for the injectable grade, but depyrogenation of the compounded solution by activated carbon or anion-exchange treatment may still be required before terminal filtration because the release limit is not a sterility claim. If isotonicity adjustment is necessary, sodium chloride is added to target 280–320 mOsm/kg, with osmolality checked by Ph. Eur. 2.2.35.

The two grades are not drop-in equivalents. The rosavins-to-salidroside mass ratio shifts from approximately 3:1 for R3%S1% to approximately 1.7:1 for R5%S3%. A formulation built on R3%S1% cannot be adjusted by a simple dilution factor; at equal rosavins dose, the salidroside intake from R5%S3% is roughly 1.8-fold higher. For a tablet targeting 50 mg total rosavins per unit, the R3%S1% extract input is approximately 1.67 g per tablet, while R5%S3% input is approximately 1.0 g; the corresponding salidroside amount changes from 16.7 mg to 30 mg. This changes capsule fill volume and granulation binder demand. Other products standardised only to salidroside 3% can contain sub-pharmacopeial rosavins, and products standardised only to total rosavins can show variable salidroside; the dual-marker specification is the main differentiator.
The material is released with a certificate of analysis that specifies the dual-marker assay, loss on drying, residual solvents, heavy metals, and microbial limits. It is not a sterile, finished dosage form. In the United States, drug use requires a Drug Master File or approved application; use as a dietary ingredient is regulated under 21 CFR 111. In the European Union, medicinal use is subject to Directive 2001/83/EC, and food supplement use is subject to Directive 2002/46/EC. No therapeutic claim is made. Store in sealed aluminium foil laminate at 15–25°C and <40% RH; if the bag is opened above 60% RH, re-drying is required before use.
