Application of 2-Amino-1,3-Propanediol; Serinol Pharma Grade API for Tablet / Capsule / Granule / Injection, Oral & Injectable
At the largest-volume downstream process for pharmaceutical-grade serinol, the material is consumed in the synthesis of non-ionic monomeric iodinated X-ray contrast agents, specifically iopamidol, iohexol, iomeprol, iopromide, and ioversol. The convergent manufacturing route reacts 5-amino-2,4,6-triiodoisophthalic acid dichloride with serinol in N,N-dimethylacetamide or 1,3-dimethyl-2-imidazolidinone at 45–55°C with sodium carbonate as acid scavenger. Reaction pH is held at 9.0–10.0 to balance acid chloride hydrolysis against free amine availability. For iopamidol, iomeprol, iopromide, and ioversol, the stoichiometric requirement is 2.0 mol serinol per mole of triiodinated isophthalic acid derivative; for iohexol the requirement is 3.0 mol because the 5-position N-(2,3-dihydroxypropyl)acetamido substituent is also constructed from serinol. Industrial charging is 2.05–2.20 mol for the bis-substituted monomers and 3.05–3.30 mol for iohexol, with the surplus consumed by acid chloride hydrolysis before complete amidation. Production equipment includes glass-lined reactors of 2.5–5.0 m³ working volume, Hastelloy C-276 agitation shafts, and 0.45 µm polytetrafluoroethylene filtration cartridges for crude clarification. After amidation, the API is dissolved in water for injection, clarified through 0.45 µm prefilters, and purified by anion- and cation-exchange chromatography to remove residual serinol and inorganic salts. Crystallization from ethanol/water at 50–60°C and vacuum drying at ≤10 kPa and ≤60°C yield a dry powder in which free serinol is controlled under ICH Q3A as a specified impurity at not more than ≤0.10% area by HPLC. Release testing uses USP <621> liquid chromatography, USP <788> particulate matter in injections, USP <85> bacterial endotoxins, USP <71> sterility, 21 CFR 211.67 equipment cleaning, and the applicable USP/Ph. Eur. Iohexol Injection and Iopamidol Injection monographs. Serinol is not present as a direct compression excipient in tablet or capsule matrices in this route; it is consumed during API synthesis. Terminal finished product types include injectable solutions at 200–370 mg I/mL, pre-filled syringes, and 50 mL, 100 mL, 200 mL, and 500 mL glass vials.

Does the Dimeric Route Change Serinol Stoichiometry and Solvent Handling?

In dimeric iso-osmolar contrast synthesis, serinol is charged at 4.0 mol per mole of the 5,5'-[(2-hydroxytrimethylene)bis(acetylimino)]bis(2,4,6-triiodoisophthalic acid) intermediate used for iodixanol. The charge is split into two amidation blocks of 2.0–2.1 mol per triiodinated ring, introduced before and after formation of the 2-hydroxytrimethylene bridge. This split-charge sequence reduces formation of crosslinked oligoamide impurities that occur when a single primary amine reacts with two acid chloride groups. Process analytical technology is required because the dimeric intermediate shows lower solubility in N,N-dimethylacetamide than monomeric precursors; the reaction solvent is shifted to 1,3-dimethyl-2-imidazolidinone or N-methylpyrrolidone at 30–50°C to maintain homogeneity. Residual serinol is monitored by charged aerosol detection HPLC after each amidation block and again before final recrystallization, with the release limit aligned to ICH Q3A at ≤0.10% area. Downstream isolation includes nanofiltration through 200 Da molecular weight cut-off membranes, anion- and cation-exchange polishing, and crystallization from ethanol at ≤55°C. Release standards include USP Iodixanol Injection, the current Ph. Eur. monograph, ICH Q7, ICH Q3C residual solvent testing by USP <467>, and 21 CFR 211.84 for incoming raw material identity and purity. The terminal finished product is an iso-osmolar injectable solution at 270–320 mg I/mL in 50 mL and 100 mL Type I glass vials, with osmolality approximately 290 mOsm/kg and intended for angiographic and CT procedures where reduced osmotic load is clinically relevant.

Intrathecal Iopamidol Injection — Serinol-Derived API Requirements

Because intrathecal injection introduces the finished solution directly into cerebrospinal fluid, the manufacturing controls for iopamidol are more restrictive than those for intravascular contrast products. Serinol is not present as an excipient in the final formulation; its function is as the starting material in the amidation of 5-(hydroxyacetylamino)-2,4,6-triiodoisophthalic acid, where it is charged at 2.0–2.2 mol per mole of intermediate. After synthesis, iopamidol is crystallized twice from aqueous ethanol and filtered through 0.22 µm sterilizing-grade membranes. The formulated product contains iopamidol at 200 mg I/mL or 300 mg I/mL, tromethamine hydrochloride as buffer, edetate calcium disodium as chelator, and water for injection; pH is adjusted to 6.5–7.0. Terminal sterilization is performed at 121°C for 15 min, validated by USP <71> sterility testing and USP <85> bacterial endotoxin testing with an endotoxin limit of <0.5 EU/mL. Applicable standards include USP <1> Injections, USP <788> Particulate Matter in Injections, USP <660> glass containers, 21 CFR 211.167 process validation, and the current USP Iopamidol Injection monograph. Filling is performed in Grade A isolators with 0.45 µm bioburden prefiltration followed by 0.22 µm sterilizing filtration; open-ampoule filling is avoided because glass microparticulates present a specific risk in cerebrospinal fluid. Terminal finished product types are single-dose 10 mL and 20 mL vials for lumbar, thoracic, and cervical myelography. When serinol-derived iohexol or iopamidol is administered orally or rectally for CT bowel opacification, the manufacturing process shifts from terminal sterilization to bioburden control and container compatibility. Adult oral iohexol solution is diluted to 12–21 mg I/mL; pediatric dilutions are set at 9–18 mg I/mL based on body weight. The upstream API synthesis still consumes serinol at 3.0–3.3 mol for iohexol and 2.0–2.2 mol for iopamidol per mole of triiodinated precursor. For ready-to-use oral contrast, the API solution is diluted with purified water, pH is adjusted to 6.0–7.5 with tromethamine hydrochloride, and the solution is passed through 0.45 µm filters before filling into unit-dose polypropylene cups or amber glass bottles. Because the product is non-parenteral, terminal sterilization is generally omitted; bioburden is controlled at ≤100 CFU/mL, and microbial challenge studies are used to establish the hold time. Published data for exact time-temperature pasteurization parameters in this specific serinol-derived oral contrast configuration is limited. Compliance relies on 21 CFR 211, ICH Q3D elemental impurity testing by USP <232> and USP <233>, USP <905> uniformity of dosage units for unit-dose containers, and USP <467> residual solvents. Terminal finished product types include ready-to-use oral contrast solutions, single-dose oral cups, and rectal enema solutions.

Serinol-Based Linker Phosphoramidite Chemistry in Oligonucleotide Manufacturing

In oligonucleotide manufacturing, serinol is converted into a non-nucleosidic phosphoramidite linker in which the primary amino group is protected with 9-fluorenylmethoxycarbonyl or trifluoroacetyl and the two hydroxyl groups are sequentially protected with 4,4'-dimethoxytrityl and cyanoethyl N,N-diisopropylphosphoramidite. This linker is coupled onto controlled pore glass or polystyrene supports at 0.1 M in anhydrous acetonitrile using 0.3 M 5-benzylthio-1H-tetrazole as activator. Coupling is performed for 180–300 s at 20–25°C; coupling efficiency is measured by DMT cation assay and must exceed 98.0% per cycle before chain elongation continues. The support loading for serinol-derived linkers is typically 200–400 µmol/g. Downstream solid-phase synthesis uses detritylation with 3% trichloroacetic acid in dichloromethane, oxidation with 0.02 M iodine in tetrahydrofuran/pyridine/water, and capping with acetic anhydride/lutidine/N-methylimidazole. Cleavage and deprotection use 30% ammonium hydroxide at 55°C for 10 h, followed by ion-exchange or ion-pair reverse-phase HPLC purification and tangential-flow ultrafiltration. Compliance is governed by ICH Q7, 21 CFR 210/211 for injectable oligonucleotide drug products, USP <621> HPLC, and the FDA Guidance for Industry: Chemistry, Manufacturing, and Control of Oligonucleotide Therapeutics; residual solvent testing is performed according to USP <467>. Terminal finished product types include 3'-amino-modified antisense oligonucleotides, GalNAc-conjugated small interfering RNA, 5'-amino-modified aptamers, internally amino-modified oligonucleotide probes used as drug substance characterization standards, and lyophilized powders for injection after conjugation.

When Serinol-Derived Contrast APIs Are Processed for Veterinary CT and Angiography

For veterinary CT and angiography, serinol-derived monomeric APIs are processed under the same synthetic conditions as human-grade contrast media, but release and stability testing follow veterinary-specific regulatory frameworks. Serinol charging is unchanged at 2.0–2.2 mol for iopamidol and 3.0–3.3 mol for iohexol per mole of triiodinated precursor. The API is synthesized under ICH Q7-aligned conditions and formulated into injectable solutions at 240–370 mg I/mL for canine, feline, equine, and exotic animal imaging. Finished product release applies VICH GL18(R2) for impurities in new veterinary drug substances, VICH GL3(R) stability testing, EU Regulation 2019/6 for veterinary medicinal products, 21 CFR 211, USP <788>, USP <85>, and USP <71>. The manufacturing process includes terminal sterilization in 50 mL and 100 mL Type I glass vials with bromobutyl stoppers; post-sterilization particulate testing is performed on every batch because rapid intravenous injection in small animal patients increases the clinical impact of particulate matter. Terminal product types include single-use veterinary contrast vials for CT angiography, portal venography, and myelography in companion animals.

    More Introduction

2-Amino-1,3-propanediol; Serinol Pharma Grade API is supplied under CAS 534-03-2, molecular formula C3H9NO2, and molecular mass 91.11 g/mol. The product is presented as a white to off-white crystalline powder or granular solid intended for tablet, capsule, granule, oral liquid, and injectable manufacture. The grade designation Serinol Pharma Grade API serves as the product model; no proprietary model number is assigned. Traceability is maintained by lot number, CAS registry number, and internal specification code. Manufacturing is performed under ICH Q7 GMP for active pharmaceutical ingredients. Release testing is aligned to ICH Q6A and includes assay, water content, related substances, residual solvents, elemental impurities, bioburden, and bacterial endotoxins. The molecule contains one primary amine and two primary hydroxyl groups, giving a hydrophilic amino alcohol with high aqueous solubility. This characteristic influences granulation behaviour, dissolution in oral liquids, and the risk of moisture uptake during storage or capsule filling.

Unlike technical-grade serinol intended for polymer or industrial synthesis, this grade is controlled for pharmaceutical dosage-form use. Technical-grade material is commonly released on assay and appearance without a full pharmacopoeial alignment. The pharma grade is tested for USP <85> endotoxin, USP <61>/<62> microbial limits, USP <232>/<233> elemental impurities, and ICH Q3C(R8) residual solvents. The injectable grade can be dissolved in Water for Injection and passed through a 0.22 µm sterilizing-grade filter. Technical-grade material is not qualified for this route because of uncontrolled particulate, endotoxin, and solvent burdens. The pharma grade also provides regulatory documentation including batch certificate, residual solvent data, elemental impurity data, and BSE/TSE statements.

What Release Specifications Govern Injectable-Grade Serinol?

The release specification matrix for the typical pharma grade is shown below. Actual lot values are reported on the certificate of analysis and may be tightened by the applicant in the drug product filing.

Because serinol has no significant UV chromophore, impurity profiling by HPLC with conventional UV detection is poorly suited. Release and stability methods therefore use evaporative light scattering detection, charged aerosol detection, or GC after derivatization. This is an analytical distinction from aromatic amine APIs that can be quantified by UV absorbance. Residual solvents are measured by headspace gas chromatography on a 30 m × 0.32 mm × 1.8 µm DB-624 column with flame ionization detection, and the procedure should meet USP <467> Option 1 sensitivity. Elemental impurities are determined by ICP-MS following microwave-assisted digestion to ICH Q3D(R2) PDE levels for oral and parenteral routes. For parenteral products, the lower PDEs for cadmium, lead, arsenic, and mercury require tighter internal limits than oral grades.

Endotoxin testing uses the kinetic chromogenic LAL method with a labelled sensitivity of 0.01 EU/mL. Sample inhibition is addressed through pH adjustment and endotoxin-specific buffer. The release limit of ≤0.50 EU/mg supports injectable compounding, but the final drug product limit must be calculated from the maximum adult dose using the K/M equation in Ph. Eur. 2.6.14 or USP <85>. For direct compression and capsule filling, the material should be milled to a controlled particle size. The hydrophilic character of serinol facilitates dissolution in aqueous granulating fluids, but can generate binder-like cohesion if overwetted. In high-shear wet granulation of similar hygroscopic amino alcohols, a target granulation endpoint of 1.5–2.5 % loss-on-drying after fluid-bed drying is common. The dried granule is passed through a 0.5 mm conical mill and blended in a diffusion mixer at ≤25 rpm for 15–20 min before compression. Tablet press tooling with a compression force range of 6–12 kN is typical for conventional excipient systems, but published data for this specific serinol formulation configuration are limited; force-displacement profiles should be generated on a compaction simulator or instrumented rotary press. Capsule filling on a dosator machine requires control of powder bed height and relative humidity below 40 % RH to prevent sticking.

Cleanroom Transfer and Aseptic Filtration in Parenteral Manufacturing

For injectable manufacture, the pharma grade is dissolved in Water for Injection at the compounding pharmacy or aseptic filling facility. The bulk solution is clarified through a 0.45 µm prefilter and then aseptically filtered through a 0.22 µm sterilizing-grade membrane at a differential pressure not exceeding 2.0 bar. Filter compatibility with the primary amine group should be confirmed with the filter manufacturer because certain membrane grades can bind amine-containing molecules or release extractables. Terminal sterilization is possible only after stability studies confirm that the process does not generate related substances above ICH Q3A thresholds. Typical steam sterilisation cycles at 121 °C for 15 min may be evaluated; however, pH shift and dimer formation must be monitored by HPLC-ELSD or charged aerosol detection. For aseptic processing, the drug product site must maintain Grade A/ISO 5 filling conditions with Grade B/ISO 7 background, and the API must be transferred through a qualified sterile transfer system. The API is not supplied as sterile; it is supplied with low bioburden for terminal filtration.

Oral liquid formulations prepared with serinol benefit from its high aqueous solubility. Simple solutions can be compounded in purified water and adjusted to target pH with hydrochloric acid or sodium hydroxide. Because the molecule contains a primary amine, pH drift and potential interaction with reducing sugars in flavouring systems should be evaluated during forced degradation. The oral liquid should be protected from light and maintained at ≤25 °C unless stability data support a wider range. Granule-filled sachets can be produced by dry-mixing serinol with mannitol and microcrystalline cellulose, then granulating with purified water or an aqueous binder solution. Because serinol dissolves readily in water, granule strength is sensitive to liquid-to-solid ratio. An L/S ratio of 0.08–0.12 is often evaluated for similar amino alcohols, but design of experiments is required for each formulation. The wet mass is passed through a 1.0 mm screen and dried in a fluid-bed dryer at inlet air temperature 50–60 °C until loss-on-drying reaches 1.0–2.0 %. This supports subsequent sachet filling with acceptable dissolution.

When Technical-Grade Serinol Is Compared with Pharma Grade API

At the analytical level, the critical difference is not chemical identity but the extent of control. Both grades may share CAS 534-03-2; however, technical-grade material is not released under a pharmaceutical quality system. The pharma grade is manufactured with dedicated or dedicated-equipment cleaning procedures validated under 21 CFR 211.67. Technical-grade streams may contain variable residual nitrite or nitro intermediates if different synthetic routes are used; these are controlled in the pharma grade by TLC or HPLC limit tests. The matrix below summarises the usual comparator criteria.

    AttributePharma Grade APITechnical Grade
    EndotoxinUSP <85>, ≤0.50 EU/mgNot controlled
    BioburdenUSP <61>/<62>, TAMC ≤100 CFU/gMay exceed 1000 CFU/g

Regulatory documentationCoA, BSE/TSE, elemental impurities, residual solventsLimited to assay
PackagingDouble antistatic liner in HDPE drum, Class 8Industrial bag or fiber drum
Suitability for injectable manufactureSuitable after aseptic filtrationNot suitable

The difference in packaging is not cosmetic. The pharma-grade liner is tested for extractables and leachables and is heat-sealed to limit moisture ingress. Industrial packaging does not provide a moisture barrier sufficient to protect the hygroscopic solid. If a technical-grade material is reprocessed or purified, the full impurity profile must be revalidated; a single assay result is not sufficient to establish pharmaceutical acceptability. Compared with technical serinol, the pharma grade also has tighter particle size control. The D90 limit of ≤200 µm for direct compression is not typical for technical material, which may contain particles above 500 µm after standard milling. Capsule filling weight variability can increase if D90 exceeds 300 µm; therefore particle size distribution is release-tested for pharma grade using laser diffraction. Stability studies under ICH Q1A(R2) are recommended because serinol is hygroscopic. At 40 °C/75 % RH, the powder may cake. The pharma grade is therefore packaged in sealed antistatic polyethylene liners with desiccant and stored at ≤25 °C and <40 % RH. The material should not be stored in unsealed containers near acids or oxidising agents because the primary amine can undergo Schiff base formation with aldehydes and ketones.
